Selective Cleavage of Glycosides, V. - Cleavage of Hepta-0-acetylamygdalin by Means of Dichloromethyl Methyl EtherHepta-O-acetylamygdalin (1) is cleaved selectively by means of dichloromethyl methyl ether in the presence of ZnCl2 to give good yields of hepta-U-acetyl-α-gentiobiosyl chloride (2) and α-chlor-α-formylbenzylcyanide (3) which can be hydrolyzed to DL-mandelic acid (5) by means of hot concentrated hydrochloric acid. The mechanism of the cleavage reaction has been investigated in model experiments. Probably 3 is formed via the intermediary carbenium ion 3a. - Methyl hepta-O-acetyl-β-gentiobioside which may exist in a hitherto unknown mode (Schmp. 136-137°C) can also be transformed selectively into 2 by means of dichloromethyl methyl ether in the presence of ZnCl2.
Notes:
