    Notes: C-phycocyanin and allophycocyanin from the green alga Spirulina platensis were isolated and crystallized by gel-acupuncture techniques. A novel two-step chromatographic procedure was used for purification. Blue hexagonal crystals were obtained by diffusing magnesium chloride into the protein solution for a week, followed by diffusion of PEG 6000 in order to complete the reduction of the solubility of the protein in the capillary tube used as a growth cell. In the case of allophycocyanin, crystals with a size of 0.4 × 0.3 × 0.3 mm were characterized by X-ray diffraction. They belong to space group P6322 with unit-cell parameters a = b = 102.04, c = 131.22 Å. The crystals of C-phycocyanin belong to either space group P6 or P63 with unit-cell constants a = b = 182.38, c = 60.87 Å, α = β = 90, γ = 120°. The crystals diffract beyond 2.4 and 2.5 Å resolution, respectively, using a rotating anode as an X-ray source.

    Notes: : Near-infrared reflectance spectroscopy (NIRS) was used for the authentication of 2 green asparagus varieties (Taxara and UC-157), grown in Huetor-Tajar (Granada, Spain) protected by the Quality Specific Appellation “Espárragos de Huétor-Tájar”. To develop the prediction model, the method chosen was modified partial least square (MPLS) regression. Two sample sets (N = 219 and N2 = 145 samples, respectively) were used to obtain the calibration equations. The standard error of cross-validation (SECV) and the r2 value were 0.082 and 0.97, respectively, for the 1st calibration set and 0.077 and 0.97 for the 2nd calibration set. The 2nd chemometric model obtained was tested with independent validation sample set (N3 = 74 samples), and the resulting values for standard error of prediction (SEP) and for r2 were 0.07 and 0.96, respectively. These results prove that NIRS is an accurate technology for identification and authentication of asparagus varieties and easily implemented in industry.

    Notes: Solutions of caffeic acid and hydroxytyrosol, the 2 orthodiphenols involved in ripe olive darkening, were used as model systems to optimize processing. Lineal, quadratic, and lineal interaction effects of pH and manganese content on oxidation rate of both orthodiphenols were significant. Manganese did not affect activation energy of caffeic acid oxidation but diminished that of hydroxytyrosol. Presence of manganese cations (0.34 mM) and high pH (9.0) also led to the darkest color in the 10 to 30°C temperature range but did not improve the color when added after caffeic acid and hydroxytyrosol oxidation. Results can lead to a faster oxidation period, which currently takes about a week, and darker olives.

    Notes: In vivo immunization and secondary culture techniques were used to generate cytotoxic T lymphocytes against a tumour cell clone obtained from a methylcholantrene-induced fibrosarcoma. Our CTLs differed from classical CTLs (Ly2 + and MHC restricted) in that our lymphocytes exhibited a non-MHC restricted cytotoxic activity directed to the original tumour and to other MCA-induced tumours, but have failed to demonstrate any killing activity against a wide range of tumour cell lines of diverse origin (including NK sensitive targets). Depletion and phenotypic studies demonstrated that these cells bear the Thy1.2 antigen but are negative for both Ly2 and L3T4 antigens. These CTLs may belong to a double negative subset (CD4−, CD8−) involved in the anti-tumour response.

    Notes: Isoantisera and syngeneic monoclonal antibodies have been produced against A7, a clone derived from a BALB/c methylcholanthrene-induced tumour named GR9. The antigens defined by the isoantisera and mAbs are widely distributed in a large number of GR9 clones as well as in other tumour cell lines of different aetiology and origin. Two GR9 clones with marked differences in H-2 expression and syngeneic in vivo growth (A7, which is H-2d-positive and highly immunogenic and B-9, which is H-2d-negative and highly malignant) were selected to further analyse the presence or absence of the antigens recognized by the syngeneic anti-tumour antibodies.Immunoprecipitation data with two different isoantisera revealed a group of proteins with a molecular weight of M, 65-70,000 present in both clones. Furthermore, immunoprecipitation with the A7-1 mAb again showed a single M, 65K band present in A7 and B9 clones. We conclude that the marked differences observed in the syngeneic growth of the two clones are not due to differences in tumour associated antigens (TAA), defined by syngeneic antibodies, and emphasize the role that class I H-2 antigens could play in tumour rejection.

    Notes: The GR9 tumour was induced with methylcholanthrene in a BALB/c mouse, adapted to tissue culture and cloned without any passage in vivo. GR9 clones were typed for H-2 with three monoclonal antibodies that define H-2Kd+ Dd, Kd and Dd antigens. A great heterogeneity of H-2d expression was found from clones which were Kd and Dd positive to clones Kd and Dd negative. These results were confirmed for A7 and B9 clones using immunoprecipitation with anti-H-2D.4 and anti-H-2K.31 alloantisera and SDS-PAGE analysis. In addition, the number of chromosomes per cell was heterogeneous amongst the clones, ranging from 38 ± 2 to pseudotetraploid clones which have 75 ± 2 chromosomes.GR9 clones were injected into syngeneic BALB/c mice to measure local tumour growth. We found that the growth correlated with the amount of H-2 antigen expressed, i.e. clones with low H-2d expression were highly malignant while clones with normal H-2d expression were highly immunogenic. Finally we found that BALB/c mice immunized against A7 (Kd, Dd-positive) protected against A7, as expected, but surprisingly also against B9 (Kd, Dd-negative).

    Notes: Storage of ripe olives in liquids with high acetic acid content under anaerobic conditions led to glucose accumulation, low ethanol, and high chemical oxygen demand (COD). In contrast, low acid concentrations and anaerobic conditions produced a high level of ethanol and slightly lower COD while aerobic conditions produced low glucose, ethanol and COD. Surface color of ripe olives improved when storage liquids were used in the second washing of the darkening step. Firmness also increased with increasing proportion of storage liquids reuse, although olives from highly acidic media were softer than those stored in low acid media.

    Notes: The effects of pouch material (aluminum, plastic), packing pH (3.8,4.3), acidifying compound (lactic, gluconic), and storage temperature were tested on polyphenol content and color of holding brines, surface color, and firmness of ripe olives. Using pouches of plastic films, pH 4.3–4.0 holding solution, and lactic or gluconic acid products with color and texture similar to traditional canned ripe olives were produced. They could be properly stored at room temperature (25°C). As storage temperature rose, color improved but texture was seriously degraded at 50°C.

    Notes: The effects of cultivar, salt concentration and temperature on changes in phenolic compounds during storage of ripe olives were studied, using a 23 factorial design. The effect of salt was negligible. Higher temperature increased phenols diffusion rate into brines for two cultivars assayed. The cultivars, Hojiblanca and Cacereña varied in poly-phenol content. Throughout storage there was hydrolysis of glucosides intially in flesh (oleuropein, verbascoside and luteolin 7-glucoside due to the acidic conditions. This effect caused the content of oleuropein in brines to always be lower than 1.5 mM and resulted in a continous increase in level of hydroxytyrosol

    Notes: Influence of several variables on ultrafiltration (UF) of green table olive brines was studied. Flux improved when temperature increased, although an interval of 30–40°C was recommended, especially when bentonite was used. To prevent darkening of regenerated brines for preservation for several days, sodium bisulfite had the best effect, although ascorbic acid was suggested. HPLC polyphenol analyses in the original brine, permeate and concentrate confirmed that UF removed a great amount of the polymerized polyphenols from the premeate. Simple phenols were not removed effectively, except for vanillin, caffeic acid and 4-hydroxybenzoic acid which were substantially eliminated.
