    Description: The human gut microbiome is critical to health and wellbeing. It hosts a complex ecosystem comprising a multitude of bacterial species, which contributes functionality that would otherwise be absent from the host. Transient and commensal bacteria in the gut must withstand many stresses. The influence of mobile genetic elements such as plasmids in stress adaptation within the ecosystem is poorly understood. Using a mobilomic approach we found evidence for plasmid-mediated osmotolerance as a phenotype amongst the Proteobacteria in healthy faecal slurries. A transconjugant carrying multiple plasmids acquired from healthy faecal slurry demonstrated increased osmotolerance in the presence of metal salts, particularly potassium chloride, which was not evident in the recipient. Pyrosequencing and analysis of the total plasmid DNA demonstrated the presence of plasmid-borne osmotolerance systems (including KdpD and H-NS) which may be linked to the observed phenotype. This is the first report of a transferable osmotolerance phenotype in gut commensals and may have implications for the transfer of osmotolerance in other niches.

    Description: The mechanistic understanding and control over transformations of multi-unsaturated hydrocarbons on transition metal surfaces remains one of the major challenges of hydrogenation catalysis. To reveal the microscopic origins of hydrogenation chemoselectivity, we performed a comprehensive theoretical investigation on the reactivity of two α,β-unsaturated carbonyls—isophorone and acrolein—on seven (111) metal surfaces: Pd, Pt, Rh, Ir, Cu, Ag, and Au. In doing so, we uncover a general mechanism that goes beyond the celebrated frontier molecular orbital theory, rationalizing the CC bond activation in isophorone and acrolein as a result of significant surface-induced broadening of high-energy inner molecular orbitals. By extending our calculations to hydrogen-precovered surface and higher adsorbate surface coverage, we further confirm the validity of the "inner orbital broadening mechanism" under realistic catalytic conditions. The proposed mechanism is fully supported by our experimental reaction studies for isophorone and acrolein over Pd nanoparticles terminated with (111) facets. Although the position of the frontier molecular orbitals in these molecules, which are commonly considered to be responsible for chemical interactions, suggests preferential hydrogenation of the CO double bond, experiments show that hydrogenation occurs at the CC bond on Pd catalysts. The extent of broadening of inner molecular orbitals might be used as a guiding principle to predict the chemoselectivity for a wide class of catalytic reactions at metal surfaces.

    Description: The dual-throat engine is one of the dual nozzle engine concepts studied for advanced space transportation applications. It provides a thrust change and an in-flight area ratio change through the use of two concentric combustors with their throats arranged in series. Test results are presented for a dual throat thruster burning gaseous oxygen and hydrogen at primary (inner) chamber pressures from 380 to 680 psia. Heat flux profiles were obtained from calorimetric cooling channels in the inner nozzle, outer or secondary chamber and the tip of the inner nozzle. Data were obtained for two nozzle spacings over a chamber pressure ratio (secondary/primary) range of 0.45 to 0.83 with both chambers firing (Mode I). Fluxes near the end of the inner nozzle were significantly higher than in Mode II when only the inner chamber was fired, due to the flow separation and recirculation caused by the back pressure imposed by the secondary chamber. As the pressure ratio increased, these heat fluxes increased and the region of high heat flux relative to Mode II extended farther upstream. The use of the gaseous hydrogen bleed flow in the secondary chamber to control heat fluxes in the primary plume attachment region was investigated in Mode II testing. A thermal model of a dual throat thruster was developed and upgraded using the experimental data.

    Description: The objective of this project was to develop a model for CO2, O2, H2O, and nitrogen use during the life cycle of wheat. Spreadsheets and accompanying graphs were developed to illustrate plant population reactions to environmental parameters established in the Controlled Ecological Life Support System (CELSS) program at Kennedy Space Center, Fl. The spreadsheets and graphs were produced using validated biomass production chamber (BPC) data from BWT931. Conditions of the BPC during the 83 day plant growth period were as follows: The BPC area is 27.8 m(exp 2), volume is 113 m(exp 3). Temperatures during the 83 day plant growth period ranged from 16.3 to 24.8 C during the light cycle (except for day 69, when the minimum and maximum temperatures were 7.7 C and 7.9 C, respectively) and 14.5 C and 23.6 C during the dark cycle (except for day 49, when the minimum and maximum temperatures were 11.1 C and 11.3 C, respectively). Relative humidity was 85 percent for the first seven days of plant growth, and 70 percent thereafter. The plant leaf canopy area was 10 m(exp 2). Presented is a list and explanation of each spreadsheet and accompanying graph(s), conditions under which the data were collected, and formulas used to obtain each result.

    Description: A propulsion system analysis of a dual fuel, dual throat engine for launch vehicle application was conducted. Basic dual throat engine characterization data are presented to allow vehicle optimization studies to be conducted. A preliminary baseline engine system was defined. Dual throat engine performance, envelope, and weight parametric data were generated over the parametric range of thrust from 890 to 8896 KN (200K to 2M lb-force), chamber pressure from 6.89 million to 34.5 million N/sq m (1000 to 5000 psia) thrust ratio from 1.2 to 5, and a range of mixture ratios for the two tripropellant combinations: LO2/RP-1 + LH2 and LO2/LCH4 + LH2. The results of the study indicate that the dual fuel dual throat engine is a viable single stage to orbit candidate.

    Description: This study identifies and evaluates promising LO2/HC rocket engine cycles, produces a consistent and reliable data base for vehicle optimization and design studies, demonstrates the significance of propulsion system improvements, and selects the critical technology areas necessary to realize an improved surface to orbit transportation system. Parametric LO2/HC engine data were generated over a range of thrust levels from 890 to 6672 kN (200K to 1.5M 1bF) and chamber pressures from 6890 to 34500 kN (1000 to 5000 psia). Engine coolants included RP-1, refined RP-1, LCH4, LC3H8, LO2, and LH2. LO2/RP-1 G.G. cycles were found to be not acceptable for advanced engines. The highest performing LO2/RP-1 staged combustion engine cycle utilizes LO2 as the coolant and incorporates an oxidizer rich preburner. The highest performing cycle for LO2/LCH4 and LO2/LC3H8 utilizes fuel cooling and incorporates both fuel and oxidizer rich preburners. LO2/HC engine cycles permitting the use of a third fluid LH2 coolant and an LH2 rich gas generator provide higher performance at significantly lower pump discharge pressures. The LO2/HC dual throat engine, because of its high altitude performance, delivers the highest payload for the vehicle configuration that was investigated.

    Description: Well defined nocturnal convection, together with distinct maximum precipitation events in Nebraska, were associated with strong diurnal wind oscillations at 850 mb in Oklahoma, Kansas and Missouri in the present study of pronounced precipitation and low level circulation variations over the Great Plains on May 5-10, 1979. A numerical prediction model, used to relate boundary layer flow modulations to synoptic scale flow for this case, is driven by a diurnal cycle. Experimental forecasts displayed diurnal wind modulations at 500 m, with rising motions of the order of 4 cm/sec at the 2200 m level, along the leading edge of the nocturnal jet.
