The Four Diastereomeric Lactones of γ-HydroxyisoleucineOf the 4 possible diastereomeric γ-hydro-L-isoleucines only 2, the (2S,3R,4S)-isomer 1 and the (2S,3R,4R)-isomer 3 have so far been described - as hydrochlorides of their lactones. In an attempt to prepare the missing ones (as the antipodes) D-allo-isoleucine was photo-chlorinated. From the mixture the hydrochloride of the (2R,3R,4R)-lactone 7 was obtained in crystalline state, but the (2R,3R,4S)-isomer 5 could only be enriched to a 70% pure preparation which, however, allowed its characterization by 1H-NMR and chromatography. By heating with barium hydroxide (130°C, 24 h) the amino acids 2, 4 and 8 were partly epimerized at C-2 to 6, 8 and 4, respectively. By heating with 6 N HCl (using the same conditions) an inversion equilibrium was established at each chiral center, presumably owing to participation of a 3,4-unsaturated intermediate.

Von den 4 möglichen diastereomeren γ-Hydroxy-L-isoleucinen waren in Form ihrer Amino-lacton-hydrochloride bisher die (2S,3R,4S)-Verbindung 1 und die (2S,3R,4R)-Verbindung 3 bekannt. Von den 2 fehlenden, die - als Antipoden - bei der Photochlorierung und Hydrolyse aus D-allo-Isoleucin entstehen, konnte die (2R,3R,4R)-Verbindung als Hydro-chlorid des Lactons 7 kristallisiert, die (2R,3R,4S)-Verbindung 5 infolge ihrer mit 7 sehr ähnlichen Eigenschaften nur auf etwa 70% angereichert werden, aber soweit, daß ihre 1H-NMR-Signale und ihre chromatographischen Eigenschaften feststellbar waren. Mit Bariumhydroxid (130°C, 24 h) wurde die 1 entsprechende offene Aminosäure 2 an C-2 teilweise zu 6 sowie 4 zu 8 und 8 zu 4 epimerisiert. Mit 6 N HCI (gleiche Bedingungen) entstanden aus jedem Diastereomeren alle 4 Isomeren. Für diesen Vorgang wird ein 3,4-ungesättigtes Zwischenprodukt verantwortlich gemacht.
