    Description / Table of Contents: Zusammenfassung In histochemischen Untersuchungen an der Mamma von Ratten wird das Verhalten der Enzymaktivitäten in Gravidität, Laktation, Involution, bei experimenteller Milchstauung und unter dem Einfluß von Oxytocin demonstriert. Die Untersuchungen wurden an 38 weiblichen Sprague-Dawley-Ratten durchgeführt. 1. Gruppe: 7 gravide Tiere, Gewebsentnahme am 12., 16. und 28. Tag post conceptionem. 2. Gruppe: 10 laktierende Tiere, Gewebsentnahme am 2., 10. und 20. Tag post partum. 3. Gruppe: 7 Tiere am 4. und 10. Tag nach Absetzung der Jungtiere (Involution). 4. Gruppe: 6 Tiere mit experimenteller Milchstauung am 5. Tag der Laktation. 5. Gruppe: 8 Tiere zur Untersuchung der experimentellen Oxytocinwirkung (0,5; 1,0; 2,0 IE/kg Körpergewicht synthetisches Oxytocin intravenös). Histochemische Darstellung der alkalischen Phosphatase (Metallsalz-, Azofarbstoffmethode), sauren Phosphatase, unspezifischen Esterase, Succinodehydrogenase, Monaminoxydase, Glukose-6-Phosphatase, Adenosintriphosphatase, 5′-Nukleotidase, Glukose6-Phosphat-Dehydrogenase, Laktatdehydrogenase. Der synthetischen Aktivität der laktierenden Mamma entspricht eine starke Erhöhung der Enzymaktivität. Im Vergleich zur Laktation ist diese in Gravidität und Involution wesentlich geringer. Bei akuter Milchstauung tritt rapider Aktivitätsverlust ein. Myoepithelzellen werden durch alkalische Phosphatase (Azofarbstoffmethode) selektiv dargestellt. Der Strukturwandel dieser Zellen in verschiedenen Funktionsphasen wird beschrieben. Elektronenoptisch ist die alkalische Phosphatase in der basalen und interzellulären Zellmembran und im Myoepithel lokalisiert. Auch saure Phosphatase, Glukose-6-Phosphatase sind in der Epithelbasis nachweisbar. Unspezifische Esterase-Aktivität ist in der Peripherie der Läppchen, im angrenzenden Fettgewebe und im Epithel nachweisbar. Adenosintriphosphatase ist nur auf mesenchymale Bestandteile der Mamma beschränkt. Succinodehydrogenase, Monoaminoxydase, Laktat- und Glukose-6-Phosphat-Dehydrogenase bilden granuläre intraepithelial lokalisierte Reaktionsprodukte. - Die histotopochemischen Befunde werden biochemischen Untersuchungen gegenübergestellt.
    Notes: Summary In histoehemical studies of the mammary gland of rats the reaction of the enzyme activity is demonstrated during pregnancy, lactation, involution, experimental galaotostasis and under the influence of oxytocin. 38 female Sprague-Dewley rats were examined. 1st group: 7 pregnant animals, excision of tissue on the 12th, 16th, and 28th day after conception. 2nd group: 10 animals in lactation, excision of tissue on the 2nd, 10th, and 20th day after delivery. 3rd group: 7 animals on the 4th and 10th day after removing the young animals (involution). 4th group: 6 animals with experimental galactostasis on the 5th day of lactation. 5th group: 8 animals for examination of the experimental influence of oxytocin (0.5; 1.0; 2.0 IE of synthetic oxytocin per kg bodyweight i.v.). The histoehemical examination showed alcaline phosphatase (method of metallic salt and azo dye), acid phosphatase, unspecific esterase, succinic dehydrogenase, monoaminoxydase, glucose-6-phosphatase, adenosine triphosphatase, 5′-nucleotidase, glucose-6-phosphate dehydrogenase, and lactic dehydrogenase. A strong increase of the enzyme activity corresponds to the synthetic activity of the mammary gland in lactation. In comparison to lactation enzyme activity is considerably smaller in pregnancy and involution. During acute galactostasis enzyme activity decreases rapidly. Myoepithelial cells are shown by alcaline phosphatase (azo dye method) selectively. The change of the structure of these cells is demonstrated in different functional phases. In electron microscopical examination alcaline phosphatase is situated in the basement membrane and intercellular membrane and in myoepithelial cells. Also acid phosphatase and glucose-6-phosphatase can be shown in the basis of epithelial cells. Unspecific esterase-aktivity can be shown in the periphery of the lobules, in the adjacent fat tissue, and in the epithelial cells. Adenosintriphosphatase is limited to mesenchymal parts of the mammary gland. Succinic dehydrogenase, monaminoxydase, lactic dehydrogenase, and glucose-6-phosphatedehydrogenase form granulated reaction products in the epithelial cells. The results of histotopochemical examination are compared to those of biochemical examinations.

Die Untersuchungen wurden an 40 weiblichen juvenilen und kastrierten Albinoratten durchgeführt, von denen die 1. Gruppe Follikelhormon (Progynon, 5 γ/d), die 2. Gruppe Corpus luteum-Hormon (Proluton, 1 mg/d) erhielten. Neben Kontrolltieren gleichen Alters wurden die Brustdrüsen unbehandelter infantiler Tiere untersucht. Am 3., 5., 10. und 20. Tag nach Injektionsbeginn wurden die Tiere getötet und die Brustdrüsen histologisch sowie enzymhistochemisch durch den Nachweis von alkalischer Phosphatase (Azofarbstoff und Schwermetallsimultanmethode), Glukose-6-Phosphatase, Adenosintriphosphatase, 5′-Nukleotidase, Glukose-6-Phosphat-Dehydrogenase, Succinodehydrogenase und Laktatdehydrogenase aufgearbeitet. Es wurde festgestellt, daß unter dem Einfluß der Behandlung mit Geschlechtshormonen ein Anstieg der Enzymaktivität einsetzt, wobei Östrogen eine dominierende Bedeutung hat. Diese zeigt sich vor allem in den ersten 5 Tagen, während danach eine gewisse Gleichförmigkeit in der Intensität dieser Reaktion eintritt. Die Ergebnisse lassen sich dem Wandel der Zellstruktur unter gleichartigen hormonalen Einflüssen zuordnen und sind damit Ausdruck hormonal induzierter Stoffwechselleistungen.
    Notes: Summary The influence of estrogene und progesterone on the activity of enzymes is to be demonstrated by histochemical examinations of the mammary gland of rats. The examinations were made with 40 female juvenile and castrated albino rats of which the 1st group was given follicle-hormone (progynon, 5 γ/d), the 2nd group corpus-luteum-hormone (proluton, 1 mg/d). Besides control animals of the same age the mammary glands of untreated infantile animals were examined. On the 3rd, 5th, 10th, and 20th day after the beginning of the injections the animals were killed and the mammary glands were treated histologically as well as enzymehistochemically by examining their reaction to alcaline phosphatase, glucose-6-phosphatase, adenosine triphosphatase, 5′-nucleotidase, glucose-6-phosphatase-dehydrogenase, succinic dehydrogenase, and lactic dehydrogenase. We found that under the influence of sex hormones the activity of enzymes increased and that estrogene had a dominating importance. This importance could be demonstrated during the first five days, while later these reactions showed a certain balance in their intensity. The results correspond to the change of the cellstructure under similar hormonal influences and thus are results of hormonally induced changes of metabolism.

    Description / Table of Contents: Zusammenfassung Im Vergleich zu lichtmikroskopischen Untersuchungen an der Mamma virilis wird anhand von 2 operativ entfernten Brustdrüsen eines 57 und 63 Jahre alten Mannes die elektronenmikroskopisch erfaßbare Zytomorphologie beschrieben. Die Befunde werden den physiologischen Wachstumsimpulsen dieses Organs gegenübergestellt und Fragen der Zelldifferenzierung, der Desquamation und apokrinen Sekretion beantwortet. Elektronenmikroskopisch werden am Drüsenepithel Basalzellen, größere Zellen der oberflächlichen Zellreihen und Myoepithelzellen unterschieden. Diese Zellen entsprechen den Gangepithelien der weiblichen Brustdrüse und besitzen intracytoplasmatische Filamente. Diese stellen ein häufiges Differenzierungsprodukt des Zytoplasmas dar. Mechanismen einer Sekretion waren nicht nachweisbar. In die Drüsenlichtung werden pseudopodienartig vorgewölbte Zytoplasmateile abgeschnürt (Extrasionsvorgang). — Superfiziale Zellen werden desquamiert, wobei die Zytolyse in den marginalen Zytoplasmaschichten erfolgt. Kern und Teile des Zytoplasmas gelangen in die Drüsenlichtung. — Die Befunde zeigen die von Lebensalter und Proliferationsreiz abhängigen Vorgänge eines permanenten Zellersatzes in der männlichen Brustdrüse an.
    Notes: Summary The ultrastructure of two mammary glands obtained operatively from a 57-year old and a 63-year old man was compared to the structure observed in the light microscope, and related to stimuli controlling growth of the gland, cellular differentiation and desquamation, and apocrine secretion. The glandular epithelium, which is analogous to that of the female mammary gland, is differentiated into basic cells, large superficial cells, and myoepithelial cells. The cells have intracytoplasmic filaments, that may be a sign of differentiation. Mechanisms for secretion were not observed, although pseudopodia-like parts of the cytoplasm are extruded into the glandular lumen. Superficial cells are desquamated, followed by cytolysis of their margins. These findings illustrate the replacement of cells due to age and altered stimuli.

    Description: [1]  We performed three field campaigns in 2004, 2007, and 2010 at the southern margin of the Jakobshavn Isbræ, West Greenland, in order to infer flow velocities and their changes from photogrammetric time-lapse imagery with a temporal resolution of 20 min and a spatial spacing of about 30 m on the glacier surface. Area-wide analysis of more than 3000 three-dimensional trajectories at individual glacier positions allow for both the mapping of the grounding line and the detailed observation of flow variations during major calving events. From 2004 to 2010, the grounding line of Jakobshavn Isbræ retreated 3.5 ± 0.2 km. Considering previously published results, the grounding line retreat amounts to 6 km since 1985. The glacier has an ephemeral floating tongue that can establish during the readvance of the glacier front and break apart after large calving events. Observations of a major calving event show that an acceleration of flow velocities coincides with the onset of the break up during which flow velocities of up to 70 m/d can be reached. Moreover, large vertical displacements of the glacier front in the order of 15 m and lowering of 8 m at positions 500 m beyond the calving front were observed 2 days before the calving event. After the break up, the glacier slowly adjusts to the new boundary conditions within the next 4–5 days. Flow velocity variations caused by calving were detected up to 1 km upstream only which indicates that individual calving events have no immediate effect on the large-scale glacier dynamics.

    Description: Among all of the existing electric power generation facilities, corrosion is considered to be the most severe on geothermal power plants. The focus of this research was set to evaluate the suitability of carbon steel API Q125 as a construction material on the geothermal sites Sibayak and Lahendong, Indonesia. The investigations were performed in the artificial geothermal brines under stagnant conditions by means of short-term electrochemical methods (Tafel extrapolation) and long-term exposure tests (up to 6 months). Furthermore, the influence of different parameters on the steels performance (temperature 70 °C, 100 °C and 175 °C,salinity 20 mg/L and 1,500 mg/L chlorides, pH 2 and 4) was assessed as well to ensure a “safer operating window” of a power plant.
