    Notes: Abstract  CD20 is a hallmark antigen of B lymphocytes. Its expression is restricted to precursor and mature B cells but it is not expressed on plasma cells. The protein is a membrane-embedded phosphoprotein that appears likely to transverse the membrane four times. Its function is unknown although CD20 has been variously proposed to play a role in B-cell activation, proliferation, and calcium transport. A unique homologue of human CD20 has been described in mouse, which also shows a B-cell-specific pattern of expression. Here we describe the generating of mice carrying a CD20 gene disruption. So far, we have failed to detect any major effect of the gene disruption on the differentiation and function of B lymphocytes as judged by the expression of surface markers, antigen receptor signaling, proliferative responses, or calcium uptake. We did note, however, that the mice homozygous for the gene disruption [generated by intercrossing (129 × C57BL/6)F1 CD20 +/- heterozygotes] showed a substantial depletion of the sub-population of peritoneal B cells that lack expression of the B220 (RA3–6B2) isoform of CD45. The loss of the IgM+ 6B2- peritoneal B cells is not, however, attributable to the CD20 gene disruption itself. Rather, it segregates with a polymorphic difference between the 129 and C57BL/6 strains that is linked to the CD20 locus which, intriguingly, is itself close to the CD5 gene. This demonstrates that caution must be exercised when comparing the phenotypes of F2 litter-mates generated from crosses between 129 embryonic stem-cell-derived chimeras and mice of other strains.

    Notes: Abstract A polymorphic microsatellite marker (D2S125) was recently reported to show significant linkage to non-insulin dependent diabetes mellitus (NIDDM) in a population of Mexican-American affected sib-pairs. We have used a simple non-isotopic screening technique employing the polymerase chain reaction (PCR) with a biotinylated primer to study the genetic linkage and allele frequency distribution of the D2S125 marker in a population of 109 British NIDDMs (62 possible affected sib-pairs). The analysis provided no evidence for linkage of the D2S125 marker in the British subjects (MLS = 0.029, P 〉 0.05). The PCR screening method used proved to be a convenient and reliable alternative to the radiolabelling of PCR products.

    Description: Changes in penguin populations on the Antarctic Peninsula have been linked to several environmental factors, but the potentially devastating impact of volcanic activity has not been considered. Here we use detailed biogeochemical analyses to track past penguin colony change over the last 8,500 years on Ardley Island, home to one of the Antarctic Peninsula’s largest breeding populations of gentoo penguins. The first sustained penguin colony was established on Ardley Island c. 6,700 years ago, pre-dating sub-fossil evidence of Peninsula- wide occupation by c. 1,000 years. The colony experienced five population maxima during the Holocene. Overall, we find no consistent relationships with local-regional atmospheric and ocean temperatures or sea-ice conditions, although the colony population maximum, c. 4,000–3,000 years ago, corresponds with regionally elevated temperatures. Instead, at least three of the five phases of penguin colony expansion were abruptly ended by large eruptions from the Deception Island volcano, resulting in near-complete local extinction of the colony, with, on average, 400–800 years required for sustainable recovery.

    Description: Dataset: Hurricane Harvey Coral Gene Expression
    Description: To capture the immediate effects of storm-driven freshwater runoff on coral and symbiont physiology, we leveraged the heavy rainfall associated with Hurricane Harvey in late August 2017 by sampling FGB coral gene expression at two time points: September 2017, when surface water salinity was reduced (∼34 ppt); and 1 month later when salinity had returned to typical levels (∼36 ppt in October 2017). This dataset includes Sequence Read Archive (SRA) and BioSample accessions under BioProject PRJNA552981 at The National Center for Biotechnology Information. It also contains sample information and species names for samples collected the east and west banks of the Flower Garden Banks National Marine Sanctuary (FGBNMS) at 80ft.     Description: Ireland and Wales have a combined coastline of approximately 9,000 km in length, comprised of a combination of rock and soft sediment. Many archaeological and historical sites along this coastline are at risk of being damaged or destroyed by the present and future effects of climate change. The EU funded CHERISH Project (Climate, Heritage and Environments of Reefs, Islands, and Headlands) of the Irish and Welsh regional seas is researching these threats. This chapter focuses on those methods and applications within the CHERISH toolkit and explores the different ways 3D data collected within a project is being used to derive seamless terrain datasets to close the data deficit (“white ribbon”) which currently exists between the terrain and marine environments. These methods are currently being developed and employed to record, map, analyse and interpret the 3-dimensional nature of both the coastal and marine historic environments.

    Description: Future anthropogenic climate forcing is forecast to increase storm intensity and frequency over Northern Europe, however understanding the significance of such a change is difficult because the natural variability of storminess beyond the range of instrumental data is poorly known. Here we present a decadal-resolution record of storminess covering the Late Holocene, based on analysis of a core taken from the peat bog of Cors Fochno in mid-Wales, UK, in 2010. Storminess is indicated by variations in the minerogenic content (aeolian sand deposits) as well as bromine deposited from sea spray. A 4 m long peat core was sampled using a Russian corer from the centre of the bog (52°30'9''N; 4°0'39''W). Chronological constraints were provided by AMS radiocarbon dating of 5 samples along the core, which showed that the core spanned a period from 4460-120 cal yrs BP.

    Description: A 4 m long peat core was sampled using a Russian corer from the centre of the bog (52°30'9''N; 4°0'39''W). Chronological constraints were provided by AMS radiocarbon dating of 5 samples along the core, which showed that the core spanned a period from 4460-120 cal yrs BP. Samples of equal volume (2.3 cm3) from 1 cm contiguous intervals along the core were processed using the loss-on-ignition method (Heiri et al. 2001), leaving the ignition residue, which can be a proxy for storminess.
