    Notes: Abstract Taxonomic diversity, physiological tolerance ranges, and nutrient utilization capabilities were determined for bacterial communities in Gulf of Alaska surface waters and sediments. Taxonomic diversity was assessed using Shannon Weaver (H′) and equitability (J′) indices. Physiological tolerance and nutritional versatility indices were developed to further assess the state of “informational heterogeneity” within the bacterial communities. The Gulf of Alaska bacterial communities were characteristically diverse; the bacterial populations in these marine ecosystems generally were eurytolerant and nutritionally versatile. The maintenance of a high degree of informational heterogeneity was found to be characteristic of these bacterial communities. It appears to be of adaptive advantage to maintain diverse populations with physiological tolerances whose ranges exceed those experienced within the natural habitat, and for the bacterial communities to possess a high degree of nutritional versatility within these marine ecosystems.

    Notes: Abstract Introducing the genetically engineered microorganism Pseudomonas cepacia AC1100 into soil microcosms resulted in elevated taxonomic diversity determined by phenotypic analyses of culturable isolates and genetic diversity determined by analysis of the heterogeneity of total microbial community DNA reannealing kinetics. The greatest impact occurred when P. cepacia AC1100 was introduced along with the herbicide 2,4,5-T, which P. cepacia AC1100 can degrade. The data suggests that both changes in the balance of populations and genetic recombination contributed to the increased diversity.

    Notes: Abstract The fears and predictions of attacks with biological weapons, which were increasing at the close of the twentieth century, were transformed into reality not long after September 11, 2001, when several anthrax-laden letters were sent through the U.S. postal system. The attack challenged our medical preparedness and scientific understanding of the epidemiology of biothreat agents. It is fortunate that this was not a massive aerosol release that could have exposed hundreds of thousands. Rapid diagnoses and medical treatments limited casualties and increased survival rates, but tragically some individuals died of inhalational anthrax. Even as physicians tested new treatment regimes and scientists employed new ways of detecting anthrax and decontaminating the mail, new predictions were made for potentially even more devastating attacks with anthrax, smallpox, plague, tularemia, botulism, or hemorrhagic fever viruses. Fear gripped the nation. Law enforcement sought to find the villain(s) who sent the anthrax letters and to deter future bioterrorist attacks. The biomedical community began to seek new ways of protecting against such future threats of bioterrorism.

    Notes: Abstract Plans to introduce genetically engineered microorganisms into the environment has led to concerns over safety and has raised questions about how to detect and to contain such microorganisms. Specific gene sequences, such as lacZ, have been inserted into genetically engineered microorganisms to permit their phenotypic detection. Molecular methods have been developed based upon recovery of DNA from environmental samples and gene probe hybridization to specific diagnostic gene sequences for the specific detection of genetically engineered microorganisms. DNA amplification using the polymerase chain reaction has been applied to enhance detection sensitivity so that single gene targets can be detected. Detection of messenger RNA has permitted the monitoring of gene expression in the environment. The use of reporter genes, such as the lux gene for bioluminescence, likewise has permitted the observation of gene expression. Conditional lethal constructs have been developed as models for containment of genetically engineered microorganisms. Suicide vectors, based upon the hok gene have been developed as model containment systems.

    Notes: Biodegradation and mineralization of petroleum, added at 1% (v/v) to freshly collected sea water, were measured using gas-liquid chromatographic, residual weight, and CO2-evolution techniques. Only 3% of the added petroleum was biodegraded and 1% was mineralized in unamended sea water after 18 days of incubation. Added individually, nitrate (10-2 M) or phosphate (3.5 × 10-4 M) supplements caused little improvement, but when added in combination, they increased petroleum biodegradation and mineralization to 70% and 42%, respectively. Attempts to clean up oil spills with the aid of microorganisms should take into consideration the nutritional deficiencies of sea water.
