    Notes: We present the first synthesis of the life history and ecology of seahorses, compare relationships for seahorses with other marine teleosts and identify research needs. Seahorses occurred primarily amidst temperate seagrasses and tropical coral reefs. Population densities were generally low, ranging from 0 to 0·51 individuals m−2, but reached 10 m−2 in some patches. Inferred life spans ranged from 1 to 5 years. Seahorses consumed live prey and possibly changed diet as they grew. Growth rates are poorly investigated to date. Reproduction and mating systems are the best-studied aspects of seahorse ecology. The relationship between size at first maturity and maximum size in seahorses conformed to that for other marine teleosts. All seahorse species were monogamous within a cycle, but some were polygamous across cycles. Direct transfer of clutches to the brood pouch of the male fish made it difficult to measure clutch size in live seahorses. After brooding, males released from c. 5 to 2000 young, depending on species and adult size. Newborn young measured from 2 to 20 mm in length, which was a narrower size range than the 17-fold difference that occurred in adult size. Newborn body size had no relationship to adult size. Both eggs and young were larger than expected among marine teleosts, even when considering only those with parental care, but brood size at release was lower than expected, perhaps because the young were more developed. The size of adults, eggs and young increased with increasing latitude, although brood size did not. Considerable research is needed to advance seahorse conservation and management, including (a) fisheries-dependent and fisheries-independent abundance estimates, (b) age- or stage-based natural and fishing mortalities, (c) growth rates and age at first maturity, and (d) intrinsic rates of increase and age- or size-specific reproductive output. Current data confirm that seahorses are likely to be vulnerable to high levels of exploitation.

    Notes: Despite differences in climate and in timing of light leaf spot epidemics between Poland and the UK, experiments provided no evidence that there are epidemiological differences between populations of Pyrenopeziza brassicae in the two countries. Ascospores of Polish or UK P. brassicae isolates germinated on water agar at temperatures from 8 to 24°C. After 12 h of incubation, percentages of ascospores that germinated were greatest at 16°C: 85% (Polish isolates) and 86% (UK isolates). The percentage germination reached 100% after 80 h of incubation at all temperatures tested. The rate of increase in germ tube length increased with increasing temperature from 8 to 20°C but decreased from 20 to 24°C, for both Polish and UK isolates. Percentage germination and germ tube lengths of UK P. brassicae ascospores were less affected by temperature than those of conidia. P. brassicae produced conidia on oilseed rape leaves inoculated with ascospores or conidia of Polish or UK isolates at 16°C with leaf wetness durations from 6 to 72 h, with most sporulation after 48 or 72 h wetness. Detection of both mating types of P. brassicae and production of mature apothecia on leaves inoculated with mixed Polish populations suggest that sexual reproduction does occur in Poland, as in the UK.

    Notes: The potential use of DNA-based methods for detecting airborne inoculum of Leptosphaeria maculans and Pyrenopeziza brassicae, both damaging pathogens of oilseed rape, was investigated. A method for purifying DNA from spores collected using Hirst-type spore samplers and detecting it using polymerase chain reaction (PCR) assays is described. For both pathogens, the sensitivities of the DNA assays were similar for spore-trap samples and pure spore suspensions. As few as 10 spores of L. maculans or P. brassicae could be detected by PCR and spores of both species could be detected against a background of spores of six other species. The method successfully detected spores of P. brassicae collected using spore traps in oilseed rape crops that were infected with P. brassicae. Leptosphaeria maculans spores were detected using spore traps on open ground close to L. maculans-infected oilseed rape stems. The potential use of PCR detection of airborne inoculum in forecasting the diseases caused by these pathogens is discussed.

    Notes: In spite of displaying the most extreme dormancy and resistance properties known among living systems, bacterial endospores retain an alert environment-sensing mechanism that can respond within seconds to the presence of specific germinants. This germination response is triggered in the absence of both germinant and germinant-stimulated metabolism. Genes coding for components of the sensing mechanism in spores of Bacillus subtilis have been cloned and sequenced. However, the molecular mechanism whereby these receptors interact with germinants to initiate the germination response is unknown. Recent evidence has suggested that in spores of Bacillus megaterium KM, proteolytic activation of an autolytic enzyme constitutes part of the germination trigger reaction.

    Notes: Antibodies were raised against purified germinationspecific cortex-lytic enzyme (GSLE) from spores of Bacillus megaterium KM which neutralized the ability of GSLE to germinate permeabilized spores. Western blotting of dormant spore and vegetative cell fractions separated by SDS-PAGE demonstrated that GSLE is spore-specific and that 〉90% of the GSLE is associated with the dormant spore cortex peptidoglycan as a phosphorylated 63 kD pro-form, which could only be visualized after lysozyme digestion of the peptidoglycan. During germination, the 63 kD pro-form of GSLE is processed to release the active enzyme, which had an apparent molecular weight of 30 kD. Inhibitor studies demonstrated that GSLE activation occurs as part of the commitment reaction and thus represents the first-identified enzymatic event to occur during germination triggering. Proteins that cross-react with anti-GSLE sera are present in spore fractions of other species.

    Notes: Abstract The accessory gene regulator (agr) and staphylococcal accessory regulator (sar) loci are important regulators of toxin production in Staphylococcus aureus. In this study we examined how environmental conditions – degree of aeration and salt concentration – affect the transcription and translation of mRNAs for α- haemolysin (Hla) and serine protease (Ssp) via these pathways and influence the stability of these proteins. Using Northern analysis, we have confirmed earlier observations that sarA is involved in the upregulation of RNAIII, the effector molecule encoded by the agr locus. However, this effect was abolished in highly aerated cultures. While sarA does appear to have an up-regulatory effect on hla transcription that is independent of agr, we propose that the PC1839 (sarA) mutant produces less α-haemolysin activity mainly as a result of post-translational inactivation by proteases. The most obvious phenotypic feature of PC1839 (sarA) is the upregulation of proteases. In this study we show that ssp is repressed by SarA at the transcriptional level. Western analysis using an anti-α-haemolysin antibody identified a major breakdown product that is only present in the supernatant of strains that are overexpressing serine protease. We have also confirmed that agr exerts a significant regulatory influence on hla at the level of translation, as well as transcription. Finally, the addition of salt upregulates ssp transcription and dramatically downregulates transcription of hla, and is an example of an environmental parameter that affects toxin production independently of agr and sarA. How environmental signals are transduced to control α-haemolysin and serine protease production, activity and stability at multiple levels are discussed.
