Summary A method is described for the production with colcemid of micronucleate cells from normal human fibroblasts (Detroit 550) and human aneuploid D98/AH2 cells. The colcemid dose is titrated to a concentration that prevents cellular attachment to, and spreading on, the substrate following attempted mitosis. Washing out the drug with growth medium followed by incubation in drug-free medium for 12 to 24 h produces micronucleate cells. The percentage of micronucleation obtainable is 50 to 70% by treatment with 0.1 ug/ml colcemid for 24 h followed by incubation of floating blocked mitotic cells in drug-free growth medium.
