    Notes: To explain the differing germination behaviour of seeds of wild type, gibberellin-deficient (gibl) or abscisic acid-deficient (sit”) mutants of tomato (Lycopersicon esculentum Mill. cv. Moneymaker), growth and water relations of fruit tissues, seeds and embryos were determined during development. The objective was to determine whether the hormones influenced water relations of fruit and seed tissues and as a consequence germinability. Despite the up to 70% lower fruit weight of the mutants, the fresh and dry seed weights were similar to that of the wild type. Water relations of pericarp and locular tissues in wild type and gibl fruits were characterized by a slowly increasing Ψa that reached a constant value Ψ of −0.60 MPa at 35–40 days after anthesis, resulting in a complete loss of turgor. However, sitw fruit tissues maintained their turgor throughout development, mainly as a result of considerably lower Ψ values but similar Ψ values compared with the other genotypes. The Ψ of wild type and gibl seeds decreased from -0.50 to -0.80 MPa and Ψ from -0.80 to -1.00 MPa between day 30 and 40. From day 40 onwards, Ψp was similar in both genotypes, approximately 0.20 MPa. As in the fruit tissues, sitw seed water relations were also characterized by higher turgor values than the other genotypes. Up to day 40, Ψp was ca 0.40 MPa, dropped temporarily to zero, and increased again to approximately 0.40 MPa at day 50. Embryo water relations of both mutants deviated from the wild type in that Ψp of the gibl and sitw embryos remained at 3.5 MPa and 2.5 MPa, respectively, from day 40 onward, whereas in wild type embryos Ψp decreased from 3.0 MPa at day 35 to approximately 1.0 MPa at 50 days after anthesis. This was mainly due to an increasing Ψn which was absent in the mutants. Throughout development there was equilibrium between Ψ of pericarp, locular tissues and seeds but between embryo and seed or fruit tissues Ψ gradients of up to 1.5 MPa were calculated in the wild type, and up to 1.0 MPa in the mutants. Thus, precocious germination is prevented by the action of the fruit's osmotic environment and ABA on the seed tissues surrounding the embryo and not the embryo itself. Embryos have a Ψ which is low enough to overcome the solute potential of the fruit tissues throughout development.

    Notes: It is not known how embryos of seeds of the Pinaceae protrude from their enclosing tissues to complete germination. Prior to protrusion of the radicle there is an increase in endo-β-1,4-mannanase (EC 3.2.1.78) activity associated with weakening of the micropylar megagametophyte/nucellus from seeds of white spruce (Picea glauca [Moench.] Voss). Mannanase activity is present as three isoforms (pI values 5.0, 4.8, 4.7) in both the embryo and surrounding structures (megagametophyte and nucellus) prior to and during imbibition. Activity of all the isoforms increases in the chalazal and micropylar megagametophyte during germination. Activity then declines after the testa splits, typically 1 day prior to radicle protrusion, due partially to its leaching from the seed into the surrounding water. Activity increases in the cotyledons and axis as the embryo commences elongation. Seeds from dormant seedlots exhibit a lower germination percentage, relative to seeds from nondormant seedlots, and the force necessary for the embryo to puncture the surrounding structures tends to be greater. Although similar mannanase activities are present in unimbibed seeds of dormant and nondormant seedlots, during germination, enzyme activity in seeds of dormant seedlots is lower. Moist chilling alleviates dormancy in the seeds of the Pinaceae and, during 3 weeks of this treatment, mannanase activity slowly increases. After 3 weeks of moist chilling and regardless of whether the seedlot was dormant or not prior to moist chilling, the force necessary to puncture the micropylar megagametophyte and nucellus is lower, and the speed of germination greater. Seeds from previously dormant seedlots also complete germination to a greater percentage, relative to unchilled seeds from dormant seedlots. Upon transfer to 25°C, mannanase activity in moist-chilled seeds decreases during germination of all seedlots regardless of their previous dormancy status.

    Notes: Abstract A current hypothesis is that endo-β-mannanase activity in the endosperm cap of tomato (Lycopersicon esculentum Mill. cv. Moneymaker) seeds is induced by gibberellin (GA) and weakens the endosperm cap thus permitting radicle protrusion. We have tested this hypothesis. In isolated parts, the expression of endo-β-mannanase in the endosperm after germination is induced by GAs, but the expression of endo-β-mannanase in the endosperm cap prior to radicle protrusion is not induced by GAs. Also, abscisic acid (ABA) is incapable of inhibiting endo-β-mannanase activity in the endosperm cap, even though it strongly inhibits germination. However, ABA does inhibit enzyme activity in the endosperm and embryo after germination. There are several isoforms in the endosperm cap and embryo prior to radicle protrusion that are tissue-specific. Tissue prints showed that enzyme activity in the embryo spreads from the radicle tip to the cotyledons with time after the start of imbibition. The isoform and developmental patterns of enzyme activity on tissueprints are unaffected when seeds are incubated in ABA, even though germination is inhibited. We conclude that the presence of endo-β-mannanase activity in the endosperm cap is not in itself sufficient to permit tomato seeds to complete germination.

    Notes: Abstract The role of cis-abscisic acid (ABA) and gibberellins (GAs) in the induction of cell-cycle activities has been studied during imbibition and subsequent germination of tomato seeds. Using flow cytometry, nuclear replication activity was investigated in embryo root tips isolated from seeds of the ABA-deficient mutant sit w , the GA-deficient mutant gib-1, and the wild-type (MM) tomato (Lycopersicon esculentum Mill. cv. Moneymaker) upon imbibition in water, 10 μM GA4+7, 5 μM ABA or 5 μM ABA+10 μM GA4+7. The nuclei of fully matured dry MM, sit w and gib-1 seeds predominantly showed 2C DNA signals, indicating that the cell-cycle activity of most root-tip cells had been arrested at the G1 phase of nuclear division. However, ABA-deficient sit w seeds contained a significantly higher amount of G2 cells (4C DNA) compared with the other genotypes, suggesting that, during maturation, cell-cycle activity in sit w seeds is less efficiently arrested in G1. Upon imbibition in water, an induction of the 4C signal, indicating nuclear replication, was observed in the root tip cells of both MM and sit w embroys. The augmentation in the 4C signal occurred before visible germination. Gib-1 seeds did not show cell-cycle activity and did not germinate in water. Upon imbibition in GA4+7, both cell-cycle activity and subsequent germination were enhanced in MM and sit w seeds, and were induced in gib-1. In ABA, the germination of MM and sit w seeds was inhibited while nuclear replication of these seeds was not affected. It is concluded that GA influences germination by acting upon processes that precede cell-cycle activation, while ABA affects growth by acting upon processes that follow cell-cycle activation.

    Notes: Summary Resumption of DNA synthetic activities and β-tubulin accumulation was studied in embryo organs of germinating cucumber (Cucumis sativus L.) seeds. Flow-cytometric analysis indicated the existence of 2C, 4C, and 8C nuclei in the radicle of mature embryos, whereas in cotyledons most of the cells contained nuclei with 2C DNA content. Upon imbibition of water, nuclear DNA replication was initiated in the radicle within 15 h, subsequently spreading towards the cotyledons. Bromodeoxyuridine incorporation preceded detectable changes in the relative amounts of DNA, implying the occurrence of putative DNA repair. Organellar DNA synthesis occurred independently of the nuclear DNA synthetic cycle. Western blotting and immunohistochemical localization demonstrated that the constitutive level of β-tubulin originated from preserved β-tubulin granules. During imbibition, disappearance of fluorescent tubulin granules, accumulation of β-tubulin, and formation of microtubular cytoskeleton were found in the radicle, but not in the cotyledon areas. Mitosis only occurred after radicle protrusion at 21 h of imbibition. It is concluded that the differences in the initiation and progress of these cellular and molecular events are associated with the discrete behaviors of the radicle and the cotyledons upon imbibition. The formation of cortical microtubular cytoskeleton and the accumulation of tubulins are important features in preparation of radicle protrusion, whereas DNA synthesis may contribute to postgerminative growth.

The equations involved + 1 spatially dependent nearest-neighbor interactions. We locate a critical fixed point and obtain the exact thermal critical indexy T = 2. A special trajectory of the full nonlinear RG transformation is found and the free energy of the corresponding initial state calculated.

    Description: Grasses are among the most resilient plants, and some can survive prolonged desiccation in semiarid regions with seasonal rainfall. However, the genetic elements that distinguish grasses that are sensitive versus tolerant to extreme drying are largely unknown. Here, we leveraged comparative genomic approaches with the desiccation-tolerant grass Eragrostis nindensis and the related desiccation-sensitive cereal Eragrostis tef to identify changes underlying desiccation tolerance. These analyses were extended across C4 grasses and cereals to identify broader evolutionary conservation and divergence. Across diverse genomic datasets, we identified changes in chromatin architecture, methylation, gene duplications, and expression dynamics related to desiccation in E. nindensis. It was previously hypothesized that transcriptional rewiring of seed desiccation pathways confers vegetative desiccation tolerance. Here, we demonstrate that the majority of seed-dehydration–related genes showed similar expression patterns in leaves of both desiccation-tolerant and -sensitive species. However, we identified a small set of seed-related orthologs with expression specific to desiccation-tolerant species. This supports a broad role for seed-related genes, where many are involved in typical drought responses, with only a small subset of crucial genes specifically induced in desiccation-tolerant plants.
