Zusammenfassung Eine Apparatur für die präparative HPLC wird beschrieben, die Arbeiten mit Säulen von 20 mm und 50 mm Innendurchmesser und 50 cm Länge zuläßt. Insbesondere werden die Probleme der Zu- und Ableitungen der Säule, des Splitsystems und des Detektors diskutiert. Die Säulen wurden mit Kieselgel und Umkehr-Phase C18 (mittlere Teilchengröße etwa zwischen 10 μ und 40 μ) gepackt. An Hand der Meßergebnisse wird gezeigt, daß die Permeabilität niedriger, die Effizienz vergleichbar ist mit analytischen Säulen. Die Anwendbarkeit der „infinite diameter“-Theorie für Säulen im Bereich der präparativen HPLC wird diskutiert. Die Belastbarkeit der präparativen Säulen ist ebenso groß wie die der analytischen Säulen, d.h. 2−·10−4 g/g stationäre Phase bei Kieselgel und 2·10−3 g/g stationäre Phase bei Umkehr-Phase C18. In der Praxis können bei 10–100facher Überladung pro Probekomponente und Arbeitsgang mehrere Gramm Substanz aufgegeben und geschnitten werden.

Summary An apparatus for preparative HPLC is described which permits operation with 20–50 mm i.d. columns, up to 50 cm in length. Particular attention is given to the problems involved with the connections to and from the column, the splitter system, and the detector. The columns were packed with silica gel and reversed phase C18 (mean particle size between about 10 μ and 40 μ). The results indicate that the permeability is lower than, and the efficiency is similar to, that of analytical columns. The applicability of the “infinite diameter” theory to preparative HPLC columns is discussed. The load capacity of preparative columns was found to be as large as that of analytical columns, i.e., (2–5)×10−4 g sample/g stationary phase for silica and 2×10−3 g/g for reversed phase C18. In practice, with 10–100 fold overloading, several grams of sample per component may be separated in one run.
