    Notes: Abstract. Polo-like kinases (Plks) have been implicated in various aspects of M-phase progression in organisms ranging from yeast to man. In vertebrates, Plks participate in centrosome maturation and spindle assembly, as well as the activation of the Cdk1/cyclin B complex. Moreover, Plks are required for the destruction of mitotic cyclins, indicating that they play an important role in the regulation of the ubiquitin-dependent proteolytic degradation machinery that controls exit from M-phase. Here, we have fused Green Fluorescent Protein (GFP) to the N-terminus of human Plk1, and expressed this chimeric construct in human cells. We found that GFP-Plk1 associates with centrosomes, the equatorial spindle midzone and the postmitotic bridge of dividing cells, confirming and extending previous results obtained with conventional immunofluorescence microscopy. In addition, however, we observed fluorescence emanating from the midbody between dividing cells, and from discrete dots associated with mitotic chromosomes. This latter staining pattern being reminiscent of centromeres, we performed double-labeling experiments with antibodies against the centromeric marker CENP-B, and reexamined the subcellular localization of endogenous Plk1 using different fixation procedures. Our data clearly show that both GFP-tagged Plk1 and endogenous Plk1 associate with the kinetochore/centromere region of human mitotic chromosomes. This novel localization of Plk1 suggests that substrates and/or regulators of Plks may be found among kinetochore-associated proteins with important functions in chromosome segregation and/or spindle checkpoint mechanisms.

    Notes: A new technique for observing domain walls and lines in materials with perpendicular anisotropy is presented. It employs laser light in a laser scan microscope and uses anisotropic diffraction by the wall/line magnetic structure. The contrast obtained can be understood simply and a numerical computation shows good agreement with experimental results. Wall configurations with numerous lines may be obtained when nucleating parallel stripes upon reduction to zero of a saturating in-plane field.

    Notes: Direct observations and Fourier transforms of amorphous networks of magnetic bubbles in ion-implanted garnets and in pure films are shown with evidence for a short coherence size. This is interpretated on the basis of a model of interaction of bubbles via dipolar interactions where only short-range effects are considered because long-range effects are totally screened by the numerous domains included in this range. Moreover these dipolar forces which are proportional to both the magnetizations stabilize the occurrence of different radii of bubbles. These two considerations enable us to understand these amorphous networks as due to short-range interactions and random fields and thence to predict a coherence size of typically 50–300 bubbles in a hexagonal arrangement. Small rotating fields in the phase of the film do not strongly perturb the size distribution of bubbles, i.e., the transverse magnetic field distribution, but induce many local displacements of bubbles. These displacements can be coherent and thus collective if the excitation frequency is associated with a soft mode of this amorphous structure. Thus local cascades of hopping motion as well as collective modes are observed as a function of the excitation frequency. The practical soft modes are localized on the grain boundaries and lead either to a decrease of the coherence size, i.e., melting or to an increase, i.e., recrystallization. Theoretical investigations of the soft mode frequency and localization are given. Experimentally direct and Fourier transform observations as a function of frequency are recorded in a video tape.

    Notes: Abstract Hybrid glasses based on 3(Trimethoxysilyl) propyl methacrylate (TMSM) and methyl methacrylate (MMA) were successfully prepared by sol-gel process. The molecular mobility of hybrid materials was studied by mean of high resolution dynamic mechanical spectrometry technique. The results of hybrid materials were compared with those of polymethylmethacrylate (PMMA) prepared in the same conditions and interpreted in term of local or large-scale molecular motion.

    Notes: A new approach called the "variational theory of complex rays" (VTCR) is developed for calculating the vibrations of weakly damped elastic structures in the medium-frequency range. Here, the emphasis is put on the most fundamental aspects. The effective quantities (elastic energy, vibration intensity, etc.) are evaluated after solving a small system of equations which does not derive from a finite element discretization of the structure. Numerical examples related to plates show the appeal and the possibilities of the VTCR.

    Description: We present measurements of fabrics and microstructures made along the Talos Dome ice core, a core drilled in East Antarctica in the framework of the TALDICE project. Fabric and average grain size data are analyzed regarding changes in climatic conditions. In particular, the fabric strength increases sharply going downward from Holocene to Wisconsin ice. Following (Durand et al., 2007), this change is associated with a positive feedback between variations in ice viscosity, due to variations in dust content, and the impact of a shear stress component, increasing with depth. A ViscoPlastic Self-Consistent modeling approach is used to simulate the fabric evolution for a “perfect dome” configuration. The discrepancies between the measured and the simulated fabrics highlight the depth ranges where shear strongly affects the fabric strengthening. Finally, the grain size and fabric analyses show the occurrence of dynamic recrystallization mechanisms (continuous and discontinuous) along the core.

    Description: The paper presents the first retrievals of clean snow properties using spaceborne hyperspectral observations via the Environmental Mapping and Analysis Program (EnMAP). The location close to the Concordia station at the Dome C Plateau (Antarctica) was selected. At this location, the atmospheric effects (except molecular light scattering and absorption) are weak, and the simplified atmospheric correction scheme could be applied. The ice grain size, snow specific surface area, and snow spectral and broadband albedos were retrieved using single-view EnMAP measurements. In addition, we propose a technique to retrieve trace gas concentrations (e.g., water vapor and ozone) from EnMAP observations over the snow surfaces. A close correspondence of satellite and ground-measured parameters was found.
