    Notes: Abstract The residual value of superphosphate and several rock phosphates was measured in three field experiments in Western Australia. The rock phosphates were Christmas Island C-grade ore, calcined C-grade ore (Calciphos) and apatite rock phosphates. The predictive capacity of the Colwell, Olsen and Bray 1 soil tests for phosphate were also evaluated. As measured by yields of variously wheat, oats, barley or clover, the effectiveness of an initial application of superphosphate decreased to about 50% of that of newly applied superphosphate between years 1 and 2, and further decreased to about 20% over subsequent years. At low levels of application, all the rock phosphates were between 10–20% as effective as superphosphate in the year of application for all experiments. Relative to newly applied superphosphate their effectiveness remained approximately constant in subsequent years for two experiments and doubled for the other experiment. The Colwell soil test predicted that the effectiveness of superphosphate decreased to about 45% between years 2 and 3, followed by a more gradual decrease to approximately 15%. At low levels of application, the effectiveness of the rock phosphates as predicted by the Colwell soil test values was initially very low relative to superphosphate (2–30%), and remained low in subsequent years (2–20%). For superphosphate treated soil, the proportion of the added phosphorus extracted generally increased as the level of application increased. By contrast, for rock phosphate treated soil, the proportion of added phosphorus extracted decreased as the level of application increased. For all three experiments there were highly significant positive correlations between amounts of P extracted by the three soil tests. Consequently all soil tests were equally predictive of yield but usually for each soil test separate calibrations between yield and soil test values were required for the different fertilizers and for each combination of fertilizer and plant species and for each year.

    Notes: Abstract  The relative importance of each of three dechlorinating species to overall organochlorine removal from bleached kraft-mill effluents (BKME) was assessed. Ancylobacter aquaticus A7, Pseudomonas P1, and Methylobacterium CP13, strains indigenous to a BKME treatment system, were tested for growth on chlorinated acetic acids and alcohols, and for adsorbable organic halogen (AOX) reduction in batch cultures of sterile BKME from three sources. A. aquaticus A7 exhibited the broadest substrate range, but could only affect significant AOX reduction in softwood effluents. Methylobacterium CP13 exhibited a limited substrate range, but was capable of removing significant amounts of AOX from both hardwood and softwood effluents. By contrast, Pseudomonas sp. P1 exhibited a limited substrate range and poor to negligible reductions in AOX levels from both effluent types. Mixed inocula of all three species combined and inocula of sludge from mill treatment systems removed as much AOX from softwood effluents as did pure populations of Methylobacterium CP13. When BKME was hydrolysed prior to AOX analysis, the subsequent estimates of recalcitrant, or non-hydrolysable, AOX levels were far less variable than their counterpart total AOX measures. It is suggested that this is a relevant and useful measure of AOX for biodegradation studies.

    Notes: Abstract Mechanisms of fatigue were studied in single muscle fibres of the cane toad (Bufo marinus) in which force, intracellular calcium ([Ca2+]i), [Mg2+]i, glycogen and the rapidly releasable Ca2+ from the sarcoplasmic reticulum (SR) were measured. Fatigue was produced by repeated tetani continued until force had fallen to 50%. Two patterns of fatigue in the absence of glucose were studied. In the first fatigue run force fell to 50% in 8–10 min. Fatigue runs were then repeated until force fell to 50% in 〈3 min in the final fatigue run. Addition of extracellular glucose after the final fatigue run prolonged a subsequent fatigue run. In the first fatigue run peak tetanic [Ca2+]i initially increased and then declined and at the time when force had fallen to 50% tetanic [Ca2+]i was 54 ± 5% of initial value. In the final fatigue run force and peak tetanic [Ca2+]i declined more rapidly but to the same level as in first fatigue runs. At the end of the first fatigue run, the rapidly releasable SR Ca2+ store fell to 46 ± 6% of the pre-fatigue value. At the end of the final fatigue run the rapidly releasable SR Ca2+ store was 109 ± 16% of the pre-fatigue value. In unstimulated fibres the nonwashable glycogen content was 176 ± 30 mmol glycosyl units/l fibre. After one fatigue run the glycogen content was 117 ± 17 mmol glycosyl units/l fibre; at the end of the final fatigue run the glycogen content was reduced to 85 ± 9 mmol glycosyl units/l fibre. [Mg2+]i did not change significantly at the end of fatigue in either the first or the final fatigue run suggesting that globally-averaged ATP does not decline substantially in either pattern of fatigue. These results suggest that different mechanisms are involved in the decline of tetanic [Ca2+]i in first compared to final fatigue runs. The SR Ca2+ store is reduced in first fatigue runs; this is not the case for the final fatigue run which is associated with a decline in glycogen and possibly related to either a non-metabolic effect of glycogen or a spatially-localised metabolic decline.

    Notes: Abstract Testing for soil phosphate (P) using the Colwell procedure is widely used in south-western Australia to estimate fertilizer applications required for crops and pastures. The relationship between plant yield, expressed as a percentage of the maximum yield, and soil test values is assumed to be constant in different years for the same soil type and plant species. Data from 11 long-term field experiments in south-western Australia show that regardless of whether percentage of maximum or absolute yield is used, the relationship between yield and soil test values is different (1)in different years, for the same site and where the same P fertilizer type has been used. This occurred irrespective of whether the same or different plant species were grown in different years; (2)where different types of P fertilizer had been used, for the same site, same year and same plant species; (3)for different plant species, for the same site, same year, and same type of P fertilizer. We conclude that considerable errors in the recommendation of fertilizer rates may result from the assumption that there is a constant relationship between soil test and yield.

    Notes: The influence of concentration (mass and volume fraction) and particle morphology on the yielding properties of filamentous broths ofAspergillus niger and Strepto-myces levoris was investigated using the rotating vane technique and compared with those of pulp suspensions. Two methods were applied to determine the volume fraction of the cells growing in filamentous form: the measurement of interstitial volume using a high molecular weight dye or dextran, and the measurement of dewatered broth filter cake volume by displacement using a pycnometer. The latter method provided the most reliable results. Cell concentrations ranging from 3 to 20 g dw/L, with corresponding volume fractions between 0.005 and 0.05, were obtained with broths generated in stirred tank and shake flask fermentations. The yield stress values obtained using the vane technique (0.1 ≤ τy ≤ 37 Pa) correlated with biomass concentration (X, grams dry weight per liter) using the equation τy = aXb with “b” ranging between 2 and 3. Increasing the length of organisms significantly increased the yield stress at a given cell concentration; this morphology effect was described using a “morphology factor” (range 0.016 to 0.11 Pa/[g dw/L]2.5), determined by setting the value of “b” to 2.5. The vane yield strain, which is the critical strain or deformation at the yield point, was independent of cell concentration, but increased with increasing length of organisms. Filamentous fermentation broths with highly branched organisms had a greater yield stress than pulp suspensions. © 1995 John Wiley & Sons, Inc.
