    Notes: Summary The major biliary metabolites of the mutagen, quercetin, the pro-mutagen rutin and taxifolin have been identified by EI-mass spectrometry, UV-spectroscopy and chromatographic methods as conjugates of the corresponding 3′-O-methyl ethers. The toxicological significance of these findings is discussed.

    Notes: Abstract Three Escherichia coli mutants defective in formate-dependent nitrite reduction (Nrf activity) were characterised. Two of the mutants, JCB354 and JCB356, synthesized all five c-type cytochromes previously characterised in anaerobic cultures of E. coli. The third mutant, JCB355, was defective for both cytochrome b and cytochrome c synthesis, but only during anaerobic growth. The insertion sites of the transposon in strains JCB354 and JCB356 mapped to the menFDBCE operon; the hemN gene was disrupted in strain JCB355. The mutation in strain JCB354 was complemented by a plasmid encoding only menD; strain JCB356 was complemented by a plasmid encoding only menBCE. A mutant defective in the methyltransferase activity involved in both ubiquinone synthesis and conversion of demethylmenaquinone to menaquinone expressed the same Nrf activity as the parental strain. The effects of men, ubiA and ubiE mutations on other cytochrome-c-dependent electron transfer pathways were also determined. The combined data establish that menaquinones are essential for cytochrome-c-dependent trimethylamine-N-oxide reductase (Tor) and Nrf activity, but that either menaquinone or ubiquinone, but not demethylmenaquinone, can transfer electrons to a third cytochrome-c-dependent electron transfer chain, the periplasmic nitrate reductase.

    Notes: Abstract Operon fusion strains of Escherichia coli K-12 have been used to demonstrate that transcription of the structural gene for NADH-dependent nitrite reductase is regulated by oxygen repression and induction by its substrate, nitrite. This two-stage regulation of nirB is totally dependent upon a functional Fnr protein. Unlike the Fnr-dependent fumarate reductase operon, nirB transcription is not repressed by nitrate. These results suggest that the Fnr protein is simply a positive control protein essential for the derepression of some, but not all, anaerobically-induced operons rather than a more general redox-sensitive regulator, as suggested by the redox control hypothesis for the regulation of gene expression in facultatively anaerobic bacteria.

    Notes: Summary The effect of alcoholic cirrhosis of the liver on the disposition and metabolism of (+)-cyanidanol-3 has been studied in three patients. Following oral intake of U-14C-(+)-cyanidanol-3 2 g, the unchanged drug was detected in plasma between 0.5 and 24 h after dosing and the metabolites, measured only as14C-levels, were still detectable at 120 h. A mean of 55% of the dose was excreted in the urine, in which the major metabolites were the glucuronic and sulphuric acid conjugates of (+)-cyanidanol-3 and 3′-O-methyl-(+)-cyanidanol-3, although one patient excreted a larger proportion as non-conjugated metabolites. The pattern of metabolism in the patients did not differ significantly from that in control subjects, suggesting that the drug is handled by the cirrhotic liver in a similar manner to the normal liver.

    Notes: Abstract Five different c-type cytochromes have been detected during anaerobic growth of various Escherichia coli strains in different media. None of these cytochromes was detectable in aerobically-grown cultures. Only a single, 43 kDa cytochrome was synthesized in response to the presence of trimethylamine-N-oxide: synthesis of this cytochrome was unaffected by the presence of nitrate or nitrite, was repressed by oxygen, but was dependent upon a funtional tor operon located at minute 22 (coordinate 1070 kb) on the E. coli chromosome. The other four cytochromes, masses 16, 18, 24 and 50 kDa, were induced by nitrite coordinately with formate-dependent nitrite reductase activity, but repressed by oxygen and nitrate. As only the 18 kDa and 50 kDa cytochromes are encoded by the nrf operon located at minute 92 (coordinate 4366 kb), there must be other loci, possibly essential for formate-dependent nitrite reduction, encoding the 16 kDa and 24 kDa cytochromes. No other c-type cytochrome was detected under any growth condition tested.
