    Notes: The shock response of single crystals of the explosive pentaerythritol tetranitrate was studied from 3.4 to 19 GPa. Four orientations, 〈110〉, 〈001〉, 〈101〉, and 〈100〉 were studied. The shock Hugoniot and shock initiation sensitivities were measured in wedge tests. Intermediate velocity transitions were noted. A sensitivity anomaly near 4 GPa was discovered for 〈110〉 orientations. Chemiluminescent phenomena associated with this anomaly were studied using framing photography and uv/visible spectroscopy. Both plane-wave explosive and gas-gun impact experiments were done. Analysis of the steric hindrance occurring during edge dislocation motion in the shear flow associated with shock-induced uniaxial strain was done. The results corroborate the hypothesis that the orientation dependence of shock initiation sensitivity is due to the anisotropy of plastic flow, that is, the relative ease of slip in different orientations.

    Notes: Dwarf mistletoe (Arceuthobium spp.) is a hemiparasite that is said to be the single-most destructive pathogen of commercially valuable coniferous trees in many regions of the world. Although its destructive nature is well documented in many respects, its effects on the physiology of its host are poorly understood. In the present study, water and carbon relations were characterized over a range of scale from leaf to whole tree in large (40- to 50-m-tall) individuals of western hemlock (Tsuga heterophylla (Raf.) Sarg.) that were either heavily infected, or uninfected with hemlock dwarf mistletoe (Arceuthobium tsugense). Specific hydraulic conductivity (ks) of infected branches was approximately half that of uninfected branches, yet leaf-specific conductivity (kL) was similar because leaf area : sapwood area ratios (AL : AS) of infected branches were lower. Pre-dawn and minimum leaf water potential and stomatal conductance (gs) were similar among infected and uninfected trees because adjustments in hydraulic architecture of infected trees maintained kL despite reduced ks. Maximum whole-tree water use was substantially lower in infected trees (approximately 55 kg d−1) than in uninfected trees (approximately 90 kg d−1) because reduced numbers of live branches in infected trees reduced whole-tree AL : AS in a manner consistent with that observed in infected branches. Maximum photosynthetic rates of heavily infected trees were approximately half those of uninfected trees. Correspondingly, leaf nitrogen content was 35% lower in infected trees. Foliar δ13C values were 2.8‰ more negative in infected than in uninfected individuals, consistent with the absence of stomatal adjustment to diminished photosynthetic capacity. Adjustments in hydraulic architecture of infected trees thus contributed to homeostasis of water transport efficiency and transpiration on a leaf area basis, whereas both carbon accumulation and photosynthetic water use efficiency were sharply reduced at both the leaf and whole-tree scale.

    Notes: Summary A new variant of human erythrocyte carbonic anhydrase II (CA II) was discovered in a single Caucasian family during routine screening of blood samples from Melbourne, Australia. The normal and variant enzymes in the heterozygous CA II mixture, as well as a minor component of the normal enzyme, were resolved by isoelectric focusing following purification by a specific affinity matrix. Specific esterase activities of all three were very similar, but quite different Michaelis-Menten constants were noted for the minor component. No differences were noted with respect to inhibition by acetazolamide, but the minor component was more sensitive to chloride inhibition. Double diffusion analysis showed the immunological identity of the normal, variant, and minor components. Both the variant CA II and the minor component were less heat stable than the normal enzyme, but all forms showed identical rates of inactivation upon dialysis against the zinc chelator pyridine dicarboxylic acid. Amino acid analyses of the whole protein and the single difference peptide were consistent with a proline to histidine substitution in the variant. This was identified as 237 Pro→His by a process of elimination involving direct sequencing of tryptic and cyanogen bromide peptides. The numbering is by homology with the human CA I sequence.

    Notes: Summary As part of a study of the ‘whey’ proteins of various mammals, a comparison is made of the α-lactalbumins and lysozymes of the kangaroo and horse. In the milk of the red kangaroo (Megaleia rufa) there is only one α-lactalbumin and it occurs throughout lactation, but no lysozyme has been detected. There are two α-lactalbumins in the milk of the grey kangaroo (Macropus giganteus), one, designated α-lactalbumin Zone B, is present throughout lactation; the second, designated α-lactalbumin Zone A, is present only in late lactation. One lysozyme is also present. The milk of the horse (Equus caballus) contains one α-lactalbumin and at least one lysozyme. Partial amino acid sequences are proposed from sequence determination and from analyses of tryptic peptides compared with the known sequences of other α-lactalbumins and lysozymes.

    Notes: Summary The occurrence of the dominant ‘whey’ protein in samples of milk from 1180 sows is examined. It exhibits genetic polymorphism with some unusual features. Although immunologically different from bovine β-lactoglobulin, it is shown by chemical studies of the isolated protein to be a β-lactoglobulin. Two homozygous genetic variants, designated porcine β-lactoglobulin A and C, are isolated and their amino acid compositions and peptide maps compared. It is shown that the C variant has +1 His, −1 Gln, and +1 Asp, −1 Glu, with respect to the A variant. These variants, containingca. 162 residues per molecule, are considered in relationship to porcine β-lactoglobulins isolated by other workers. The sequence of the first 50 residues is determined and compared with sequence of the bovine protein. The sequences ofca. 70% of the remaining residues is proposed on the basis of the composition of tryptic peptides and assumed homology.

    Notes: Summary The occurrence of the ‘whey’ protein, α-lactalbumin, in pig (Sus scrofus) milk samples from 904 sows is examined. A semi-discontinuous buffer system has been developed to detect the existence of genetic polymorphism. There are two homozygous variants, designated A and B. Both variants are isolated and it is shown by peptide and sequencing studies that the A variant differs from the B variant by having an Arg residue substituted for the Lys residue at the N-terminus of the molecule. The sequence of the first thirty residues is determined and compared with those of related α-lactalbumins.
