    Notes: : A survey of 34 open hand dug wells was performed in the Senegal River basin of West Africa. Nitrate concentrations were determined on the well water samples over a six-month period. With the exception of two wells, the wells indicated varying levels of nitrate contamination. The range of concentrations was 0.10 to 880 mg/I as nitrate. These data when compared to physical characteristics, land use, and age using a chi-square analysis did not suggest any strong association. The fact that these wells are open and in a semiarid climate may be of such importance that the previously considered factors are of minor relative importance. Contamination may be primarily the result of foreign matter entering the well mouth.

    Notes: Summary The effects on cell survival of maintaining bone marrow cells (CFU-S) in situ following irradiation and before assay by transplantation was investigated. When the CFU-S cells are maintained in situ following irradiation survival drops and plateaus at about 9 h post-irradiation. Evidence is presented that this decrease in survival may be due to potentially lethal damage repair (PLD) inhibition caused by post-irradiation in situ holding. This effect on PLD repair is different than that usually found in cells in vitro and in vivo tumors in that it mainly alters the shoulder rather than the slope of the survival curve of CFU-S cells. It is different than PLDR found in vivo for normal mammary and thyroid gland epithelial cells because in situ holding decreases rather than increases the survival of CFU-S cells. Evidence is also presented that the radiation survival curve for in situ bone marrow cells (CFU-S) may not have a shoulder.

    Notes: Abstract. Seventy-six novel microsatellite markers with various simple sequence repeat (SSR) motifs are reported in this paper. They were generated on the basis of non-radioactive library screening procedures from flow-sorted rat Chromosome (Chr) 5-specific DNA, and were mapped in three rat backcross populations. Fifty-four of these markers mapped to Chr 5, while the other 22 mapped to other chromosomes of the rat genome. The marker D3Uwm8 is a new microsatellite marker for the rat syndecan 4 (ryudocan) gene. A genotyping protocol based on agarose gel electrophoresis is also provided in this paper.

    Notes: Abstract. Fifty-five novel rat microsatellite markers were isolated from libraries specific for rat chromosomes (Chrs) 1, 2, and 7. The markers were mapped in three backcross rat populations. Thirty of these markers mapped to Chrs 1, 2, or 7, while the other 25 mapped to other chromosomes. New markers for two genes, liver-specific transporter gene (Livtr) and insulin-responsive glucose transporter (Glut4), were also mapped to rat Chrs 9 and 10, respectively. Three provisionally assigned markers from previous studies were also confirmed. Detailed methodologies for the generation and enrichment of clones containing repeat sequences and for the isolation of chromosome-specific markers are presented, since they represent unique combinations and modifications of previous protocols. Such methods and the newly presented markers should be useful for both specific and general mapping studies in the rat.

    Notes: Breast cancer is an important disease site for chemopreventive intervention. The current in vivo rodent models for breast cancer do not adequately approximate the human disease. Thus, we developed a new series of models based on the direct introduction of activated oncogenes into in situ mammary ductal cells in the rat. So far we have introduced both activated ras and neu into the mammary parenchyma of the rat. Both oncogenes cause the development of mammary carcinomas without any additional exogenous intervention. The activated neu gene is, however, 200 times more penetrant than the activated ras gene. The carcinomas induced by both genes are more aggressive than those induced by chemical carcinogens. They are more often transplantable, locally invasive, and metastatic. Tumors arising from the introduction of the neu oncogene have a greater histopathological resemblance to human breast cancers than those associated with ras insertion. For example, within a week post-introduction of neu, lesions resembling ductal carcinomas in situ are observed in the rat mammary gland. These do not occur following ras introduction or exposure to chemical carcinogens. These models are helping to better define the cellular and molecular events associated with the multistage progression of breast cancer. They are also being used to develop models to evaluate chemopreventive approaches for breast cancer. For example, in the area of cancer prevention we have shown that hormonal intervention is more effective in preventing neu-initiated breast cancers than ras-initiated breast cancers. We have also shown that the chemopreventive monoterpene, d-limonene, can prevent ras-induced mammary tumors in this model. We therefore feel that these models will be useful in identifying and characterizing agents for chemoprevention in breast cancer, as well as in defining and evaluating intermediate biomarkers for chemoprevention trials.

    Notes: Individual genetically determined susceptibility to cancer as well as acquired epigenetic and genetic organ specific alterations are important considerations in choosing target populations for chemopreventive trials. These individual epigenetic and genetic alterations can also serve as potential biomarkers for chemoprevention clinical trials. In order to model these potential markers for chemoprevention investigations, we are examining a series of interrelated rat models.Inbred rats vary in their susceptibility to mammary cancer induction by environmental agents. For example, the WF strain is highly susceptible to chemically induced mammary cancer while the Cop rat is almost completely resistant. The F344 is intermediate in susceptibility to chemically induced mammary cancer. These differential susceptibilities are inherited in a dominant pattern. For example, resistance is due to the inheritance of Mcs gene(s) which likely act by altering the differentiation lineage of mammary epithelial cells.As tumors form in the mammary glands of these rats, they acquire additional epigenetic and genetic alterations. Epigenetic initiation is a very frequent cellular event following carcinogen exposure which may predispose cells to genetic change including allelic imbalance. For example, following a standard dose of NMU or DMBA over 1% of cells are epigenetically initiated. During the carcinogenesis process, initiated cells may acquire genetic change such as oncogene activation and allelic imbalance. Interestingly, the pattern of allelic imbalance appears to be an inherited trait. For example, a non-random loss of heterozygosity (LOH) in rat chromosome 1 following DMBA only occurs in certain strains, such as Cop rats. Interestingly this change does not occur following initiation by ionizing radiation.It will thus be important to identify these epigenetic and genetic events which underlie mammary carcinogenesis as well as determine their patterns of inherited predisposition and temporal occurrence.     Notes: Monoterpenes, found in a wide variety of plants, are a major component of plant essential oils. The unsubstituted monocyclic monoterpene limonene has been shown to prevent carcinogen-induced mammary cancer at both the initiation and the promotion/progression stages. This terpene also causes the complete regression of the majority of advanced rat mammary cancer when added to the diet. Modification of limonene by hydroxylation at various positions increases both its chemopreventive and therapeutic efficacy. For example, the naturally occurring hydroxylated limonene analog perillyl alcohol is 5-10 times more potent than limonene and has a similar therapeutic index.Several cellular, metabolic and molecular activities are associated with terpene exposure. These include induction of phase I and II hepatic detoxification enzymes, selective inhibition of protein isoprenylation, inhibition of CoQ synthesis, and induction of the mannose 6-phosphate/IGFII receptor and TGFβ.Due to the therapeutic efficacy of monoterpenes in experimental model systems, clinical evaluation of this class of compounds has begun in advanced cancer patients. A Phase I trial of limonene is in progress in the UK. Efforts in the US will target perillyl alcohol for Phase I testing. Pre-IND toxicology is currently being completed. Phase I trials are anticipated to begin in the Spring of 1995. We feel that the results of these therapeutic trials, if positive, will facilitate the development of current terpenes and more potent analogs for future chemoprevention trials.

    Description: This position paper is the outcome of a joint reflection by a group of international geographic and environmental scientists from government, industry, and academia brought together by the Vespucci Initiative for the Advancement of Geographic Information Science, and the Joint Research Centre of the European Commission. It argues that the vision of Digital Earth put forward by Vice- President Al Gore 10 years ago needs to be re-evaluated in the light of the many developments in the fields of information technology, data infrastructures, and earth observation that have taken place since. It focuses the vision on the nextgeneration Digital Earth and identifies priority research areas to support this vision. The paper is offered as input for discussion among different stakeholder communities with the aim to shape research and policy over the next 5-10 years.
