    Notes: Summary Previous studies have shown that transformation of Escherichia coli by plasmid DNA modified in vitro by carcinogens leads to RecA-dependant recombination between homologous plasmid and chromosomal DNA sequences. The mechanism of this recombination has now been studied using recombination-deficient mutants, and the influence of induction of the SOS response on the level of recombination investigated. Plasmid pNO1523, containing the str + operon (Sms), has been modified in vitro by either irradiation with UV light, or by reaction with (±) trans-benzo(a)pyrene-7,8-dihydrodiol-9,10-epoxide (BPDE) and used to transform streptomycin-resistant hosts. The formation of Ampr transformants which also carry streptomycin resistance was used as a measure of the level of recombination between plasmid and chromosomal DNA. Transformation of recB and recC mutants produced no change in the level of recombination while in the recF mutant a significant decrease was observed compared to the wild type host. Thermal induction of the SOS response in tif-1 and tif-1 umuC mutants followed by transformation led to a four-fold increase in recombination in both cases. The results suggest that the streptomycin-resistant transformants arise exclusively via a recombinational pathway which is largely dependant on the recF gene product, and that this pathway is influenced by induction of the SOS response. These results are discussed in terms of the mechanism of this recombination.

    Description: Rapid changes in ocean circulation and climate have been observed in marine-sediment and ice cores over the last glacial period and deglaciation, highlighting the non-linear character of the climate system and underlining the possibility of rapid climate shifts in response to anthropogenic greenhouse gas forcing. To date, these rapid changes in climate and ocean circulation are still not fully explained. One obstacle hindering progress in our understanding of the interactions between past ocean circulation and climate changes is the difficulty of accurately dating marine cores. Here, we present a set of 92 marine sediment cores from the Atlantic Ocean for which we have established age-depth models that are consistent with the Greenland GICC05 ice core chronology, and computed the associated dating uncertainties, using a new deposition modeling technique. This is the first set of consistently dated marine sediment cores enabling paleoclimate scientists to evaluate leads/lags between circulation and climate changes over vast regions of the Atlantic Ocean. Moreover, this data set is of direct use in paleoclimate modeling studies.

    Description: Ice cores are powerful archives for reconstructing volcanism as they contain both soluble (i.e. aerosols) and insoluble (i.e. tephra) products of volcanic eruptions and for more recent periods have high-precision annually resolved chronologies. The identification and geochemical analysis of cryptotephra in these cores can provide their volcanic source and latitude of injection, complementing records of sulphur injections from volcanic eruptions developed using continuous flow ice-core analysis. Here, we aim to improve the volcanic record for the Southern Hemisphere using a sampling strategy for cryptotephra identification based on coeval deposition of sulphate and microparticles in ice cores from the interior of East Antarctica covering the Mid-to Late Holocene. In total, 15 cryptotephras and one visible horizon were identified and geochemically characterised. Through comparisons to proximal deposits a range of possible sources were isolated for these horizons including the South Sandwich Islands, South Shetland Islands, Victoria Land (Antarctica) and South America. This new tephra framework contributes to the volcanic history of the region by extending the known geographical range of tephra deposition for previously identified events and providing a potential indication of phases of eruptive activity from key sources. Using the tephra-based source attributions and comparison of the timing of the events to a database of sulphur injections from Holocene volcanic eruptions it is possible to refine injection latitudes for some events, which can lead to improved estimates of their radiative forcing potential. The relatively low magnitude of the volcanic stratospheric sulphur injections related to the events in the tephra framework indicates they would have had a limited impact on Southern Hemisphere climate. Further work is required to improve source attributions for some events and/or to determine the magnitude of sulphur injections for individual events during years when coeval eruptions occurred. One limitation of the framework is the dominance of cryptotephra from regional volcanic sources and a lack of tephra from tropical sources, which hampers the refinement of eruption parameters for these large magnitude and often climate-impacting eruptions. This issue could be explored further through increased sampling of these events and/or development of additional analytical techniques for the identification and robust geochemical analysis of glass tephra shards less than 5 μm in diameter. Such investigations could be coupled with model experiments to determine the likelihood that past tropical eruptions deposited glass tephra shards over Antarctica and the potential size range and geographical spread of deposition.

    Description: Recently, indirect evidence was obtained for inhibition of soil net N mineralization by sterols in soil organic matter, which could have been caused by their antioxidant or antimicrobial properties. The objective of this study was to test the effect of potential inhibitors ( i.e., individual compounds with known antioxidant and/or antimicrobial properties) on soil microbial mineralization processes during incubation for 7 and 14 d. A sandy agricultural soil was amended with four substances: two phenolic acids differing in their antioxidant capacity (AOC) (acetovanillone with no AOC, ferulic acid with large AOC), Trolox, an analogue of vitamin E (large AOC), and β-sitosterol (no AOC, but potential antimicrobial properties). The two compounds with large AOC (ferulic acid and Trolox) showed no significant inhibition of C and net N mineralization; and the Trolox amendment actually caused a significant increase in C and net N mineralization after 7 d of incubation. Acetovanillone with no measurable AOC caused a significant increase in C mineralization (109% of substance C added), indicating degradation of the substance, and a very pronounced negative net N mineralization within 7 d (–356%), which was interpreted as N immobilization. Only β-sitosterol showed strong inhibition of net N mineralization after 7 and 14 d (–59% and –26%, respectively) which was not interpreted as N immobilization, since there was no concomitant increase in C mineralization. Thus, an antimicrobial effect of β-sitosterol specificly on microorganisms of the N cycle was suggested, but there was no clear inhibitory effect caused by the antioxidant compounds.

    Description: Drug‐drug interactions have been demonstrated to alter cytochrome 2D6 (CYP2D6) enzyme phenotype due to inhibitor ingestion, although it is unclear how substrate interactions affect phenotype. This was a pragmatic clinical trial examining the kinetics of a CYP2D6 enzyme probe drug with and without CYP2D6‐dependent substrates. Patients were enrolled into an inpatient study unit, and orally administered a 2 mg microdose of dextromethorphan (DM) to probe enzyme activity with and without CYP2D6‐dependent drug‐drug interactions. Thirty‐nine subjects were enrolled in this trial. Twelve subjects were on no CYP2D6‐dependent drugs and 27 were on one or more CYP2D6‐dependent drugs. There were 1 poor metabolizer, 5 intermediate metabolizers, 31 normal metabolizers, and 2 ultra‐rapid metabolizers. Those with co‐ingestion of another CYP2D6‐dependent drug were 9.49 (95% confidence interval (CI): 1.54–186.41; P = 0.01) times more likely to have genotype‐phenotype discordance based upon the 3 hours dextrophan/dextromethorphan (DX/DM) ratio. CYP2D6 substrate co‐ingestions can cause genotype‐phenotype discordance.
