Notes:
All preparations bioassayed were sterile. In addition, incorporation of antibiotics during preparation of the skin extract did not lessen the mortalities. The toxicity of the dialyzed mouse skin extract is not due to the presence of bacteria. In searching for factors which cause death in burn patients receiving therapy, we investigated the skin as a source of toxic factors. Both normal and burned skin, when processed as described, yielded sterile extracts which caused death in saline-treated scalded mice. By means of ammonium sulfate precipitation and Sephadex chromatography of this extract, fractions having enzymatic activity have been obtained. These fractions also proved toxic to saline-treated scalded mice. Isolation and identification of the active material may contribute to understanding the cause of death in burn patients and hopefully to therapy.
