    Notes: Abstract Although feline salivary glands have been used in investigations on secretion and microlithiasis and both processes involve calcium, nothing is known about its distribution in these glands. Therefore we have demonstrated the presence of calcium by a histochemical technique using glyoxal bis(2-hydroxyanil) and a biochemical technique using dry ashing. The histochemical technique stained serous acinar cells weakly and rarely found mucous acinar cells strongly in the parotid gland, mucous acinar cells moderately to strongly and serous acinar cells weakly in the sublingual gland, and central and demilunar acinar cells moderately to strongly in the submandibular gland. The biochemical technique revealed less calcium in the parotid than in the submandibular and sublingual glands. Both techniques revealed a decrease of calcium in submandibular and sublingual glands following parasympathetic stimulation. The histochemical distribution of calcium, which corresponds to that of acinar secretory glycoprotein, and the loss of calcium following parasympathetic stimulation, which causes release of secretory granules, indicate the presence of calcium in secretory granules. The concentration of calcium in the different types of acinar cell corresponds to the acidity of the secretory glycoprotein and suggests that calcium is present as a cationic shield to allow the condensation of polyionic glycoprotein in secretory granules.

    Description / Table of Contents: Zusammenfassung Myoepithelzellen der Speicheldrüsen lassen sich bei der Katze — nicht jedoch bei Mensch und Hund — mittels der alkalischen Phosphatase-Reaktion darstellen. Der Nachweis für Adenosintriphosphatase fällt in diesen Zellen beim Menschen, nicht bei Katze und Hund, positiv aus. Elektronenmikroskopisch-cytochemisch zeigt sich, daß das Reaktionsprodukt der jeweiligen Methode bei der Katze und beim Menschen an den Plasmamembranen der Myoepithelzellen auftritt und zwar am regelmäßigsten und meistens am stärksten in der Nachbarschaft der Azinuszellen. Beim Hund fällt in der Ohrspeicheldrüse sowie den nicht-mukösen Acini der Glandulae submandibularis und sublingualis das Reaktionsprodukt der Adenosintriphosphatase-Reaktion an der Begrenzung der Azinuskanälchen und der Lumenoberfläche aus. Alkalische Phosphatase ist in der Glandula sublingualis in den Wänden der Kanälchen und an den Lumina lokalisiert. Diese bemerkenswerten Unterschiede zwischen den verschiedenen Tierarten zeigen, daß keine der verwendeten Methoden zur Universalmarkierung von Myoepithelzellen der Speicheldrüsen geeignet ist. Außerdem ist die Aktivität des Myoepithels bei den einzelnen Drüsen uneinheitlich. Dies legt die Vermutung nahe, daß man sich selbst bei Anwendung der richtigen Methode nicht darauf verlassen kann, alle in einem Gewebsschnitt vorhandenen Myoepithelzellen zu erfassen.
    Notes: Summary Salivary myoepithelial cells were demonstrated by alkaline-phosphatase techniques in cat, but not in man or dog, and by an adenosine-triphosphatase technique in man, but not in cat or dog. Electron-microscopical cytochemistry showed that the reaction product from the respective techniques in cat and man was associated with the myoepithelial plasma membrane and that it was most constant and usually strongest at the plasma membrane adjacent to the acinar cells. In the dog, the reaction product from the adenosine-triphosphatase technique was found lining the canaliculi and lumina of the acini of the parotid gland, and of the non-mucous acini of the submandibular and sublingual glands. Alkaline-phosphatase reaction product was found lining the canaliculi and lumina in the sublingual gland. These remarkable species differences indicate that neither technique can be regarded as a universal marker of salivary myoepithelial cells. Inconsistencies in activity were found within the myoepithelium of individual glands and suggest that even the appropriate technique may not be relied upon to demonstrate all the myoepithelial cells present in a tissue section.

    Notes: Summary Some ductal cells of pleomorphic adenomas showed evidence of secretory activity, with apical secretory granules, thiamine pyrophosphatase activity in the Golgi apparatus, and acid phosphatase activity in GERL-like structures and in immature secretory granules. Alkaline phosphatase activity was demonstrated rarely at the luminal plasma membrane and in intracellular vesicles, suggesting resorptive activity. ATPase reaction product was associated with contiguous surfaces of tumour cells, particularly of those cells adjacent to the basement membrane, these latter cells apparently differentiating in a different manner to the luminal cells. A comparison of luminal ductal cells of the tumours with normal salivary glands revealed most similarity with intercalary ductal cells.

    Notes: Synopsis Submandibular, sublingual and parotid glands of cat have been studied following periods of ductal ligation ranging from 1 day to 1 year. An increased prominence of granules of acid phosphatase, β-glucuronidase and E600-resistant esterase reaction products was sometimes seen in acinar cells and atrophic striated ductal cells, and probably represents increased lysosomal enzymic activity, which may be of importance in the adaptation of the parenchyma to the altered environment. Other reaction products often appeared to be at normal levels in parenchymal structures that were not very atrophic, and at reduced levels in those that were very atrophic, suggesting a reduction of functional activity in these structures.

    Notes: Abstract This investigation concerns the natural history of microlith in the salivary glands of cat. Microliths were detected in more sublingual than submandibular glands and were almost absent in the parotid. They were found intraparenchymally, intraluminally and interstitially, and ultrastructurally in phagosomes of acinar, ductal and myoepithelial cells, intermixed with the cytoplasm of degenerate acinar cells, and in intraparenchymal macrophages and a multinuclear giant cell. They appear to form in healthy acinar cells during autophagocytosis, and possibly to be discharged luminally, laterally or basally, and to form in the debris of degenerate cells intraparenchymally and intraluminally. They appear to be removed by expulsion in the saliva, scavenging macrophages, and possible eventual degradation in the parenchymal phagosomes. The greater occurrence of microliths in the sublingual gland may relate to a low level of secretory activity, and the near absence of microliths in the parotid to a low level of calcium. The feline salivary glands were found to be an outstanding model for the investigation of microlithiasis.

    Notes: Synopsis The localization of reaction product arising from incubation with thiamine pyrophosphate at pH 7.2 and β-glycerophosphate or nitrophenylphosphate at pH 5 has been examined in central and demilunar acinar cells, myoepithelial cells, and cells of the striated ducts. Thiamine pyrophosphate reaction product was seen in the inner saccules of the Golgi apparatuses of the different types of cell. β-Glycerophosphate and nitrophenylphosphate reaction products were seen in lysosomes, and in GERL-like structures in all except the cells of the striated ducts, in which the reaction products were observed in the Golgi apparatus. In central cells, the latter two reaction products were also seen in secretory granules and the cytosol.

    Notes: Summary Lumina and adluminal cells in human salivary gland pleomorphic adenomas were found to contain neutral, carboxylated, and occasionally sulphated glycoproteins. A variable component of luminal contents and secretory granules did not appear to contain glycoprotein and possibly consisted of protein. Glycosaminoglycans, which appeared to be hyaluronic acid and chondroitin sulphate, were demonstrated rarely in lumina, often between epithelial cells, and forming the matrix of myxoid tissue and, together with collagen, chondroid tissue. No differences were seen between tumours from parotid glands and those from submandibular glands. Glycoproteins demonstrated in the epithelium are similar to those of intercalary ducts of parotid and submandibular glands, and may represent a primitive form of salivary secretion. Glycosaminoglycans secreted intercellularly by epithelial cells cause their increasing separation to form myxoid or chondroid tissue. This stromalization extends to lumina to produce a loss of epithelium. Pleomorphic adenoma appears to be a manifest example of variable derepression of the genotype.

    Notes: Summary Microliths in Araldite-embedded pieces of submandibular and sublingual glands of cat were stained in semithin sections by Methylene Blue and Azure II followed by Basic Fuchsin, and were examined in ultrathin sections by electronmicroscopical X-ray microanalysis. Calcium and phosphorus were detected in substantial aggregates of crystals that were stained by Basic Fuchsin and appeared to be hydroxyapatite, but were not detected in granular material that was stained by Methylene Blue and Azure II and appeared to be organic. The polychromatic stain thus appears to be a useful indicator of calcified material. The majority of microliths in acini contained substantial aggregates of crystals, whereas the majority of those in ducts did not. This corresponds to the distribution of the glandular calcium, and suggests that microliths are variously enriched with calcium according to its local level.

    Notes: Synopsis Lower labial, buccal and sublingual glands have been examined. The lower labial glands contained a greater proportion of seromucous cells than the buccal and sublingual glands. In the latter glands there was a greater proportion of mucous cells that are stainable with the Alcian Blue critical electrolyte concentration technique with 0.2m MgCl2 and above than in the lower labial glands. Granules of acid phosphatase, β-glucuronidase and E600-resistant esterase reaction product, presumbly, representing lysosomal enzyme activity, were usually more prominent in seromucous than in mucous cells. Thiamine pyrophosphatase and inosine diphosphatase activities were demonstrated in a Golgi-like distribution, which was more prominent in mucous cells. Diffuse non-specific esterase, NADH-and NADPH- diaphorases and peroxidase activities were more prominent in seromucous cells. However, diffuse acid phosphatase activity was more prominent in mucous cells. Granules of β-glucuronidase, acid phosphatase and E600-resistant esterase reaction product were seen in ductal cells, and were often concentrated in the supranuclear region. Arylamidase activity was present lining the lumen of some ducts and acini.
