    Notes: SUMMARY: The activity of glutamic-oxaloacetic transaminase (GOT) and glutamicpyruvic transaminase (GPT) of bovine and porcine muscle tissue and muscle press juice was determined. The total GPT activity of muscle tissue is about one tenth of the GOT activity. There are no remarkable differences in the activities of GOT and GPT between these slaughter animals and other species (rat, rabbit and man). The GOT activity of the longissimus dorsi muscle of pigs is significantly higher than that of the same bovine muscle. The mitochondrial (GOTM) and sarcoplasmic isozymes (GOTB) of GOT in skeletal muscles of cattle and pigs were determined after electrophoretic separation. The ratio GOTM:GOTS in skeletal muscle was found to be about 1:1. There is only a small decrease in GOT activty during storage of muscle tissue at 0 or +4°C for several weeks postmortem. The small activity of GOTM in the muscle press juice does not substantially change during storage of muscle tissue under these conditions, indicating that there is no drastic change of the mitochondrial structure during aging of meat. Bacterial spoilage of meat, however, results in the release of GOTM from the mitochondria.

    Notes: SUMMARY: The total activity of glutamic-oxaloacetic transaminase (GOT). the relative activity of the mitochondrial GOT isozyme (GOTM) and the total activity of glutamic-pyruvic transaminase (GPT) in five different muscles of four pigs and two cows were determined. A highly significant, positive correlation was found between either GOT or GPT activity and the amount of muscle pigments. Tonic “red”) (muscles had high transaminase activities and tetanic (“white”) muscles had low transaminase activities. However, the relative GOTm activity (GOTM activity in percent of the totaf GOT activity) of porcine muscle was found to be almost constant in the different types of muscle. Likewise, in bovine muscles, higher GOT and GPT activities were found in “red” than in “white” muscles. However, the relative GOTK activity seemed to be higher in the “red” than in the “white” muscles. A discussion of the transaminase activities and their relationship to muscle function is presented.

    Notes: SUMMARY: Freezing and thawing of bovine and porcine muscle cause a remarkable release of the mitochondrial isozyme of the glutamic-oxaloacetic transaminase (GOTM) from the mitochondrial structures resulting in an increase of GOTM activity in the muscle press juice. The lower the freezing temperature the stronger is this effect. Repeated freezing and thawing increases the release of GOTM. It is considered that the level of GOTM activity in the muscle press juice indicates the extent of mitochondrial damage. On the basis of these results a simple and rapid routine method was developed which allows a reliable differentiation between nonfrozen and frozen and thawed meat.

    Description / Table of Contents: Summary A report is given of a method of determining the calcium, magnesium, zinc, and iron content of an animal tissue (1 to 5 g) in a single incineration solution. The hydrochloric acid solution of the dry ash is passed through a cation exchange column. In this manner, phosphate and 90 to 95% of the iron is separated from calcium, magnesium, and zinc. The three latter elements are retained by the column by exchange. After elution with 5N hydrochloric acid they are determined by two complexometric titrations. Iron is determined in the eluate combined with the rinsings; the o-phenanthroline reaction is employed. Details are given of the incineration conditions and the determination of the iron by means of phenanthroline.
    Abstract: Résumé On décrit une méthode qui permet de déterminer sur une solution unique provenant de la calcination de tissus animaux (1 à 5 g) les teneurs en calcium, magnésium, zinc et fer. La solution chlorhydrique des cendres sèches est traitée sur une colonne d'un échangeur de cathions ce qui permet de séparer les phosphates et le fer (ce dernier dans une proportion de 90 à 95%), du calcium, du magnésium et du zinc. Les trois derniers éléments sont retenus par échange sur la colonne et, après élution par l'acide chlorhydrique 5 N, ils sont dosés au moyen de deux titrages complexométriques. Dans le liquide qui s'est écoulé de la colonne, auquel on réunit l'éluat, on dose le fer par la réaction à l'orthophénanthroline. Les conditions de la calcination et le dosage du fer par la phénantroline sont l'objet de précisions détaillées.
    Notes: Zusammenfassung Es wird über ein Verfahren berichtet, den Calcium-, Magnesium-, Zinkund Eisengehalt tierischen Gewebes (1 bis 5 g) in einer einzigen Veraschungslösung zu bestimmen. Die salzsaure Lösung der Trockenasche wird durch eine Kationenaustauscher-Säule gegeben und hierdurch Phosphat und 90 bis 95% des Eisens von Calcium, Magnesium und Zink getrennt. Die drei letzteren Elemente werden von der Säule unter Austausch zurückgehalten und nach Elution mit 5-n Salzsäure durch zwei komplexometrische Titrationen bestimmt. In dem mit dem Eluat vereinigten Durchlauf wird Eisen mittels der o-Phenanthrolin-Reaktion erfaßt. Auf die Veraschungsbedingungen und die Eisenbestimmung mit Phenanthrolin wird näher eingegangen.

    Notes: Der von Mauthner bei der Einwirkung von KCN auf Cystin neben Cystein erhaltenen Säure kann nicht die Struktur einer α-Amino-β-rhodan-propionsäure zukommen. In der Säure lassen sich keine freien Rhodangruppen nachweisen und es fehlen ihr Eigenschaften einer α-Aminosäure. Die Rhodanaminopropionsäure ist zwar primäres Reaktionsprodukt, jedoch kommt es infolge der Nachbarschaft von SCN- und NH2-Gruppe zu einer Cyclisierung unter Bildung eines Thiazolinringsystemes. In der Mauthner-Säure liegt 2-Aminothiazolin-carbonsäure-(4) bzw. 4-Carboxy-thiazolidon-(2)-imid vor. Bei der Einwirkung von KCN auf Cystamin wird in glatter Reaktion das 2-Amino-thiazolin gebildet. Die Grundreaktion der Cyanideinwirkung auf Diaminodisulfide, die den bekannten Nitroprussidnatrium-Test auf SS-Bindungen etwa in Peptiden und Eiweißstoffen ermöglicht, ist damit geklärt.

    Notes: Es werden die Bedingungen der Umsetzung zwischen Cystin und Kaliumcyanid, die zu Cystein und α-Amino-β-rhodan-propionsäure führt, näher untersucht. Auch das Cystein läßt sich dabei präparativ abtrennen. Mit Wasserstoffperoxyd als Oxydationsmittel, das das primär entstandene Cystein oxydiert, gelingt es bei dieser Umsetzung das Cystin quantitativ in die α-Amino-β-rhodan-propionsäure umzuwandeln. Die Rhodangruppe läßt sich in der α-Amino-β-rhodan-propionsäure durch Behandlung mit Säuren oder nascierendem Wasserstoff nicht durch die SH-Gruppe ersetzen. Mit Alkalien gelingt die Hydrolyse zum Thiol, jedoch reagiert das Cystein dabei sofort weiter mit der gleichzeitig entstehenden Cyansäure unter Bildung einer Ureidosäure, die als Cystein-bzw. Cystinhydantoin festgelegt wird.
