    Notes: Abstract A method for purifying TBP2 from N. meningitidis has been developed using affirnity chromatography on Tf-agarose followed by ion exchange chromatography; the Tf-binding activity of fractions was evaluated by a dot-blot technique. This method allowed the purification of the TBP2 in milder conditions than those used previously and to a high degree of homogeneity as was demonstrated by Coomassie brilliant blue or Silver training after SDS-PAGE electrophoresis. The SDS-PAGE profile of the material obtained in the Tf-agarose affinity chromatography step shows only two detectable proteins, identified as the TBP1 and the TBP2, with a small amount of contamination. Passage through a MonoQ HR anion exchange column, allowed the isolation of TBP2 in the absence of TBP1. Our results demonstrate the conservation of the antigenic reactivity of this protein, which produces monospecific serum with the antibodies elicited reacting exclusively with the TBP2 in whole outer membrane vesicles.

    Notes: Abstract: Peroxidase-conjugated transferrin was used to detect transferrin receptors both in intact outer membrane vesicles (OMVs) from Neisseria species in a dot blot assay, and in SDS-PAGE-separated OMV proteins after transferring to introcellulose membranes. All N. meningitidis strains produced transferrin receptors after culturing in either iron sufficiency or iron restriction although expression was higher in the latter case, whereas only six N. lactamica and two N. sicca (among 20 commensal species) were able to bind transferrin. Molecular mass (MM) of the receptors were mainly between 78 kDa and 85 kDa (87.5% of strains), 12.5% had receptors with MM close to 70 kDa, and 5% showed receptors with MM over 85 kDa. Our results confirm the molecular mass heterogeneity of the transferrin receptors in N. meningitidis, completely disagree with the ‘universal’ 98 kDa receptor proposed by some authors, and show a low expression of the receptor in commensal Neisseria.

Tritrichomonas foetus was cultivated in Diamond's medium without phosphates or agar; 10% glycerol was added to the medium and the protozoa were frozen slowly and stored at −28 or −95 C. About half of the trichomonads died within the first day at either temperature. Thereafter, they continued to die off slowly during storage at −28, and none remained alive at or after 1024 days. New cultures could not be initiated with them after 64 days. At −95, almost as many trichomonads were alive after 256 days as after 1 day. Thereafter, however, they decreased in numbers, but 11% were still alive after 2048 days (5.6 years). The survivors appeared in good condition, and new cultures were readily initiated with them.

    Notes: SYNOPSIS. Tritrichomonas foetus survived much better on extended storage at -95 than at -28d̀C following slow freezing in the presence of 1.0 M glycerol. There was no significant difference between these temperatures in survival up to 8 days, but thereafter the protozoa continued to die off slowly at -28d̀, whereas their numbers remained essentially constant at -95d̀ for 128 to 256 days. The trichomonads' motility was much better after storage at -95d̀ than after storage at -28d̀, and fresh cultures could be initiated from the former much more readily.Other constituents of the suspending medium besides glycerol affect the survival of the protozoa upon freezing. Survival was much better when the protozoa were frozen in the original Diamond's trypticase-yeast extract-maltose-cysteine-ascorbic acid-serum medium in which they had been grown than when they were frozen in physiological salt solution or in fresh Diamond's medium. There was no significant difference between survival in the latter two suspending media. The speed and time of centrifugation needed to remove the trichomonads from the medium in which they had been grown had no effect on their survival upon subsequent freezing. Presumably some product or products of the trichomonads' metabolism have an additional protective action which supplements that of glycerol.When frozen in the original Diamond's medium in which they had been grown plus 1.0 M glycerol, an average of 15% of the trichomonads were alive after 128 days' storage at -28d̀ and an average of 38% were alive at -95d̀C. When frozen in physiological salt solution plus 1.0 M glycerol, these percentages were 8% and 12% respectively.

    Notes: Abstract The lack of experimental models suitable for the study of meningococcal pathogenicity led us to investigate if thoses actually in use (culture in iron-restricted media and animals models) provide results comparable with the responses observed in vivo during infection. In this work we studied three invasive strains cultured both in laboratory media and in human plasma, analysing the immune response elicited in mice against membrane antigens and comparing them with those seen using homologous human convalescent sera. Outer membrane protein profiles observed after culture in plasma were different and more complex than those obtained after growth in laboratory media. Analogous differences were observed in the antigenic profiles, detecting some antigens recognized by human, but not mouse sera, and vice sersa. However, the response to one of the major iron-regulated outer membrane antigens, the transferrin binding protein 2 (TBP2), was unaffected by the culture medium or the model, human or mouse, used for the analysis. In conclusion, we have found that results of antigenic analysis change depending on the culture conditions and animal models used. For the meningococcal antigen TBP2, growth in iron-restricted laboratory media and a mouse model provide results which correlate well with those observed using convalescent human serum from individuals recovered from infections. We suggest that careful analysis and evaluation of experimental results and their comparison with in vivo elicited immune responses are essential in order to get accurate extrapolations for experimental vaccine designs.

    Notes: Abstract When grown under iron restriction, Neisseria meningitidis expresses new outer-membrane proteins, some of which are antigenic and potentially useful as vaccine components. This is particularly relevant to N. meningitidis serogroup B, against which neither polysaccharide nor conjugate vaccines are effective. We investigated recognition of N. meningitidis serogroup B outer-membrane antigens by three sera from patients recovered from meningitis. Recognition of antigens from the homologous strain provided information on in vivo expression during infection and immunogenicity, while cross-reactivity with outer membrane proteins from the other two strains and from another five strains in our collection allowed evaluation of antigenic heterogeneity. Our results demonstrate that transferrin-binding protein 2 (TBP2) is immunogenic in humans, to varying degrees depending on the strain, and that TBP2s (like the equivalent proteins of Haemophilus influenzae type b) are among the most important iron-regulated outer membrane antigens expressed during infection. Other immunogenic outer membrane proteins (some iron-regulated) are also expressed during infection; in a previous study in mouse, three of these proteins (with Mr of 50, 70 and 77 kDa) did not induce an immune response. Our cross-reactivity data provide some support for Robki et al.'s two-group classification of N. meningitidis strains, and provide evidence against the possibility that the antigenic domains shared by the TBP2s of all N. meningitidis strains induce immune responses in vivo.

    Notes: Abstract The effect of praziquantel in vitro at concentrations of 5, 50 and 500 ppm for 1 h resulted in the progressive breakdown of the tegument and in morphologic distortion of adultEchinococcus granulosus when compared to controls. Scanning electron microscopy of all specimens treated in the various concentrations of praziquantel showed loss of most, if not all, of the rostellar hooks and changes in the structure of the suckers. Many of the tapeworms immediately detached from the host's gut upon being placed in the drug, and all treated cestodes exhibited contraction or swelling, particularly in the penultimate proglottid. Intense contraction was apparent in the worms exposed to the higher drug concentrations. Characteristic conical microtriches on the terminal proglottid, as observed in the control specimens, became fused and matted when exposed to 5 ppm of praziquantel. At a drug concentration of 50 ppm, the tegumental surface developed grooves or furrows between clumps of fused microtriches, while 500 ppm caused production of holes within the denuded tegument of the parasite. Ovoid bodies, presumed to be eggs, were observed on the outer surfaces and just below the tegument of tapeworms treated with concentrations of 50 ppm. These structures also appeared to adhere to the outer surfaces of specimens exposed to 500 ppm. In view of the foregoing, special care should be taken in handling and disposing of feces from infected or suspect dogs after praziquantel treatment, since the breakdown in the tegumental surface ofE. granulosus presumably results in the release of potentially infective eggs.

    Use of low-cost daylighting system: custom design and application consequences in an office environment (2011)

    Description: Nowadays, many working environments are located within facilities that are not visually connected with the exterior. This is due to the new architectural paradigms as regards the design, based on the possibility to substitute natural light for artificial lighting even in daytime. Accordingly, the need of reducing energy costs and the user's wish to optimize lighting and visual comfort levels raises the option of innovative natural lighting systems implementation. For a luminous retrofitting case, located in Mendoza, Argentina, a specific methodology used determined that the more adequate natural lighting strategy would be redirecting direct sunlight, working on the reflected light design. The methodology proposed is formed by an analysis of the initial situation, conditions modelization and design resolving proposals, through luminous evaluation, real and virtual scale model construction, and performance evaluation using heliodon and simulation software. This work points to use natural lighting to reduce energy consumption and get better luminous ambient through the application of a low-cost daylighting system. In the mentioned case, strategies of redirection of the solar component capable of optimizing indoor lighting levels up to a 40% were applied.
