    Notes: Red light effects on the uptake of 14C and of 32P was studied by observing leaf sections from 8-day-old etiolated corn leaves that were placed on various substrates following a brief exposure to red radiant energy. There was a general increase of 14C uptake over dark levels into all metabolite fractions that were prepared. This is in contrast with results obtained previously in which leaf samples were first floated on substrate and then irradiated (Mitrakos et al. 1967, Price et al. 1965). The latter tests resulted in a general decrease in sugar and starch as well as 14C content of all fractions. However, under both types of experimental conditions the red light effect manifested itself as an increase in hexosemonophosphate turn-over rate and accumulation of radioactivity in the cell wall polysaccharide fraction. The present data further substantiate the previous work in that they demonstrate the regulatory influence of the hexose pool size on the intermediary metabolism and the manifestation of the phytochrome responses. From the data thus far obtained it cannot be determined as to whether or not phytochrome mediates by controlling phosphorylating activity or through control of specific enzymatic processes that lead to observable cell wall polysaccharide synthesis and leaf unrolling.

    Notes: Abstract  Members of the Tcf/Lef family interact with β-catenin to activate programs of gene expression during development. Recently β-catenin was shown to be essential for establishing cell fate along the animal-vegetal axis of the sea urchin embryo. To examine the role of Tcf/Lef in sea urchins we cloned a Strongylocentrotus purpuratus Tcf/Lef homolog. Expression of SpTcf/Lef was maximal when β-catenin became localized to nuclei of vegetal blastomeres, consistent with its acting in combination with β-catenin to specify vegetal cell fates. Expression of a dominant-negative SpTcf/Lef inhibited primary and secondary mesenchyma, endoderm, and aboral ectoderm formation in a manner similar to that observed when nuclear accumulation of β-catenin was prevented. Our results suggest that SpTcf/Lef functions by interacting with β-catenin to specify cell fates along the sea urchin animal-vegetal axis.

    Notes: Abstract Three data sets of suspensate concentrations, collected under different sampling plans, were analyzed to develop a minimal sampling period, consistent with a predetermined precision. Individual samples collected during any part of a given day were within ±50% of the daily average. In the main channel of the estuary, a monthly sampling scheme closely approximated a weekly pattern; however, seasonal sampling was unsatisfactory. Outside the main channel, weekly, monthly, and seasonal sampling patterns did not differ greatly. Variability in suspensate concentrations must be considered when calculating sediment flux in estuarine and coastal waters.

    Notes: Abstract Observations made with primary leaves of Phaseolus vulgaris L. demonstrated that phytochrome modulates light-induced stomatal movement. Removal of the far-red-absorbing form of the pigment (Pfr) with far-red (FR) radiation decreased the time required by the stomata to reach maximal opening following a dark-to-light transition; this effect of FR was fully reversible with red. Removal of Pfr with FR also decreased the time required to reach maximal closure following a light-to-dark transition, and the rate of closure was dependent on the final irradiation treatment before darkness. No evidence was found for phytochrome involvement in determining stomatal aperture under constant conditions of either darkness of light.

    Notes: Summary A survey of the distribution of tracer in preparations from etiolated corn leaf sections which had been fed with uniformly labeled glucose (glucose-U-C14) and then exposed to ten minutes of red light, failed to show the presence of any new or specifically labelled metabolite. The earliest consequence of the light-treatment was a loss of tracer from the soluble sugars accompanied by an accumulation of C14 in the cell-wall polysaccharides. Other light effects were reduced labelling of amino acids and higher phosphate turnover rates. The survey did not elucidate the nature of the first biochemical response after an exposure of an etiolated seedling to radiant energy, but it suggests several areas of further investigation.

    Notes: Abstract The influence of far-red (FR; 700–800 nm) radiation on steady-state stomatal conductance and net photosynthesis in P. vulgaris has been studied. Whereas FR radiation alone was relatively ineffective, addition of FR to a background of white light (WL; predominantly 400–700 nm) resulted in increased stomatal conductance. Stomata exhibited a marked diurnal sensitivity to FR. The action maximum for enhancing stomatal conductance was near 714 nm. A combination of FR and infra-red (IR; 〉800 nm) enhanced net photosynthesis when added to a background of WL. When IR alone was added to WL, there was a net decrease in photosynthesis, indicating that it is the FR waveband which is responsible for the observed photosynthetic effects. Naturally occurring levels of FR radiation (235 μmol·m-2·s-1) in vegetation-canopy shade enhanced net photosynthetic CO2 gain by 28% when added to a background of 55 μmol·m-2·s-1 WL.
