    Notes: Abstract. The rational approach to pharmaceutical drug design begins with an investigation of the relationship between chemical structure and biological activity. Information gained from this analysis is used to aid the design of new, or improved, drugs. Primary considerations during this investigation are the geometric and chemical characteristics of the molecules. Computational chemists who are involved in rational drug design routinely use an array of programs to compute, among other things, molecular surfaces and molecular volume, models of receptor sites, dockings of ligands inside protein cavities, and geometric invariants among different molecules that exhibit similar activity. There is a pressing need for efficient and accurate solutions to the above problems. {Often, limiting assumptions need to be made, in order to make the calculations tractable. Also,} the amount of data processed when searching for a potential drug is currently very large and is only expected to grow larger in the future. This paper describes some areas of computer-aided drug design that are important to computational chemists but are also rich in algorithmic problems. It surveys recent work in these areas both from the computational chemistry and the computer science literature.

    Notes: Abstract The mouse is an ideal species for investigation at the interface of lung biology and lung function. As detailed in this review, there are well-developed methods for the quantitative study of lung function in mice. These methods can be applied to mice in both terminal and nonterminal experiments. Terminal experimental approaches provide more detailed physiological information, but nonterminal measurements provide adequate data for certain experiments. In this review, we provide two examples of how these models can be used to further understanding of the primary pathobiology of airway responsiveness in both the absence and the presence of induced airway inflammation. The first model is a dissection of chromosomal loci linked to the variance in airway responsiveness observed in the absence of any manipulation to induce airway inflammation. The second model explores the role of T-cell costimulatory signals in the induction of airway hyperresponsiveness. As the number of mice with targeted deletions of effector genes or insertion of informative transgenes grows, additional examples are likely to accrue.

    Notes: Near-field scanning optical microscopy (NSOM) has been used to image and record domains in thin-film magneto-optic (MO) materials (e.g., a Co/Pt multilayer). In this technique, a subwavelength-sized source or detector of visible light is placed in close proximity to a sample and raster scanned to read or write data on a scale inaccessible to traditional lens based systems. The technique, therefore, represents a hybridization of conventional magnetic and MO storage technologies. In the imaging mode, resolution of 30–50 nm has been consistently obtained, whereas in the recording mode, domains down to −60 nm have been written reproducibly. Data densities of −45 Gbits/in.2 have been achieved, well in excess of current magnetic and MO methods. A brief analysis of speed and other issues indicates that the technique may represent a viable alternative to these and other methods for anticipated high density data storage needs.

    Notes: A distance regulation method has been developed to enhance the reliability, versatility, and ease of use of near-field scanning optical microscopy (NSOM). The method relies on the detection of shear forces between the end of a near-field probe and the sample of interest. The system can be used solely for distance regulation in NSOM, for simultaneous shear force and near-field imaging, or for shear force microscopy alone. In the latter case, uncoated optical fiber probes are found to yield images with consistently high resolution.

    Notes: 2 O production, and denitrification in soil amended with animal slurries were studied. The effects of the abiotic factors on the biological processes were monitored for 8 wk in repacked soil cores amended with pig or cattle slurry. The soil cores were incubated at 43, 57, and 72% water-filled pore space (WFPS) and at 10, 15, and 20°C with or without addition of 10% acetylene. The amount of N2O lost at 72% WFPS corresponded to 8 to 22% of the slurry's NH+ 4 content, but to only 0.01 to 0.9% at 43 to 57% WFPS. Denitrification losses at 72% WFPS accounted for 17 to 58% of the slurry's NH+ 4 content, but for only 0.01 to 1.2% at 43 to 57 WFPS. The amount of available C accounted for by denitrification was 8 to 16% of total respiration at 72% WFPS, but only 0.03 to 0.4% at 43 to 57% WFPS. Both N2O production and denitrification peaked earlier in the cattle-slurry treated soil than in the pig-slurry treated soil, whereas the total N loss was greatest from the latter. Neither amendments nor soil water contents seemed to affect the Q10-values for the CO2 production, resulting in values between 1.6 and 2.6 at 72% WFPS, N2O production and denitrification had Q10-values ranging between 3.3 and 5.4. High temperatures enhanced both aerobic respiration and denitrification by consuming oxygen, resulting in strong sensitivity of denitrification to temperature.

    Notes: Summary In low phosphate, vitamin D-deficiency rickets normal mineralization is reversibly arrested, and rickets is thus a suitable model for studying factors influencing the mineralization process. Partly on the basis of their distribution within the growth cartilage, the so-called matrix vesicles are considered to play an important role in the process of mineralization and it has been claimed that the distribution pattern is the same in rickets as in normal animals. With the use of modern stereological techniques, our group recently demonstrated a matrix vesicle distribution between the zones of the epiphyseal growth plate in normal rats different from that described earlier. The same bimodal distribution pattern was observed in rachitic rats in the present study, the highest volume density being found in the resting and upper hypertrophic zones and the lowest in the proliferative zone. The volume density differences are explained by differences in the number of vesicles between zones, the variation in mean caliper diameter being small. Our findings are discussed in relation to the proposed theories on matrix vesicle origin. The results seem to support the dynamic cell debris theory for matrix vesicle origin presented earlier, but the existence of subpopulations of matrix vesicles with a specialized function and origin cannot be ruled out.
