    Description: As vertebrate embryos develop to adulthood, their organs undergo marked changes in size and tissue architecture. The heart acquires muscle mass and matures structurally to fulfil increasing circulatory needs, a process that is incompletely understood. Here we used multicolour clonal analysis to define the contributions of individual cardiomyocytes as the zebrafish heart undergoes morphogenesis from a primitive embryonic structure into its complex adult form. We find that the single-cardiomyocyte-thick wall of the juvenile ventricle forms by lateral expansion of several dozen cardiomyocytes into muscle patches of variable sizes and shapes. As juvenile zebrafish mature into adults, this structure becomes fully enveloped by a new lineage of cortical muscle. Adult cortical muscle originates from a small number of cardiomyocytes--an average of approximately eight per animal--that display clonal dominance reminiscent of stem cell populations. Cortical cardiomyocytes initially emerge from internal myofibres that in rare events breach the juvenile ventricular wall, and then expand over the surface.     Description: In acute myeloid leukaemia (AML), the cell of origin, nature and biological consequences of initiating lesions, and order of subsequent mutations remain poorly understood, as AML is typically diagnosed without observation of a pre-leukaemic phase. Here, highly purified haematopoietic stem cells (HSCs), progenitor and mature cell fractions from the blood of AML patients were found to contain recurrent DNMT3A mutations (DNMT3A(mut)) at high allele frequency, but without coincident NPM1 mutations (NPM1c) present in AML blasts. DNMT3A(mut)-bearing HSCs showed a multilineage repopulation advantage over non-mutated HSCs in xenografts, establishing their identity as pre-leukaemic HSCs. Pre-leukaemic HSCs were found in remission samples, indicating that they survive chemotherapy. Therefore DNMT3A(mut) arises early in AML evolution, probably in HSCs, leading to a clonally expanded pool of pre-leukaemic HSCs from which AML evolves.     Notes: An interferometer is presented which is capable of measuring the free-surface velocities and displacements of both specular and diffuse surfaces. The setup utilizes a previously used principle of producing a virtual image of one mirror at the same distance from the photodiode as the second mirror of the interferometer, albeit with considerable simplification. It is shown that use of a He–Ne laser of only 5-mW power can produce high contrast displacement fringes from surfaces of materials with nonuniform microstructure, including composites. Substrates of carbon–carbon composites and polycrystalline alumina with nonuniform microstructure on the scale of 5–10 μm, and with peak velocities up to 150 m/s were considered. An experimental strategy which allows one to covert the optical setup to either a velocity or a displacement interferometer is also discussed. It is further shown that use of a fast photodiode and a high-speed digitizer with a 5-ps rise time provides a time resolution of 0.2 ns for recording the displacement fringes, and allows measuring free surface velocities up to 800 m/s. This is demonstrated by measuring such transient surface velocities with rise times of 1 ns on a specular Si surface. In all the experiments reported here, the surface velocities were produced by the reflection of a stress wave, which in turn was generated on the back surface of the substrate, using a Nd:YAG laser pulse.

    Notes: A system, based on the principle of magnetic induction coupling, to track a particle nonintrusively in three dimensions is presented. It consists of small transmitters mounted inside the particle being tracked, and a set of receiving antennae surrounding the experimental apparatus. In Part I of the sequence of two papers, the focus is on the theoretical aspects, in particular, on developing a computational technique to solve the inverse problem of finding the three-dimensional position as well as orientation of the particle from the voltages induced in the antennae. In Part II the focus is on the actual system development, including all hardware and data acquisition aspects. The results presented here are comprehensive, as they include details of hardware/electronics, comparison of induced voltage model and actual measurements, and a set of actual tracking results. Through test experiments that include a variety of real-time trajectories, this system is tested and its success is demonstrated. Results of real trajectories of a single ball rising in a mass of other spheres in a vibrated bed are also shown. Although this system is intended for use in experimental studies of dry granular flows, it has wide applicability due to its nonintrusive nature, and is particularly useful when optical tracking techniques are not feasible. © 1998 American Institute of Physics.
