    Notes: Summary The molecular cloning of an α-glucosidase gene isolated from a Candida tsukubaensis (CBS 6389) genomic library in Saccharomyces cerevisiae is reported. The cloned gene is contained within a 6.2 kb Sau3A DNA fragment and directs the synthesis and secretion of an amylolytic enzyme into the extracellular medium of the recombinant host, S. cerevisiae. The cloned enzyme was found to have an unusually broad substrate specificity and is capable of hydrolysing α-1,2, α-1,3, α-1,4 and α1,6 linked, as well as aryl and alkyl, d-glucosides. On the basis of its substrate specificity profile, the cloned enzyme was classified as an α-glucosidase (E.C. 3.2.1.20). It has a pH optimum in the range 4.2–4.6, a temperature optimum of 58°C and is readily inactivated at pasteurization temperature (60°C). Southern blot analysis failed to reveal any homology between the cloned gene and genomic DNA isolated from other well characterized amylolytic yeasts. A rapid plate-assay, based on the utilization of a chromogenic substrate X-α-d-glucoside to detect the expression of the cloned α-glucosidase in S. cerevisiae transformants, was developed.

    Notes: Abstract Until recently, the micro mechanical origins of soil behaviour have remained illusive, but it is now known that that the constitutive behaviour of a soil is largely determined by its particle size distribution. This paper examines the specific boundary problem associated with the penetration of a model pile into two different gradings of dry calcareous sand in a geotechnical centrifuge, in order to establish the effect of the inclusion of fine particles on the pile end bearing resistance. The first grading of sand comprised particles smaller than 0.5 mm; the second grading contained particles of nominal size d such that 0.15 mm 〈 d 〈 0.5 mm. Each test was performed on each of two samples of each grading. Tip resistance was observed to rise to a peak at shallow depths, and then fall; a micro mechanical explanation is presented for this instability. Following the centrifuge tests, particles were retrieved from the centres of the soil samples, where the pile had previously been driven, for subsequent particle size analysis. It was found that insignificant crushing had occurred in the sand retrieved from depths less than the depth of peak resistance, but that significant crushing had occurred in the sand retrieved from greater depths. The peak in tip resistance was a factor of two larger for the well-graded sand, but the ultimate tip resistance at greater depths was found to be approximately independent of the initial particle size distribution for all four tests. A micro mechanical explanation is also proposed for this observation.

    Notes: Abstract This paper presents a simple micro mechanical model for the behaviour of overconsolidated soils. The simplest case of isotropic unloading and reloading is considered for a soil which has been subjected to isotropic plastic (clastic) or “normal” compression. Such a material has a wide disparity in particle sizes, such that small particles are kinematically unstable, and large particles are stable. A simple unit cell is defined with one of the largest grains at the centre, surrounded by a fractal matrix of finer particles. On unloading, the unit cell exhibits elastic behaviour followed by “kinematic yielding”, which is the onset of sliding of grains. On reloading, kinematic yielding may not occur, but the inhomogeneous stresses in the cell may induce “clastic yielding” before re-joining the normal compression line. The unit cell is shown to reproduce micro mechanical features of a real aggregate having particle size disparity, and leads to realistic hysteresis loops which permit possible ratchetting on cyclic loading. A simple dilatancy rule is shown to offer micro mechanical insight into the form of the unloading curves evident in much of the soil mechanics literature.

    Notes: In this paper, a fourth-order displacement (cubic strain) Serendipity quadrilateral element is presented. The shape functions and integration scheme are introduced, followed by a series of convergence tests, indicating that the element does not have any spurious zero-energy modes, and passes the patch test and single-element tests. Modifications are then made to enable the element's use in effective stress analysis problems. Examples are finally solved using the element and a comparison is made between some computed and closed-form solutions. It is also shown that the cubic strain quadrilateral may be used in conjunction with or as a substitute for the cubic strain triangle when predicting collapse loads for undrained plane or axisymmetric problems in the fully plastic range.

    Notes: This paper describes the computer algorithms used in a numerical simulation of the compression of an aggregate of crushable grains. It has been used in a model for the evolution of a granular medium under one-dimensional compression, in which the probability of fracture for individual particles is a function of applied stress, particle-size and co-ordination number. The information relating to the particles is represented in a compact way on the computer which allows the number of particles produced to become sufficiently large for satisfactory comparisons to be made with experimental data and which allows information, such as the positions and sizes of the particles, to be easily extracted. An algorithm based on the representation is used to locate neighbouring particles in a way which does not deteriorate unacceptably in terms of speed as the number of particles increases. © 1997 by John Wiley & Sons, Ltd.
