Zusammenfassung Hydration und partielles spezifisches Volumen stromafreier Chloroplaste wurden aufgrund von Auftriebs- und Dichtemessungen in verdünnten wässrigen Glyzerinlösungen bei 20 °C untersucht. Das partielle spezifische Volumen wurde im Mittel zu $$\left\langle {\bar v_2 } \right\rangle = 0,833 \pm 0,002$$ cm3/g bestimmt. Für die Hydration wurden im Durchschnitt 〈δ1〉 = 0,85 ± 0,04 g H2O/g unbehandelte stromafreie Chloroplasten und 1,53 ± 0,08 g H2O/g ultraschall-behandelte stroma-freie Chloroplasten gefunden. Entsprechende Messungen an Rinderserumalbumin ergaben $$\left\langle {\bar v_2 } \right\rangle = 0,741 \pm 0,001$$ cm3/g und 〈δ1〉 = 0,18 ± 0,01 g H2O/g trockenes Protein.

Summary The hydration and partial specific volume of stroma-freed chloroplasts are determined by buoyant weight and density measurements in diluted aqueous glycerol solutions at 20 °C. For the partial specific volume an average value of $$\left\langle {\bar v_2 } \right\rangle = 0.833 \pm 0.002$$ cm3/g was measured. The average hydration values were found to be 〈δ1〉 = 0.85 ± 0.04 g of H2O per g of untreated stroma-freed chloroplasts and 1.53 ± 0.08 g of H2O per g of ultrasonicated stroma-freed chloroplasts. Corresponding measurements with bovine serum albumin yielded $$\left\langle {\bar v_2 } \right\rangle = 0.741 \pm 0.001$$ cm3/g and 〈δ1〉 = 0.18 ± 0.01 g of H2O per g of dry protein.
