    Notes: A non-glycosylated form of horseradish peroxidase c extracted from Escherichia coli inclusion bodies and refolded in the presence of haem and Ca2+ ions has been used to grow protein crystals suitable for X-ray diffraction analysis. The crystals are prisms in the trigonal space group P3112 or P3212 with a = b = 158.9 and c = 114.3 Å, and diffract to 1.9 Å. There are four molecules, each of 34 kDa, in the asymmetric unit. The molecules of the asymmetric unit are related by approximate translational symmetry, resulting in pseudo-centerings. Data to approximately 15 Å can thus be described by a lattice of a′ = b′ = 91.7 Å and c′ = 57.1 Å, α = β = 90° and γ = 120°, including four molecules.

    Notes: Electrolyte and water balance studies have been carried out on 20 patients with total body surface burns ranging from 15 to 85%. All patients were treated with 0.5% aqueous AgNo3 applied directly and continuously to the burn wound. The balance studies showed that the loss of Na through the burn wound was directly related to the square area of burn surface and was constant from day to day. Based on this steady and predictable loss of Na in the uninfected burn, a formula has been derived predicting the daily Na replacement necessary to cover the obligatory loss via the burn wound plus the requirements of normal metabolism. This formula has provided an accurate prediction of daily Na requirements in the treatment of thermal burns of widely varying extent, in patients of widely different ages, as judged by water and electrolyte balance studies.The formula provides a useful guide to the level of Na replacement required. Exact tailoring must be carried out to fit the needs of each patient. The direct monitoring of the accuracy of Na replacement by way of the serum Na level has been found cumbersome but intermittently necessary. In the burn patient in metabolic balance and taking a reasonable diet, the level of urinary excretion of Na has been found to be an accurate prediction of serum Na level. A urinary excretion of 40 mEq of Na per liter or above has uniformly reflected a normal serum Na level. The urinary Na has been used as an indicator of accurate replacement therapy replacing the majority of serum Na determinations.

    Notes: Abstract We have used DNA-mediated gene transfer to introduce intoDrosophila melanogaster cells DNA sequences for which no selective criteria exist. We have introduced a Drosophila heat-shock locus into culturedDrosophila cells by calcium phosphate cotransfection with the copia vector pCV31gpt and selection for xanthine utilization. We recovered cell lines containing between three and about 50 copies of both pCV31 gpt and pMH10A, a cloned 87A7hsp70 heat-shock locus that encodes a mutant 40,000-dalton heat-shock protein (hsp40). The stable inheritance of the transformed DNAs argues that the input DNAs have integrated into the genome. We show that this is indeed the case for one cell line by cloning back the transfected DNA and detecting the flanking chromocentral sequences by in situ hybridization. Surprisingly, the integrated hsp70 genes are not expressed. This report represents the first example of the cointroduction of DNA sequences intoDrosophila cells by cotransfection with a dominant selectable marker.

    Notes: Abstract At the nonpermissive temperature, the shibirets gene of Drosophilacauses adult paralysis and cell death during development. We have used DNA-mediated gene transfer with wildtype genomic DNA to rescue the lethal phenotype of shits cultured cells and have isolated cell lines which grow at the nonpermissive temperature of 29° C. By using restriction endonucleases, we confirm that this rescue is DNA sequence dependent. Further, cells cotransfected with wildtype genomic DNA and plasmid DNA contain plasmid sequences when selected for survival at 29° C. This confirms the uptake of DNA by the transformed cells and provides a simple temperature selection technique allowing coselection of genes which cannot themselves be selected for.
