    Notes: Solute transport systems are one of the major ways in which organisms interact with their environment. Typically, transport is catalysed by integral membrane proteins, of which one of the largest groups is the ATP-binding cassette (ABC) proteins. On the basis of sequence similarities, a large family of ABC proteins has been identified in Arabidopsis. A total of 60 open reading frames (ORFs) encoding ABC proteins were identified by BLAST homology searching of the nuclear genome. These 60 putative proteins include 89 ABC domains. Based on the assignment of transmembrane domains (TMDs), at least 49 of the 60 proteins identified are ABC transporters. Of these 49 proteins, 28 are full-length ABC transporters (eight of which have been described previously), and 21 are uncharacterized half-transporters. Three of the remaining proteins identified appear to be soluble, lacking identifiable TMDs, and most likely have non-transport functions. The eight other ORFs have homology to the nucleotide-binding and transmembrane components of multi-subunit permeases. The majority of ABC proteins found in Arabidopsis can, on the basis of sequence homology, be assigned to subfamilies equivalent to those found in the yeast genome. This assignment of the Arabidopsis ABC proteins into easily recognizable subfamilies (with distinguishable subclusters) is an important first step in the elucidation of their functional role in higher plants.

    Notes: Gradients of gene expression in the developing fourth leaf of the grass Lolium temulentum L. strain Ba 3081 were investigated using cell-free translation and Northern analysis of RNA extracted from succesive leaf segments. The transition from growth at the leaf base to photosynthesis in the blade was reflected in relative levels of transcripts from the plastid genome (psb A, pet A and rbc L, encoding plastid polypeptides) and nuclear genome (cab and PCR, encoding plastid polypeptides, and histone H3 and PAL encoding extra-plastidic polypeptides). These data combined with information on polypeptide synthesis in vivo indicate that post-transcriptional regulation plays an important part in controlling gene expression in this tissue.

    Notes: Cellular and compositional changes related to tissue growth and assembly of the photosynthetic apparatus were established with reference to the developmental gradient along the expanding fourth leaf of Lolium temulentum L. Ba 3081. The number of cells and the fresh weight per leaf segment fell with increasing cell age (distance from the leaf base). Components of the photosynthetic apparatus increased in concentration towards the more mature part of the leaf. Appreciable levels of chlorophyll and carotenoid could be detected in basal tissue enclosed in the sheaths of previous leaves prior to emergence into full light. The distributions of NADPH-protochlorophyllide oxidoreductase (PCR. EC 1.6.99.1) and cytochrome f along the age gradient were visualized by Western blotting. Leaf base tissue contained two forms of PCR, 41 and 39 kDa, the smaller of which diminished with increasing cell age and proximity to full light. Cytochrome f comprised a 52 kDa species, which also declined with distance from the base, and a group of polypeptides at around 30–33 kDa which greatly intensified with tissue maturity. The significance of multiple forms of plastid proteins and the role of the light gradient penetrating the leaf sheath in regulating growth and plastid assembly processes are discussed.

    Notes: China's soils tend to be phosphate deficient. Application of phosphorus fertilisers to the soil is yield and cost ineffective as much of the phosphate applied is rapidly locked-in and is inaccessible to the crop. Chinese Institutes have established intensive wheat breeding programmes to generate wheat varieties that produce adequate yields and grain quality in such soils. Three such wheat cultivars have been identified with good performance characteristics in the field. These three cultivars are thought to harbour chromosome translocations that may confer enhanced phosphate scavenging abilities to the plants. The isolation and study of the expression of high-affinity phosphate transporters in tissues of these wheats, in two of the donor wheatgrasses and in another widely planted Chinese wheat variety is presented and the first full-length sequence of a wheat phosphate transporter and partial clones of several other putative phosphate transporters are reported. Relative quantitative reverse-transcription – polymerase chain-reaction was used to demonstrate that different phosphate transporters have different expression patterns within a given variety and respond differently to phosphate deprivation. The significance of the genetic background for these findings and for the different phosphate acquisition properties of the wheats under study is discussed.

    Notes: Abstract Cultured oat (Avena sativa L. — naked form) aleurone protoplasts were employed as a model system for following changes which accompany the development of vacuoles during in-vitro incubation. Over a 5-d period, the aleurone grains progressively grew and fused to form a large central vacuole and the volume of the protoplasts increased sevenfold. The growth of the vacuole was accompanied by a progressive acidification of the vacuolar sap. Vacuolation was inhibited by high concentrations of mannitol and by cycloheximide and cordycepin applied at various times during the incubation period. Neither cycloheximide nor cordycepin affected the initial phases of vacuolation but cycloheximide retarded subsequent stages, particularly if added early in the incubation. Cordycepin inhibited only the later stages of vacuolation. Radiolabelling studies identified at least three novel microsomal proteins, with relative molecular masses of approximately 34, 47 and 48 kDa, which appeared during vacuolation and whose synthesis was markedly affected by these inhibitors.

    Notes: Abstract A non-competitive enzyme-linked immunosorbent assay (ELISA) which enables detection of as little as 0.1 ng cytochrome f in leaf extracts has been developed. No evidence for specific or non-specific interference by proteins other than cytochrome f was found. The assay was applied to a comparative study of age-related changes in the cytochrome f content of leaves of Festuca pratensis Huds. cv. Rossa, and a non-yellowing mutant genotype (Bf993) having a lesion in the mechanism responsible for thylakoid membrane disassembly. Cytochrome f in senescent leaves of the latter genotype was found to be present at significantly higher levels than in the wild-type, implying an inability on the part of the mutant to degrade this protein. The results obtained by ELISA were confirmed by antibody probing of Western blots.

    Notes: Novel fundamental solutions for elastodynamics of fully incompressible media are derived, by reason of their many applications to engineering and physical problems. It is demonstrated that, when Poisson's ratio equals 1/2, the frequency domain fundamental solutions (Stokes's solutions) are independent of the dilatational wave velocity, which tends to infinity. Consequently, dynamic problems involving incompressible materials can be rigorously analysed by the boundary element method without special difficulty. Numerical solutions to geotechnical problems of incompressible half-spaces are presented to demonstrate the applicability and accuracy of the formulations.

    Notes: The axial and lateral load-displacement response of single piles and pile groups during the secondary consolidation (creep) phase of deformation is obtained by idealizing the soil as a linear viscoelastic material. The boundary element method is used to model the soil halfspace and the piles, which are assumed to behave elastically, are idealized as beam-columns. By using the correspondence principle, the problem is solved in the Laplace transform domain and the final solution is obtained by numerical inversion of the transformed data. The salient features of the viscoelastic response are identified in several non-dimensional plots which encompass the spectrum of geometrical and material parameters commonly encountered in practice. The results of the analysis compare favourably with the limited data available in the literature.
