Notes: Abstract The focus of hematopoiesis arising after transplantation of a fragment of bone marrow from a C57BL mouse beneath the capsule of the kidney of a syngeneic mouse contains more hematopoietic cells than if transplanted into a semisyngeneic (CBA x C57BL)F1 recipient. Experiments with repopulation of the graft, when depopulated by irradiation, by injected hematopoietic cells of the same genotype as the graft (C57BL) showed that these differences are due to the smaller size of the hemotopoietic microenvironment in the focus formed in the hybrid than in the syngeneic system. Hybrid resistance is thus manifested not only against hematopoietic cells, but also against stromal precursors transferring the hematopoietic microenvironment.

Notes: Abstract A fragment of bone marrow transplanted beneath the capsule of the kidney creates a focus of ectopic hematopoiesis the size of which, measured according to the number of hematopoietic cells, is proportional to the size of the implant. In irradiated recipients the focus was 1.5–2.5 times larger than in intact recipients. Cells creating a focus of heterotopic hematopoiesis in intact and irradiated recipients differed in their radiosensitivity: their D0 values were about 160 and 350 rad, respectively. In this connection it is postulated that two populations of precursor cells participate in the creation of the focus. Their possible relations with predetermined and inducible osteogenic precursor cells are discussed.

Notes: Abstract An irradiated fragment of bone marrow was implanted beneath the capsule of the kidney. Karyologic analysis showed that all hematopoietic cells in the newly formed focus were of recipient origin. Meanwhile, resistance to the bone marrow graft was of donor type. Cells of hematopoietic origin capable of migration, including lymphocytes and macrophages, play no part in the phenomenon of hybrid resistance.

Notes: Abstract Foci of heterotopic hematopoiesis were obtained by transplanting bone marrow of C57BL/6 or (CBAxC57BL)F1 mice beneath the kidney capsule of (CBA/T6T6xC57BL)F1 mice carrying a chromosomal translocation. Cytogenetic analysis of hematopoietic cells from these foci 20–120 days after transplantation showed that only the recipient's hematopoietic cells proliferate in 40% of the grafts, whereas the rest are mosaic and contain on average less than 20% of the donor's cells in both syngeneic and semisyngeneic systems. These characteristics remain stable for at least 4 months. It is concluded that the heterotopic focus is populated in one stage by a very small number (fewer than 10) hematopoietic stem cells. The stability of the clones is evidence that in the dynamic equilibrium of hematopoiesis the exchange of cells between different parts of the hematopoietic system is extremely small or is absent altogether.
