    Notes: Abstract A comparison has been made of the amount of DNA coding for ribosomal RNA present in a polyploid series of plants of Datura innoxia. The plants which were produced by in vitro culture of pollen grains were haploid, diploid, triploid, tetraploid and hexaploid. In all instances there was a similar proportion of DNA coding for ribosomal RNA. The implications of these results are discussed, and the data compared with that available from other series of plants of different DNA content or ploidy.

    Notes: Road traffic accident involvement rates show clear age and gender differences which may in part be accounted for by differences in risk perception and perceptions of driving competence. The present study extends and replicates that of Matthews and Moran (1986). Young (18–30 years) and older (45–60 years) male and female drivers responded to a questionnaire on perceived accident risk and driving competence (judgment and skill) with respect to themselves and four target groups, and also rated a series of videotaped driving sequences with respect to likelihood of accident occurrence and perceived driving competence. Results showed that effects of rater characteristics were generally confined to the questionnaire. Younger males were perceived as most likely to experience an accident and were judged to be lower than other groups in driving competence. Younger groups showed little bias against older groups and vice versa, but gender-related bias was apparent. The findings of Matthews and Moran were generally confirmed. The results are discussed with reference to four main issues: (1) demographic bias effects—which are generally weak; (2) stereotyping on the basis of gender and/or age of driver; (3) group-specific bias; (4) self-appraisal bias.

    Notes: Summary Synchronous populations of diploid cells ofChlamydomonas reinhardi were exposed to a bifunctional (Nitrogen Mustard) and a monofunctional (Diethyl Sulphate) alkylating agent at various stages of the pre-meiotic interphase and meiosis, and the effect on recombination in the arg 1/pab 2 region of linkage group I assessed. Both compounds affected recombination—the monofunctional one depressing it during two short periods, one just pre-meiosis when the main DNA duplication for meiosis is thought to occur, and the other in early prophase. The bifunctional compound on the other hand depressed recombination over an extended period. These effects have been related to the ability of the agents used to affect DNA synthesis and the different temporal patterns of response interpreted in terms of differential repair activities. The results provide additional evidence of the existence of two periods during which recombination can be affected.

    Notes: Abstract In order to detect low copy number sequences in pea using biotin-labelled probes we optimised some aspects of the in situ hybridization technique. We found protoplast preparations to be superior to standard squashes in terms of their signal: noise ratio. Heat and alkali denaturation of chromosomal DNA were both more effective than acid denaturation. A comparison of antibody-fluorochrome and streptavidin-enzyme conjugates showed the streptavidin-alkaline phosphatase conjugate to be the most sensitive detection system. Using the optimised method, we were able to detect a single site for a 13.5 kb legumin gene clone.

    Notes: Summary Variation in cell population size and cell weight in the cotyledons are important factors in determining seed weight in Pisum sativum. The regulation of these parameters as well as that of growth rate has been examined. The use of reciprocal crosses between varieties of contrasting seed size has allowed the recognition of two systems of control—an intrinsic one dependent on the seed's own genotype, and the extrinsic control of the maternal environment. It is shown that the use of particular kinds of crosses can aid in discriminating the separate roles of sinks and sources as determinants of seed size.
