    Notes: Abstract The partitioning of six phenothiazines was determined between phosphate buffer (pH 6.0) and the lipid phases of cyclohexane, n-octanol and dimyristoyl phosphatidylcholine (DMPC). For DMPC liposomes studies were carried out both below and above the phase transition temperature (Tc) of the liposomes. The partitioning of chlorpromazine hydrochloride between n-octanol and phosphate buffer was both pH and concentration-dependent. A linear relationship between the absolute temperature (T−1) and the logarithm of the equilibrium partition coefficient (ln K) was derived. The temperature dependence of the partition coefficient (K) over the temperature range 20–40° C in cyclohexane and n-octanol, and 5–40° C in DMPC liposomes, permitted the calculation of free-energy (G), enthalpy (H) and the entropy (S) of partitioning. Both the entropy and the enthalpy of partitioning of phenothiazines were positive in the three systems studied. In general, the partitioning of phenothiazines in cyclohexane, n-octanol and DMPC liposomes (both above and below the phase transition temperature (Tc)) is entropically controlled. Correlation was not however found between the free-energy of oil-water partitioning and liposome-water partitioning which may be attributed to the formation of surface associated phenothiazine in high concentrations at the liposome water interface. The concentration dependent partitioning of chlorpromazine in DMPC liposomes may be attributed to the adsorbed fraction of drug.

    Notes: Abstract Sensitive analytical methods indicate the presence of hundreds of chemical contaminants in our environment. However, concentration of these pollutants is usually at the low parts per million or parts per billion level. At such low levels, toxicants induce long-term (chronic) rather than short-term (acute) toxicities. Experiments are designed to evaluate chronic toxicity using early bioindicators. Recently, fish have been used as experimental animals because some species show early (weeks as opposed to years in other research animals), sensitive responses. Thus, medaka (Oryzias latipes), following exposure to diethylnitrosamine (DEN), exhibited liver tumors in several weeks; more interestingly its DNA was modified (to a ‘DNA-adduct’) after only 24–48 hours exposure. Such adducts show promise as early bioindicators because they are formed within hours of exposure. The formation of DNA-adducts were monitored in medaka and rainbow trout (Salmo gairdneri) exposed to DEN by aqueous or intra-peritoneal routes. Using HPLC-Fluorimetry, O 6-ethylguanine (O 6-EtGua) was detected and monitored in acid thermal hydrolysates of DNA isolates. (Detection limit for O 6-EtGua was as low as 3 ng, 1.7×10-11 moles). Fourier transform cyclotron resonance mass spectrometry with IR laser desorption/ionization was used advantageously to establish the structure of nucleotides, bases and nucleosides directly, without further cumbersome derivatizations. Other aspects of DNA-adducts are discussed.

    Notes: Abstract Mechanical and electrical behaviour of the “Magnex DC” conductive polymer composite is reported. X-ray diffraction patterns and scanning electron microscopy (SEM) show that the composite has a semicrystalline, isotropic, and defected structure. It was found that both the elastic modulus and yield stress increase with decreasing temperature; the yield stress increases nonlinearly with increasing strain rate due to the voids in the composite. In addition, the activation energy and the activation volume of a single rate-activated process observed at relatively high strain rates are determined. The dependence of the impedance behaviour on frequency is investigated. It was observed that the a.c. conductivity is almost independent of frequency below 10 kHz and increases with frequency above this range. The values obtained for the conductivity indicate that the composite has a relatively small electrical conduction which is rather less than that for the pure semiconductors.

    Description: The Transporter Classification Database (TCDB; http://www.tcdb.org ) is a freely accessible reference database for transport protein research, which provides structural, functional, mechanistic, evolutionary and disease/medical information about transporters from organisms of all types. TCDB is the only transport protein classification database adopted by the International Union of Biochemistry and Molecular Biology (IUBMB). It consists of more than 10 000 non-redundant transport systems with more than 11 000 reference citations, classified into over 1000 transporter families. Transporters in TCDB can be single or multi-component systems, categorized in a functional/phylogenetic hierarchical system of classes, subclasses, families, subfamilies and transport systems. TCDB also includes updated software designed to analyze the distinctive features of transport proteins, extending its usefulness. Here we present a comprehensive update of the database contents and features and summarize recent discoveries recorded in TCDB.
