    Notes: Abstract We have tested 380 strains of our Fungal Culture Collection (MZKIBK), for the activity of the inhibitor of HMG CoA reductase, mevinolin. All fungal strains were cultivated in a two-step submerged fermentation on selected substrates. For the first screening, samples of broth extracts were tested by TLC and positive results were further evaluated by HPLC analysis. In addition to already known microbiol producers of HMG CoA reductase inhibitors and some newly identified strains having moderate activity, we found that fungi of the basidiomycetous genus Pleurotus, especially species P. ostreatus, P. saca and P. sapidus, are an as yet unknown but promising source of the hypocholesterolaemic agent.

    Notes: Abstract Screening of different yeast species showed that they are able to synthesize hydroxymethylglutaryl-CoA (HMGCoA) reductase inhibitors. Crude methanol extracts and the purified inhibitors from Pichia labacensis and Candida cariosilignicola were tested for their biological activity on the solubilized microsomal HMGCoA reductase from Chinese hamster ovary cells. Identification of the inhibitors was studied by thin layer chromatography, high pressure liquid chromatography and mass spectroscopy.

    Notes: Six different species of the known teleomorphic food-borne xerophilic genus Eurotium were repeatedly isolated in a mycodiversity study of hypersaline waters. At salinities above 17% NaCl, E. amstelodami was detected most consistently, followed by E. repens and E. herbariorum, while E. rubrum, E. chevalieri and a potentially new species, “Eurotium halotolerans”, were detected only occasionally at lower salinities. The qualitative secondary metabolite profiles produced by Eurotium spp. from salterns were not different from those of Eurotium spp. from foods and other habitats. Spatiotemporal frequency of occurrence and in vitro determined adaptive ability of propagules to survive prolonged exposure to hypersaline conditions indicate that E. amstelodami, E. herbariorum, and E. repens contribute to the indigenous fungal community in hypersaline water environments, while E. rubrum, E. chevalieri and “E. halotolerans” are only temporal inhabitants of brine at lower salinities.

    Notes: Penicillium crustosum is common in food and feed both in subtropical and temperate regions. Recently, it has also been found occurring frequently in glacier ice, sea ice and sea water of Arctic regions of Svalbard. The aim of the study was to compare isolates of the same fungal species from widely different habitats and geographic regions to see if the nutritional physiology and the profile of secondary metabolites were consistent or depended on the isolation source. All 121 strains examined produced the following families of secondary metabolites: penitrems (100%), roquefortines (100%), terrestric acids (99.2%) and viridicatols (100%), whereas 81 of 83 Arctic isolates additionally produced andrastin A. However, only 8 of 38 non-Arctic isolates produced detectable andrastin A. The quantitative profiles of 96 strains were compared using cluster, principal component and correspondence analyses. There was no clear grouping of Arctic versus non-Arctic, creatine positive versus creatine negative strains.

    Notes: The ascomycetous black yeasts Hortaea werneckii, Phaeotheca triangularis, and Trimmatostroma salinum are halophilic fungi that inhabit hypersaline water of solar salterns. They are characterized by slow, meristematic growth and very thick, darkly pigmented cell walls. The dark pigment, generally thought to be melanin, is consistently present in their cell walls when they grow under saline and non-saline conditions. We used the inhibitor tricyclazole to test the fungi in this study for the presence of 1,8-dihydroxynaphthalene (DHN)-melanin biosynthesis, since fungal melanins reportedly are derived either from DHN, tyrosine via 3,4-dihydroxyphenylalanine, γ-glutaminyl-3,4-dihydroxybenzene, or catechol. Tricyclazole-treated cultures of the fungi were reddish-brown in color and contained typical intermediates of the DHN-melanin pathway, as demonstrated by high-performance liquid chromatography. This investigation showed that the three fungi synthesized DHN-melanin under saline and non-saline growth conditions.

    Notes: Different microfungi were isolated from salty soil and hypersaline water samples, taken from the extreme environment of operating marine solar salterns. The filamentous fungus most frequently and repeatedly isolated was identified as the pathogen Aspergillus fumigatus. Because of its great morphological variability and mycotoxin production ability, 50 A. fumigatus isolates were selected for additional characterisation by their secondary metabolites profile, which was determined by thin layer chromatography (TLC) and high performance liquid chromatography (HPLC). At standard conditions 45% of the isolated strains produced different mycotoxins and in ecological conditions prevailing in the saltern (high salinity and temperature) production of important mycotoxins (verruculogen, fumitremorgins B and C, fumigaclavine A) was lowered or inhibited.

    Notes: Hypersaline waters in salterns have so far been considered to be populated only with halophilic algae and bacteria and completely lacking halophilic fungi. In this paper we present population dynamics of polymorphic black yeasts, isolated from hypersaline waters (3–30% NaCl) of a saltern, in relation to different physicochemical parameters. Hortaea werneckii, Phaeotheca triangularis, Trimmatostroma salinum, Aureobasidium pullulans and Cladosporium spp. were detected with the highest frequency just before the peak of halite (NaCl) concentration. Since H. werneckii, P. triangularis and T. salinum are not known outside saline environments, these results suggest that hypersaline water is their natural ecological niche.

    Notes: The development of crop plants with increased salt tolerance necessitates the study of naturally salt-tolerant eukaryotic species. We studied the bio-synthesis of glycerol as a compatible solute in the halophilic eukaryotic microorganism, black yeast Hortaea werneckii. A restriction fragment–differential display technique was used to investigate the transcriptome of the organism. Eight differentially expressed genes were identified in response to growth at different salinities. Although the putative functions of their products, P-type ATPase, ubiquinone reductase, aconitase, RNA helicase, Asn-tRNA ligase, isoamyl alcohol oxidase, and phosphatidylinositol-3-kinase, are not intimately related within the cellular machinery, the results presented here are sufficient to propose a model which describes how H. werneckii adapts to extremely high salinities. Some of these mechanisms of adaptation to raised environmental salinity are similar to those in other salt-sensitive species, e.g. glycerol accumulation, there also appear to be novel mechanisms present such as the use of different energy production mechanisms and post-transcriptional regulation of gene expression. Our results have also provided new data on two genes from two other fungal species, the Neurospora crassa B1D1.130 gene and the Aspergillus ustus amdS-A gene.

    Notes: Abstract By using different techniques and media, microfungi were isolated from cadavers of the cave cricket Troglophilus neglectus, collected on the walls of a Slovenian cave. The isolated mycobiota was consistently different for adult and larval stages of T. neglectus cadavers. Fungi isolated with the highest frequency from the larval stage were represented by five different species of the genus Mucor, the prevailing one being a newly described species, M. troglophilus. The dominating fungus isolated from the adult stage was the well-known fungal entomopathogen Beauveria bassiana. The other isolated fungi were mainly primary and secondary saprotrophic colonizers.
