    Notes: Abstract. The effects of UV-B radiation generated in the laboratory and as a component of sunlight on the viability and particular biochemical activities of the bacterium Staphylococcus aureus have been examined. UV-B radiation progressively inhibits protein synthesis (assayed as 3H-alanine incorporation) and kills cells. Cell respiration, and RNA and DNA synthesis (3H-uridine and 3H-thymidine incorporation) were not greatly affected by UV-B irradiation. The OH˙ and 1O2-free radical scavengers protected cells against killing and inhibition of protein synthesis by UV-B, suggesting that such radicals mediate the effects of UV-B on this organism. A similar protective effect using a ferric ion chelator suggests an important role for metallic ions in UV-B lethality.

    Description / Table of Contents: Zusammenfassung Die Fluoreszenz des Laserpigments Coumarin-1 (C1; 7-Diethylamino-4-methyl-coumarin) reagiert auf die Anwesenheit von Peroxodisulfat (S2O 8 2− ). Die Emissionsintensität nimmt mit steigender Peroxodisulfatkonzentration ab. Die Abnahme wird einem Löschvorgang zugeschrieben, der mit einer Oxidation des Grundzustands durch Peroxodisulfat verbunden ist. Die Kinetik der Reaktion wurde bei 382 nm spektrophotometrisch untersucht. Sie verläuft nach zweiter Ordnung (nach erster Ordnung bezüglich jedes Reaktanden). Die Reaktionsgeschwindigkeit istpH-abhängig; in alkalischen Medien werden höhere Geschwindigkeitskonstanten gefunden. Zusatz von Natriumdodecylsulfat (SDS) hemmt die Oxidation beträchtlich.
    Notes: Summary The fluorescence of coumarin-1 laser dye (C1; 7-diethylamino-4-methyl-coumarin) proved to be sensitive to the presence of peroxodisulfate (S2O 8 2− ). The emission intensity decreases with increasing in peroxodisulfate concentration. The decrease is assigned to a quenching process connected with a ground state peroxodisulfate oxidation of the dye. The kinetics of the reaction have been investigated spectrophotometrically at 382 nm. The reaction follows second order kinetics, being first order for each reactant. The reaction rate ispH dependent; higher rates are observed in alkaline media. Addition of sodium dodecyl sulfate (SDS) retards the oxidation process remarkably.

    Description / Table of Contents: Zusammenfassung Die Löschung der Fluoreszenz von 1,4-Dimethoxynaphthalin (1) und 1,8-Dimethoxynaphthalin (2) durch Tetraphenylporphyrin (3), 9,10-Diphenylanthracen (4) und 3-Cyano-4-phenyl-6-(p-tolyl)-pyridin-2-on (5) in Chloroform wurde untersucht. Die Löschung verläuft über einen Resonanzenergietransfermechanismus. Die Geschwindigkeitskonstante für den Energietransfer (k ET) ist bei gleichem Akzeptor für den Donor2 niedriger als für1. Daraus läßt sich schließen, daß in allen untersuchten Systemen der sterische Effekt über den Effekt des Ionisierungspotentials dominiert. Die berechneten kritischen Transferdistanzen betragen 17–72 Å. Im Gegensatz zu diesen Beobachtungen verläuft der vorherrschende Relaxationsmechanismus bei der Fluoreszenzlöschung von1 und2 durch 7,7,8,8-Tetracyanochinon-dimethan (6) über einencharge-transfer-Prozeß. Die Einflüsse von Temperatur und Geometrie der Verbindungen auf die Effizienz der Fluoreszenzlöschung von1 und2 durch den Akzeptor6 wurden ebenfalls untersucht.
    Notes: Summary The fluoroescence quenching of 1,4-dimethoxynaphthalene (1) and 1,8-dimethoxynaphthalene (2) by tetraphenylporpyrin (3), 9,10-diphenylanthracene (4), and 3-cyano-4-phenyl-6-(p-tolyl)-pyridin-2-one (5) has been studied in chloroform solution. The quenching occursvia a resonance energy transfer mechanism. The rate constant for the energy transfer (k ET) of donor2 is slower than that of1 by the same acceptors, indicating that the steric effect dominates the ionization potential effect in all systems. The calculated critical transfer distances (R 0) are 17–72 Å. In contrast, charge transfer is the predominant pathway of electronic deexcitation in the fluorescence quenching of donors1 and2 by 7,7,8,8-tetracyanoquinone-dimethane (6) in chloroform. The roles of temperature and geometrical structure of the donors on the efficiency of fluorescence quenching of1 and2 by acceptor6 have also been studied.

    Notes: Abstract Streptomyces sp A21 is a cellulolytic strain isolated from soil which was assigned to the genus Streptomyces on the basis of distinctive morphological features. A genomic library of A21 DNA has been constructed and transformed into Escherichia coli K-12 using a high-copy-number vector. One of the recombinant plasmids activates the cryptic bgl operon when inserted into appropriate strains. The complete sequence of the 1629-bp A21 DNA fragment has been determined. The analysis revealed the presence of an ORF whose putative product shows a high degree of similarity to RNA polymerase sigma factors; we therefore designated the gene psfS (Putative sigma factor, S treptomyces). Mapping of the 5′ terminus of transcript by primer extension indicated that PsfS induces transcription initiation within the bgl promoter-silencer region.

    Notes: Ultrahigh molecular weight polyethylene has been irradiated using a cobalt 60 source to give received dose between zeor and 50 Mrad. Irradiacted specimens were subjected to tensile characterization, dynamic mechanical analysis, and differential scanning calorimetry. Changes in tensile and dynamic mechnaical properties following irradiation arise from both molecular rearrangement and from increased crystallinity following scissionof long interlamella tie chains. The effects of post-irradiation aging on mechanical properties are associated with increasing crystallinity resulting from decomposition of metastable groups formed in the amorphous region during irradiation. Irradiated materials have been subject to sinusoidal stressing between 0.275 and 0.55 of yield stress for 100,000 cycies, and changes in mechnaical and physical properties measured. Increased resistance to creep during stressing was observed with the irradiated materials, behavior which is consistent with previsouly observed changes in crystallinity and crosslink density. Overall property changes measured following stressing were small compared with those induced by the initial irradiation.
