    Notizen: The thermophilic filamentous phototroph Chloroflexus aggregans was able to form a bacterial mat-like dense cell aggregate rapidly. The aggregate formation, which was observed in growing cells in a liquid medium in a bottle, occurred every time within 20–30 min after the cells were dispersed by shaking. The aggregation depended on the energy supplied by photosynthesis or respiration. Cells aggregated most rapidly under temperature and pH conditions that support maximum growth. The aggregation was also accelerated by the addition of 3-isobutyl-1-methylxanthine that inhibits cyclic 3′,5′-AMP phosphodiesterase. Microscopic observation revealed that the bacterium has a fast gliding mobility (1–3 μm s−1). The distinctive cell aggregation of C. aggregans was due to this rapid gliding movement.

    Notizen: Isotachophoretic analysis of ions was performed on guard cells of Vicia faba cv. Ryosai Issun with either open or closed stomata. In guard cells of open stomata, K+ and malic acid concentrations were 5–7 and 5–10 times higher, respectively, than in guard cells of closed stomata. The content of citric acid (plus isocitric acid) also increased during stomatal opening, but the increment was smaller than that of malic acid. Sodium ions, phosphoric and glyceric acids were present in low concentrations but did not increase during the opening. Other cations and anions could not be measured because of low concentrations. Malic acid provided 68–79% of the counter anions for the potassium taken up by guard cells during stomatal opening.

Phylogenetic trees were drawn and analyzed based on the nucleotide sequences of the 1.5-kb gene fragment coding for the L and M subunits of the photochemical reaction center of various purple photosynthetic bacteria. These trees are mostly consistent with phylogenetic trees based on 16S rRNA and soluble cytochrome c, but differ in some significant details. This inconsistency implies horizontal transfer of the genes that code for the photosynthetic apparatus in purple bacteria. Possibilities of similar transfers of photosynthesis genes during the evolution of photosynthesis are discussed especially for the establishment of oxygenic photosynthesis.

    Notizen: Abstract Malate formation in guard cells of Vicia faba leaves is enhanced by light. The action spectrum for this effect was determined for epidermal strips of Vicia faba, and two different spectra were obtained under different light conditions and with and without background irradiation with high-irradiance red light (〉600 nm, 3.0 mW cm−2) superimposed on monochromatic light of other wavelengths. The spectrum obtained at quantum fluxes of 1.7–2.2 nE cm−2 s−1 of monochromatic light without background red light showed a sharp peak at 433 nm with a shoulder around 475 nm and a lower peak at 670–680 nm; the spectrum obtained at much lower quantum fluxes of 0.05–0.07 nE cm−2 s−1 of monochromatic light with red-light background had two peaks of comoparable heights at 380 and 460 nm. The formation of malate with 430-nm blue light was saturated at a quantum flux of 3 nE cm−2 s−1 without the background red light but at a much lower quantum flux of 0.2 nE cm−2 s−1 with the background red light. At this low intensity, blue light was practically ineffective without background red light. A synergistic action of red light presumably absorbed by the chlorophylls, and blue light absorbed by a yellow pigment is thus demonstrated by these experiments. The action maxima at 380 and 460 nm for the blue-light effect in the presence of background red light agree with the absorption maxima of flavins.

    Notizen: Abstract Bacteriochlorophyll(Bchl)-protein complexes were isolated from a strictly aerobic and facultative methylotrophic bacterium Pseudomonas radiora strain MD-1. They were identified as the reaction center (RC)-B870 and the B870 complexes on the basis of their absorption spectra, light induced spectral changes and polypeptide compositions. The RC-B870 complex of this bacterium showed similar properties to those of typical purple photosynthetic bacteria, and contained c-type cytochrome which was oxidized upon illumination.

    Notizen: Abstract The light-harvesting chlorosome antennae of anaerobic, photosynthetic green sulfur bacteria exhibit a highly redox-dependent fluorescence such that the fluorescence intensity decreases under oxidizing conditions. We found that chlorosomes from Chlorobium tepidum contain three isoprenoid quinone species (chlorobiumquinone, menaquinone-7, and an unidentified quinone that probably is a chlorobiumquinone derivative) at a total concentration of approximately 0.1 mol per mol bacteriochlorophyll c. Most of the cellular chlorobiumquinone was found in the chlorosomes and constituted about 70% of the total chlorosome quinone pool. When the quinones were added to artificial, chlorosome-like bacteriochlorophyll c aggregates in an aqueous solution, a high redox dependency of the fluorescence was observed. Chlorobiumquinones were most effective in this respect. A lesser redox dependency of the fluorescence was still observed in the absence of quinones, probably due to another unidentified redox-active component. These results suggest that quinones play a significant, but not exclusive role in controlling the fluorescence and in inhibiting energy transfer in chlorosomes under oxic conditions. Chlorosomes from Chloroflexus aurantiacus contained menaquinone in an amount similar to that of total quinone in Chlorobium tepdium chlorosomes, but did not contain chlorobiumquinones. This may explain the much lower redox-dependent fluorescence observed in Chloroflexus chlorosomes.

    Notizen: Abstract Bacteriochlorophyll(Bchl)-protein complexes were isolated from obligate aerobic bacteria, Erythrobacter longus and Erythrobacter species OCh 114. The apparent molecular weights, absorption spectra and polypeptide compositions of the light-harvesting complexes were, in general, similar to those of the light-harvesting Bchl-protein complexes of purple photosynthetic bacteria. The reaction center complexes of these bacteria also showed similar properties to those of the purple bacteria except for slightly altered polypeptides. However, the following characteristic features of the light-harvesting systems were found in these aerobic bacteria. Major carotenoids were not bound to the Bchl-protein complex in E. longus. In Erythrobacter sp. OCh 114, a new type of Bchl-protein complex which showed a single absorption band in the near infrared region at 806 nm was obtained. The reaction center of strain OCh 114 was associated with a c-type cytochrome.

    Notizen: Abstract The complete carotenoid composition of the thermophilic green sulfur bacterium Chlorobium tepidum strain TNO was determined by spectroscopic methods. Major carotenoids were four kinds of carotenes: γ-carotene, chlorobactene, and their 1′,2′-dihydro derivatives (1′,2′-dihydro-γ-carotene and 1′,2′-dihydrochlorobactene). In lesser amounts, hydroxyl γ-carotene, hydroxyl chlorobactene, and their glucoside fatty acid esters were found. The only esterified fatty acid present was laurate, and OH-chlorobactene glucoside laurate is a novel carotenoid. In other strains of C. tepidum, the same carotenoids were found, but the composition varied from strain to strain. The overall pigment composition in cells of strain TNO was 4 mol carotenoids and 40 mol bacteriochlorophyll c per mol bacteriochlorophyll a. The effects of nicotine on carotenoid biosynthesis in C. tepidum differed from those in the thermophilic green nonsulfur bacterium Chloroflexus aurantiacus.

    Notizen: Abstract About 20 different carotenoids were found in a strictly aerobic photosynthetic bacterium, Erythrobacter longus. All the carotenoids except the highly polar ones were identified as C40-skeletal carotenoids, which could be devided into three groups: (1) bicyclic carotenoids: β-carotene and its hydroxyl derivatives; β-cryptoxanthin, zeaxanthin, caloxanthin and nostoxanthin, (2) monocyclic carotenoids: rubixanthin, bacteriorubixanthin and bacteriorubixanthinal, which was a unique cross-conjugated carotenal, and (3) acyclic carotenoids: anhydrorhodovibrin and spirilloxanthin. Bacteriorubixanthinal and zeaxanthin were the major components. (3R)-3-Hydroxy-β-ionone has rarely been found in carotenoids of purple photosynthetic bacteria, while the acyclic carotenoids have been found exclusively in photosynthetic bacteria. Thus, this bacterium is interesting in its composition of carotenoids.
