    Notes: Summary Responses of single olfactory cells on the antennal club ofIps pini have been recorded electrophysiologically. The majority of cells were strongly activated by either/or of the two behavior-modifying chemicals, the aggregation pheromone ipsdienol and the aggregation inhibitor ipsenol. Simultaneous stimulation with these terpence alcohols showed that ipsenol had no inhibitory effect on the receptor responses to ipsdienol. It appears that the behavioral inhibition by ipsenol on the attraction of ipsdienol results from central integration of the information from separate receptor cells rather than blockage at the peripheral receptors.

    Notes: Abstract Electrophysiological recordings inIps pini were made from single olfactory cells stimulated by six concentrations of eleven compounds—either pheromones or host constituents. The receptor cells were grouped according to their differential responses to these biologically significant compounds. Cells specialized to either ipsdienol or ipsenol (pheromones for severalIps species) appeared to be relatively uniform in their sensitivity to all compounds tested. Another group of cells specialized tocis- andtrans-verbenol were more variable in their sensitivities to the substances tested. The activation of separate groups of cells by the different pheromones and host compounds shows (by exclusion) that behaviorally expressed syner-gistic as well as inhibitory action mainly is due to an interaction in the central nervous system rather than an interaction of the compounds on the receptors. All “ipsdienol cells” responded to both enantiomers of ipsdienol. Although some differences between responses to the enantiomers by individual cells were observed, these did not form a consistent pattern, and no net differences could be found between the eastern and western populations ofI. pint.

    Notes: Abstract Electroantennogram (EAG) and single-cell recording techniques have been used to demonstrate the presence of separate receptors for (−)-threo- and (−)-erythro-4-methyl-3-heptanol on the antenna ofS. scolytus. The majority of single-cell recordings showed spikes of two different amplitudes. The cell giving spikes of larger amplitude responded to the (−)-threo stereoisomer while the cell with the small-amplitude spikes responded to (−)-erythro-4-methyl-3-heptanol. It is suggested that in most recordings the two cells are associated with a single sensillum basiconicum.

    Notes: Abstract The stimulation effect of the two enantiomers of the male-produced aggregation pheromone, ipsdienol, was tested by electrophysiological recordings from single olfactory cells in females of two species,Ips pini andI. paraconfusus. The results demonstrated two types of receptor cells, each specialized to one of the optical configurations. This suggests that separate acceptors (membrane receptors) for (+)- and (−)-ipsdienol are produced by the beetles' receptor cells. The dose-response curves obtained for the 92% “pure” enantiomers and racemic mixtures indicated no synergistic or inhibitory interaction of the enantiomers on the receptor cells. The results could be explained by activation of one acceptor type in each cell group. I. paraconfusus apparently had the majority of its ipsdienol cells keyed to the (+)-enantiomer. Conversely, the westernI.pini had more cells tuned to (−)- than to (+)-ipsdienol. This difference is consistent with behavioral responses where these species are sympatric in California. The (+)- and (−)-ipsdienol are aggregation pheromone components ofI. paraconfusus anaI. pint, respectively, and the opposite enantiomers act as aggregation inhibiting allomones. More (−)- than (+)-ipsdienol cells were also obtained in the easternI. pint, even though this population produces more (+)- than (−)-ipsdienol (63∶35) and requires both enantiomers for aggregation behavior. However, the difference in the numbers of (+)- and (−)-ipsdienol cells recorded from the eastern population was insufficient for an acceptable level of statistical confidence.

    Notes: Abstract Laboratory bioassays and field tests demonstrated that a Swiss population ofS. multistriatus responded much more strongly to α- than to δ-miltistriatin in combination with 4-methyl-3-heptanol and α-cubebene. High concentrations of brevicomin appeared to replace α-multistriatin in evoking a response byScolytus species, but this effect can be explained by the fact that the brevicomin was contiminated with small amounts of α-multistriatin. Frontalin, another bicyclic ketal, showed no biological activity. Field tests indicated thatS. pygmaeus aggregates to the same attractant mixture asS. multistriatus. S. scolytus also responded preferentially to this mixture, but the relative amounts of α-multistriatin to 4-methyl-3-heptanol do not appear to be as important as forS. multistriatus.
