    Notes: This paper presents the architecture of a system for robotic welding of complex tasks. The system integrates off-line programming, control of redundant robots, collision-free motion planning and sensor-based control. An implementation for pipe structure welding made at Odense Steel Shipyard Ltd., Denmark, demonstrates the system can be used for automatic welding of complex products in one-of-a-kind production.

    Notes: Abstract A crossing programme including 30 species and 40 cytotypes within the genusHordeum was undertaken. Viable hybrids were obtained in 302 combinations, 15 of which were intraspecific. Differences in seed set and in germination were observed in crosses between different groups of species. Obtaining crosses between different taxonomic groups was generally more difficult when diploid material was used. Some species, e.g.,H. lechleri, H. jubatum, andH. brachyantherum showed a higher crossability than others. The chromosome numbers of the hybrids were usually those expected from the parental numbers but aneuploid series around the expected numbers were rather frequent. Three cases of unreduced gametes were found. Selective chromosome elimination was restricted to combinations including eitherH. vulgare orH. bulbosum.—Despite a very diverse morphology, all South American diploid species together with the two North American diploidsH. intercedens andH. pusillum appear to be closely related. The hexaploid American speciesH. procerum, H. lechleri, andH. arizonicum are also related. The two North American tetraploid speciesH. jubatum andH. brachyantherum sometimes form semifertile hybrids. The Asiatic speciesH. roshevitzii appears to be related to both North and South American taxa.

    Notes: Abstract The similar-looking basic genomes ofHordeum bulbosum (2x and 4x) have five rather similar metacentric, one submetacentric, and one satellited choromosome. C-banding patterns are characterized by one or two centromeric, or juxtacentromeric, small to larger bands in most chromosomes, by bands at the nucleolar organizers, by small or very small telomeric bands, and by the nearly complete lack of intercalary bands. Banding pattern polymorphism is widespread. Banding patterns supported by chromosome morphology enable identification of homologues, and discrimination between non-homologues inH. bulbosum (2x). The C-banded karyotype ofH. bulbosum (4x) supports an autopolyploid origin, but it was possible to identify only homologues of submetacentrics and SAT-chromosomes.

    Notes: Summary The intergeneric hybrid Hordeum vulgare x Psathyrostachys fragilis was fairly easily obtained. During each growing season the intermediate, perennial hybrid yielded haploid tillers of H. vulgare. Late in one season few, hybrid tillers headed. The morphology, cytology and enzymatic patterns of hybrid and haploid tillers were investigated.

    Description: Abstract 4195 Background: Daclizumab, a humanized monoclonal antibody, binds CD25 and blocks formation of the high affinity IL-2 receptor on T cells. A multi-center, randomized, double-blind clinical trial using daclizumab to perturb IL-2 signaling as a way of reducing T cell mediated graft versus host disease (GVHD) after allogeneic unrelated bone marrow transplantation (BMT) was completed 18 years ago. We report long term outcomes and biological correlates. Methods and Patients: Between April, 1993 and December, 1994, 209 adult or pediatric patients receiving marrow from an unrelated donor for hematologic malignancies or severe aplastic anemia at 12 sites in Europe and North America were randomized to one of three arms: 5 weekly doses of placebo (arm A, n=64); 0.3 mg/kg daclizumab (arm B, n=69); or 1.2 mg/kg daclizumab (arm C, n=76) to a maximum of 100 mg beginning on the day before BMT. Conditioning included total body irradiation (TBI) (1200–1440 cGy). All patients received methotrexate (15 mg/m2 day 1; 10 mg/m2 days 3, 6, 11) plus cyclosporine for at least 180 days. The study was designed to provide 80% power to detect a reduction in the incidence of acute GVHD (grade II-IV, requiring treatment with corticosteroids to day +100) from 80% to 55%. Randomization was stratified by donor HLA (matched vs. mismatched) and age (=20). Arms were well balanced for HLA matching, age, disease risk, TBI dose, and diagnosis. Diagnoses were 31 ALL, 30 AML, 118 CML, 16 MDS, and 14 other. Median age was 31 years (range 1–54); 61% were male; 45% had high risk disease (14% AML with active disease or beyond 2nd remission, 20% CML beyond first chronic phase); 29% had a HLA-A, -B or -DR mismatched donor. T cells from a subset of patients (n=107) were collected at days 11–35, days 36–80, and days 81–100 and analyzed by flow cytometry for expression of total CD25, daclizumab binding, and free CD25 binding sites. Samples from arm A and arm C, including those from 1 year long term follow up (LTFU) were also evaluated for T cell phenotype. Results: The incidence of grade II-IV acute GVHD for arms A, B, and C were 34%, 42%, and 45%, and results did not differ between arm A vs. arm B (p=0.60) or arm C (p=0.38). Since other clinical outcomes did not differ significantly between arm B and arm C, the two daclizumab dose arms were combined for post-hoc exploratory analysis. There was a suggestion that the daclizumab arms showed decreased risk of relapse (HR 0.57, 95% CI: 0.32–1.00, p=0.06) and increased risk of chronic GVHD (HR 1.49, 95% CI: 0.96–2.31, p=0.07) compared to the placebo arm. Daclizumab did not increase OS (HR 0.89, 95% CI: 0.61–1.29, p=0.54) compared to placebo, and there was no interaction between disease risk and OS.     Description: Patients who receive allogeneic hematopoietic stem cell transplants have an increased risk for new malignancies because of several risk factors, including conditioning with radiation and chemotherapy, immune modulation, and malignant primary disease. The frequency of and risk factors for malignant neoplasm in long-term survivors should be assessed. A former analysis of the EBMT observing the 1036 patients of this study with a median observation time of 10.7 years showed older patient age and immunosuppressive treatment of chronic graft-versus-host disease as main risk factors for secondary malignancies. We have tried to determine the cumulative incidence and define potential risk factors for new malignancies in long-term survivors after marrow transplantation in a retrospective multi center follow-up study. This study of the Late Effects Working Party was performed with 45 transplantation centers cooperating in the European Cooperative Group for Blood and Marrow Transplantation. 1036 consecutive patients who underwent transplantation for leukemia, lymphoma, inborn diseases of the hematopoietic and immune systems, or severe aplastic anemia. Patients were transplanted before December 1985 and had survived more than 5 years. Reports on malignant neoplasms were evaluated, and the cumulative incidence was compared to that in the matched general population. Patient age and sex, primary disease and disease stage at transplantation, histocompatibility of the donor, conditioning regimen, type of prophylaxis of graft-versus-host disease, development of acute and chronic graft-versus-host disease, and treatment of chronic graft-versus-host disease were evaluated as variables. Univariate analysis was performed using the log rank test for the time until malignancy occurred; significant risk factors were studied in multivariate analysis (Cox regression). Median follow-up since transplantation was 17.9 years (range, 5 to 32.3 years). Malignant neoplasms were seen in 114 patients; the cumulative incidence was 4.0% at 10 years, 8.5% at 15 years, 14.0% at 20 years and 21.0% at 25 years.     Description: Studies of genetically identical-twin transplants are a novel opportunity to study how transplants work because: (1) there is no allogeneic effect; (2) no leukemia cells in the graft; and (3) no graft exposure to therapy. We conducted an international study that identified 19 subjects who received syngeneic bone marrow (N=11) or blood cell (N=8) transplants after myeloablative conditioning. 11 were males; age 51 y (range, 37–68 y). 18 received total body radiation. None had Richter transformation. Interval from diagnosis to transplant was 27 mo (5–171 mo). At transplant 8 had Rai stage 3/4, 5 had 〉50x10e9/L lymphocytes, 10 received ≥3 prior therapies, 8 had prior fludarabine, and 5 had a prior complete remission (CR). 18 engrafted and 13 achieved posttransplant CR; median time to CR was 3 mo (1–5 mo). Probability of 100 d survival was 89% (95% CI, 72–99%).10 subjects are alive (8 disease-free) at median follow-up of 63 mo (9–116 mo). Ten subjects either never achieved CR (N=6) or relapsed posttransplant (N=4). 5-y cumulative incidence of relapse was 52% (27–77%). Estimated 5-y survival and disease-free survival were 59% (34–81%) and 43% (20–67%), respectively. Causes of death included interstitial pneumonitis (N=1) and leukemia (N=8). 5-y cumulative incidence of treatment-related mortality (TRM) is 5% (0–20%). We used a highly sensitive (10e-4 to 10e-5) PCR method to examine post transplant blood (2 pts) or bone marrow (2 pts) samples for the tumor specific IgH gene (CDR)III to assess minimal residual disease (MRD). IgH CDR III was PCR amplified in pre transplant B-CLL samples from 4 pts to obtain the sequence to design tumor-specific primer probes for MRD. No evidence of MRD was detected in two pts at 12 and 21 mo posttransplant. A very weak clonal signal was identified in one pt at 64 mo. All three of these pts were in continuous clinical CR at 12, 60, and 66 mo, respectively. In one pt, who relapsed with B-CLL 6 y after transplant, molecular studies at 10 y follow-up demonstrated a very strong molecular signal but of a different clone. Additional investigation identified familial CLL where the donor was also diagnosed with B-CLL soon after marrow donation. Molecular analysis of the donor B-CLL showed a clone identical to the recipient’s post-transplant relapse, strongly indicating B-CLL transmission at the time of transplant. This study demonstrates that identical twin transplants can be performed in advanced B-CLL with little TRM and with a high-rate of durable clinical and molecular remissions. The 5-y leukemia relapse rate of 52% is higher than that in studies of similar subjects receiving allotransplants but lower than after autotransplants. We also report B-CLL transfer from a twin donor demonstrating the need for careful evaluation of allogeneic donors prior to graft collection.

    Description: The work presents an experimental investigation into the motion of and hydrodynamic forces along a single flexible stem in regular waves. The experiment covers a large range in relevant non-dimensional parameters: the drag-to-stiffness ratio, the inertia-to-stiffness ratio, the Keulegan-Carpenter number and the Reynolds number. The two first parameters relate to the response of the stem in waves and thus account for material properties, while the two last parameters are relevant for hydrodynamic forces on the stem. The displacement of the stem was captured with a digital video camera and the displacement along the stem was captured for every 2.5 mm at 25 Hz. This unique laboratory data set allowed for the following analyses: (i) Determination of the relevant non-dimensional parameter to predict the stem motion and shape. (ii) A direct comparison between the measured force for mimics of two lengths (0.15 m and 0.30 m) illustrating the force reduction potential for flexible mimics. (iii) Direct evaluation of the average force coefficients (drag) and (inertia) for the flexible stems. (iv) The distributed external hydrodynamic loading and the internal shear forces were estimated from the laboratory experiments. The distribution of the shear force helped to understand the breakage mechanisms of flexible stems. (v) A linkage between phase lags and internal shear forces was suggested. The data set is considered valuable as validation material for numerical models of stem motion in waves. © 2019 Cambridge University Press.
