    Notes: Summary Routine electrophoretic surveys for genetic variation in natural populations depend primarily upon detecting differences in the net charge carried by a protein. We have calculated the proportion of base substitutions which would yield an electrophoretically detectable mutant protein, and the relative mutation rates among different charge classes, under a variety of simplifying assumptions. These calculations indicate that: (i) only 25 per cent of all single base mutations would lead to a charge change on a protein molecule. (ii) five distinct classes of electrophoretic variants can be generated from a specified protein by single base substitutions. (iii) the relative mutation rates differ markedly among the different charge classes which can be generated by single base substitutions. The estimates of the proportion of electrophoretically detectable mutant proteins and relative mutation rates among charge classes were relatively robust to changes in assumptions concerned with the kind and site of base substitutions and the amino acid composition of the protein.

    Notes: Eighty-three third backcross lines which comprise a set of near isogenic lines (NIL's) of the barley cultivar ‘Clipper’ but each carrying a different chromosomal segment from Hordeum spontaneum, marked with a distinct isozyme, were tested for resistance to three races of the barley leaf rust pathogen (Puccmia hordei). Fourteen lines showed resistance to at least one race and three showed resistance to all three races. The resistance in two of these lines was controlled by separate, single partially dominant genes. In one case the resistance gene named Rph1O was on chromosome 3 and linked (r = 0.15 ±0.05) with the isozyme locus Est2. In the second case, the gene (Rph11) was on barley chromosome 6 and linked (r = 0.07±0.02) with the isozyme locus Acp3 and (r = 0.11±0.02) with Dip2.

    Notes: Most genes for resistance to barley leaf scald map either to the Rrs1 locus on the long arm of chromosome 3H, or the Rrs2 locus on the short arm of chromosome 7H. Other loci containing scald resistance genes have previously been identified using lines derived from wild barley, Hordeum vulgare ssp. spontaneum. A single dominant gene conditioning resistance to scald was identified in a third backcross (BC3F3) line derived from an Israeli accession of wild barley. The resistance gene is linked to three microsatellite markers that map to the long arm of chromosome 7H; the closest of these loci, HVM49, maps 11.5 cM from the resistance gene. As no other scald resistance genes have been mapped to this chromosome arm, it is considered to be a novel scald resistance locus. As the Acp2 isozyme locus is linked to this scald resistance locus, at 17.7 cM, Acp2 is assigned to chromosome 7H. Molecular markers linked to the novel scald resistance gene, designated Rrs15, can be used in breeding for scald resistance.

    Notes: The carbohydrate content of Hordeum spotaneum lines collected from 23 diverse environments in Israel was investigated. The range in composition was sucrose: 0.83—1.15%; trisaccharides): 0.48—1.16% total free and combined fructose: 1.2—4.2%; pentosans: 4.2—9.9%; and fl→3), (1→4)-β-glucans: 4.5—13.2%. The high pentosan and β-glucan content in some lines would cause quality problems if found in malting barleys.

    Notes: Isoelectric focusing (IEF) and immunoblotting were used to detect genetic variants of malt endopeptidase (MEP1), an enzyme related to malting quality in barley and coded by the CepB locus on barley chromosome 3 (= 3H). A variant was found in a Turkish accession of wild barley (Hordeum vulgare ssp. spontaneum). The self progeny of a hybrid between this accession and the barley cultivar ‘Clipper’ were analyzed for recombination between the CepB locus and other isozyme loci. The estimates of recombination linked the CepB locus to an NADH diaphorase locus (Ndh2), which in turn was linked to the seedling esterase complex (Est1, Est2, and Est4) situated near the distal end of the long arm of chromosome 3. The Ndh2 locus was independent of two other NADH dehydrogenase loci (Ndh3, Ndh5) which were mapped on barley chromosome 5 (= 1H) in relation to the three hordein loci (Hor1, Hor2, and Hor3).

    Notes: Fifty-six accessions of Hordeum spontaneum from various Israeli habitats were hybridized with, and their progeny backcrossed three times to H. vulgare cv. ‘Clipper’. Selected chromosome segments marked by one of 24 isozyme loci were transferred to form 84 backcross lines which were nearly isogenic with ‘Clipper’. Each line was made homozygous for a single isozyme-marked segment. These lines were evaluated in preliminary and advanced field trials. The yield, malt extract percentage and grain size of the lines approached or equalled, and sometimes significantly surpassed, that of ‘Clipper’ and more recent commercial cultivars. Three isozyme-marked segments were combined in pairs and the three resultant doubly-marked lines were compared with the reconstituted parental, singly-marked lines and with the ‘Clipper’ recombinant from the same F2-array. The additive effects of single segments on yield or gram size were not significant, whereas half of the six interactions were significant, but in the unfavorable direction. It was concluded that near-isogenic lines can identify segments of wild barley germplasm that are useful for improving yield, but that pairs of these from different habitats may interact unfavourably.

    Notes: The scald susceptible barley cultivar ‘Clipper’ and a third-backcross (BC3) line homozygous for the Rrs14 scald resistance gene that originally came from Hordeum vulgare ssp. spontaneum were grown in replicated field trials. The level of resistance that Rrs14 confers against field populations of the pathogen Rhynchosporium secalis, the causal agent of scald disease, was evaluated. The Rrs14 BC3 line exhibited 80% and 88% less leaf damage than ‘Clipper’ in 1995 and 1996, respectively. Given this effectiveness of Rrs14, research was undertaken to identify a linked marker locus suitable for indirect selection of Rrs14. Based on linkage to a set of previously mapped loci, Rrs14 was positioned to barley chromosome 1H between the seed storage protein (hordein) loci Hor1 and Hor2, approximately 1.8 cM from the latter locus. The Hor2 locus is thus an ideal codominant molecular marker for Rrs14. The tight linkage between Rrs14 and Hor2 and the availability of alternative biochemical and molecular techniques for scoring Hor2 genotypes, permits simple indirect selection of Rrs14 in barley scald resistance breeding programmes.

    Notes: Summary Genetic structure of populations ofHordeum spontaneum, the wild progenitor of barley, was studied electropho-retically in proteins encoded by 28 gene loci in 1179 individuals representing 28 populations covering the entire ecological range in Israel; for comparison, the same test was conducted in 100 random seeds of Composite Cross XXI (generation 17) of cultivated barley,Hordeum vulgare. The results indicate that: a)H. spontaneum in Israel is genetically highly polymorphic, b) clinal, regional and local genetic patterns are significantly correlated with and predictable by climatic and soil variables, suggesting the operation of natural selection, and c) natural populations are on the average more variable than the tested Composite Cross generation. This genetic variation awaits testing and exploitation in breeding programs.

    Notes: Summary Legumin and albumin are the fractions of pea seed proteins preferred to vicilin because of their high sulfur amino acid contents. The joint inheritance of legumin and albumin contents was studied in a cross between to contrasting lines of peas — one with high legumin and low albumin, and the other with low legumin and medium to high albumin. Single seed determinations were made in the parental, F1; F2 and backcross generations using rocket immunoelectrophoresis. In the non-segregating generations (P1, P2 and F1), legumin and albumin contents were negatively correlated (r=−≅0.50). The estimates of correlation coefficients in the segregating generations (F2, BC1 and BC2) were also about −0.5. However, the two estimates based on the round and on the wrinkled seeds separately in the F2 generation were not significantly different from zero. At least four individual round F2 seeds showed the desired recombination of high legumin with high albumin indicating that the unfavorable correlation can be broken. In this cross legumin content showed predominantly additive genetic variation whereas the dominance variance was the largest component for albumin content. A combined “relative sulfur index”, proposed as a convenient measure for selection, showed a narrow sense heritability of 47%. In general these results support the view that sulfur amino acid content of peas can be improved by breeding, but that the required selection regime must take both legumin and albumin content into account.

    Notes: Summary For the measurements of outcrossing rates in plant populations, current electrophoretic procedures permit many loci to be scored per individual progeny. Given that the total experimental effort or cost is limited, the choice exists then between assaying a large number of loci on a restricted number of individuals, or assaying a large number of individuals at a few loci. Using simple models and the criterion of minimising the variance of the estimate, several factors which affect this choice are considered (levels of polymorphism, heterozygosity, linkage disequilibrium, pollen or outcrossing heterogeneity). The general conclusion is that the actual level of outcrossing is a major factor in determining experimental strategy. Maximum efficiency for estimating outcrossing in predominantly inbreeding plants comes from large samples assayed for few polymorphic loci. In contrast, in predominantly outcrossing plants, more loci should be assayed at the expense of sample size for improved statistical efficiency.
