    Notes: Abstract Hunter disease is an X-linked mucopolysaccharidosis caused by deficiency of the lysosomal enzyme iduronate-2-sulfatase (IDS). Using the IDS cDNA and DNA probes corresponding to loci flanking the IDS locus, we performed molecular genetic studies in two patients with Hunter syndrome. An interstitial deletion spanning the middle part of the IDS gene was found in the first patient. The second patient carries a gross gene rearrangement that can be detected after HindIII or EcoRI digestion of genomic DNA, and is similar to that found recently in seven unrelated Hunter patients. Our data suggest that the structural aberration observed is a partial intragenic inversion. As the same altered hybridization pattern is also revealed by the recently described anonymous DNA probe II 10, which recognizes a frequent TaqI restriction fragment length polymorphism at the DXS466 locus, we conclude that DXS466 maps within the IDS gene, probably in an intron.

    Notes: Abstract Gaucher disease is the most common lysosomal storage disease with a high prevalence in the Ashkenazi Jewish population but it is also present in other populations. The presence of eight mutations (1226G, 1448C, IVS2+1, 84GG, 1504T, 1604T, 1342C and 1297T) and the complete deletion of the β-glucocerebrosidase gene was investigated in 25 unrelated non-Jewish patients with Gaucher’s disease in Germany. In the Jewish population, three of these mutations account for more than 90% of all mutated alleles. In addition, relatives of two patients were included in our study. Restriction fragment length polymorphism analysis and sequencing of PCR products obtained from DNA of peripheral blood leukocytes was performed for mutation analysis. Gene deletion was detected by comparison of radioactively labelled PCR fragments of both the functional β-glucocerebrosidase gene and the pseudogene. Among the unrelated patients, 50 alleles were investigated and the mutations identified in 35 alleles (70%), whereas 15 alleles (30%) remained unidentified. The most prevalent mutation in our group of patients was the 1226G (370Asn→Ser) mutation, accounting for 18 alleles (36%), followed by the 1448C (444Leu→Pro) mutation, that was found in 12 alleles (24%). A complete gene deletion was present in two alleles (4%). The IVS1+2 (splicing mutation), the 1504T (463Arg→Cys) as well as the 1342C (409Asp→His) mutations were each present in one allele (2%). None of the alleles carried the 84GG (frameshift), 1604A (496Arg→His) or the 1297T (394Val→Leu) mutation. This distribution is different from the Ashkenazi Jewish population but is similar to other Caucasian groups like the Spanish and Portuguese populations. Our results confirm the variability of mutation patterns in Gaucher patients of different ethnic origin. All patients were divided into nine groups according to their genotype and their clinical status was related to the individual genotype. Genotype/phenotype characteristics of the 1226G, 1448C, and 1342C mutations of previous studies were confirmed by our results.

    Notes: The integrase (Int) proteins encoded by bacteriophages HK022 and λ catalyse similar site-specific integration and excision reactions between specific DNA regions known as attachment (att) sites. However, the Int proteins of HK022 and λ are unable to catalyse recombination between non-cognate att sites. The att sites of both phages contain weak binding sites for Int, known as ‘core-type’ sites. Negatively acting nucleotide determinants associated with specific core sites (λ B′, HK022 B′, HK022 C) are responsible for the barrier to non-cognate recombination. In this study, we used challenge phages to demonstrate that the λ and HK022 Ints cannot bind to core sites containing non-cognate specificity determinants in vivo. We isolated mutants of the HK022 Int, which bind the λ B′ core site. Two mutants, D99N and D99A, have changed a residue in the core-binding (CB) domain, which may be directly contacting the core site DNA. We suggest that binding to the λ B′ site was accomplished by removing the negatively charged aspartate residue, which normally participates in a conflicting interaction with the G4 nucleotide of the λ B′ site. We showed that, although our mutants retain the ability to recombine their cognate att sites, they are unable to recombine λatt sites.

    Notes: Summary To examine the importance of covariance between stages in traits related to foraging, we quantified the relationships between reproductive success and sizerelated variability in weight gain in juvenile and adult instars of the crab spider Misumenoides formosipes (Araneae: Thomisidae). Prereproductive weight and fecundity are both highly correlated with carapace width, a linear measure of size which does not change within an instar. In field populations, adult females with larger carapaces gain more weight and are more likely to reproduce than females with smaller carapaces. The growth rate of spiders fed ad libitum in the laboratory is unrelated to size, suggesting that size-related differences in the field are due to variation in prey-capture success. Adult females with a carapace width less than 3.4 mm comprised 22% of the population, but were never found to reproduce. Of the individuals that did reproduce, a 17% increase in carapace width resulted in a 100% increase in fecundity. Juvenile stages must be examined to understand adult foraging and reproductive success, because the net weight gained by juvenile instars determines adult size. The final weight gained by spiders in the antepenultimate and penultimate instars explained nearly all the variation in carapace width in the penultimate and adult instars, respectively. We found that constraints on foraging in late juvenile stages are different from the adult stage. Penultimate foraging behavior differs from that of adults, because of constraints on foraging in the period preceding ecdysis. Additionally, in both late juvenile instars, carapace width had little or no effect on the final weight gained within the instar suggesting that factors that affect foraging are different between the juvenile and adult stages. These analyses stress the fact that to fully understand the effects of foraging on reproductive success, we must examine stage-specific constraints throughout an organism's life history.

    Notes: Abstract. A new method for the evaluation of one- and two-centre magnetic and electric multipole integrals for Slater-type functions is presented. The method is strictly analytical in that no approximations of any kind are involved. Two simple functions, ℐ1 aug and ℐ2 aug, are introduced, which employ only functions that are well known in electronic structure theory. With the use of augmentation exponents these functions apply to multipole integrals as well as other one-electron integrals, such as nuclear attraction integrals. The proposed method includes the analytic determination of derivatives of the integrals with respect to atomic displacements. Some illustrative test calculations are presented and compared to results from the literature.

    Notes: Abstract The effect of explant size, configuration, and duration of benzyladenine exposure on shoot organogenesis in Petunia x hybrida ‘Ultra Salmon’ leaf discs was determined. Leaf discs of 13, 10, 8, 5, and 3 mm diameter, and 10 mm with a 5 o4 3 mm hole and 13 mm with an 8 or 5 mm hole were tested. The smallest size and configuration for maximum shoot production and consistency of regeneration was a 10 mm whole disc. This was followed by the 8, 10 with a 3 mm hole, 5, 10 with a 5 mm hole and 3 mm disc respectively. Of all configurations tested, the 13 mm disc and the 13 mm disc with a 5 mm hole produced the most shoots. Exposure of leaf discs to benzyladenine-containing medium for 4 days or less resulted in no shoots being formed. Exposure for 10 days or more induced shoot regeneration on all discs tested. A delay of more than 2 days after excision, before plating on benzyladenine-containing medium, decreased shoot regeneration significantly. The explant configurations tested showed that: the area to cut edge ratio was a good predictor of shoot number for either of the two configurations when considered separately; and there are factors other than area to cut edge ratio involved in relating shoot production to explant configuration and size.

    Notes: Iliac crest biopsies are important in the detection of human skeletal dysplasias. Therefore, culture of these cells may serve as a valuable method for studying proteoglycan metabolism in chondrocytes of individuals with skeletal abnormalities. Morphological and biochemical studies were performed on human iliac crest chondrocytes grown in monolayer and in agarose gels. Two proteoglycan populations of different hydrodynamic size and glycosaminoglycan composition were synthesized by cells grown in monolayer. Chondrocytes cultured in an agarose gel for 2 weeks synthesized proteoglycans identical to those of the native tissue with respect to hydrodynamic size and glycosaminoglycan chain length. However, the ratio of chondroitin-6-sulfate to chondroitin-4-sulfate was higher than in the native tissue. This ratio was not influenced by different sulfate concentrations in the medium. Moreover, treatment with ascorbic acid did not influence proteoglycan synthesis; however, there was a pericellular accumulation of proteoglycans. © 1994 Wiley-Liss, Inc.
