    Notes: The cells of an established mouse fibroblast line, 3T3, have a high plating efficiency and grow rapidly in sparse culture, but stop growing at a very low saturation density in comparison with other lines, because 3T3 is extremely sensitive to contact inhibition of cell division. After each medium change, however, there occurs in a small fraction of the cells in a saturation density culture a series of changes that results in a single rather synchronized division 30 hours later. This is due to a macromolecular substance in the serum which appears to act by reducing the sensitivity of the cells to contact inhibition. The first recognizable event following the addition of serum to a stationary phase culture is a ten fold increase in the rate of RNA synthesis, occurring within 30 minutes. An increase in the rate of protein synthesis follows several hours later. DNA synthesis does not begin before 12 hours, but by two hours after medium change an appreciable fraction of the cells become committed to eventual DNA synthesis and cell division. The sequence of event suggests that regulation of RNA synthesis is the means by which contact inhibition controls cell division.

    Notes: The alpha granules of circulating platelets are dynamic structures that acquire endogenous and exogenous components by synthesis and uptake, respectively. The uptake of exogenous components is a result of either receptor-mediated endocytosis or fluid-phase pinocytosis. Despite many detailed studies on the function and content of α-granules, little is known of the impact of platelet age on these organelles. In this report, we describe the use of platelet biotinylation to identify and isolate aged platelets for the analysis of α-granule contents. When aged platelets were permeabilized and examined by flow cytometry utilizing fluorescently labeled antibodies, two exogenously acquired proteins, fibrinogen and immunoglobulin G, were found to increase significantly with platelet age. The levels of intracellular fibrinogen were found to be elevated relative to control, 114 ± 2% and 119 ± 5% on days 4 and 5 postbiotinylation, respectively; the life span of dog platelets is 6.0 days. Intracellular immunoglobulin G content increased similarly. Levels of two endogenously synthesized proteins, thrombosponding and P-selectin, were not elevated in aged platelets. Confirmation of the flow cytometric data was obtained by isolating aged, biotinylated platelets by fluorescence-activated cell sorting and quantitating the fibrinogen levels with an ELISA assay. For platelets averaging 4.6 days of age, the fibrinogen level was elevated to 128 ± 23% of the level for the entire platelet population. These data demonstrate that age-dependent changes in exogenously acquired α-granule proteins do occur and that the uptake mechanism for these proteins is active through out the platelet life span. © 1994 Wiley-Liss, Inc.

    Notes: The response of Volvox to ultraviolet irradiation was analyzed. Young individuals isolated from a synchronous culture were exposed to UV light (120 J/m2) and subjected to variable lenght periods of dark following irradiation. The major effect of the UV treatment was the inability of the gonidia present in the colonies at the time of irradiation to continue and complete the developmental program. Individuals show a heightened sensitivity to UV for a limited period immediately following inversion and are insensitive at other stages of development. The cytotoxic effect of UV during this interval is completely reversed by the immediate exposure to white light and is increased with longer periods of dark treatment prior to exposure to white light. The temporal profile of the sensitivity defines a smooth curve in which the maximal sensitivity occurs three hours after inversion. The response to higher doses of UV (up to 500 J/m2) is a nonlinear increase in cytotoxicity and is disproportionanately greater in those individuals just prior to the period of maximal sensitivity than those later in development. The results suggest that Volvox has at least two pathways for the repair of UV damage and that one of these, the principal dark repair pathway, is temporarily deficient in the gonidia of young individuals.

    Description: The 'satellite-gauge model' (SGM) technique is described for combining precipitation estimates from microwave satellite data, infrared satellite data, rain gauge analyses, and numerical weather prediction models into improved estimates of global precipitation. Throughout, monthly estimates on a 2.5 degrees x 2.5 degrees lat-long grid are employed. First, a multisatellite product is developed using a combination of low-orbit microwave and geosynchronous-orbit infrared data in the latitude range 40 degrees N - 40 degrees S (the adjusted geosynchronous precipitation index) and low-orbit microwave data alone at higher latitudes. Then the rain gauge analysis is brougth in, weighting each field by its inverse relative error variance to produce a nearly global, observationally based precipitation estimate. To produce a complete global estimate, the numerical model results are used to fill data voids in the combined satellite-gauge estimate. Our sequential approach to combining estimates allows a user to select the multisatellite estimate, the satellite-gauge estimate, or the full SGM estimate (observationally based estimates plus the model information). The primary limitation in the method is imperfections in the estimation of relative error for the individual fields. The SGM results for one year of data (July 1987 to June 1988) show important differences from the individual estimates, including model estimates as well as climatological estimates. In general, the SGM results are drier in the subtropics than the model and climatological results, reflecting the relatively dry microwave estimates that dominate the SGM in oceanic regions.

    Description: Climatologies of convective precipitation were derived from passive microwave observations from the Special Sensor Microwave Imager using a scattering-based algorithm of Adler et al. Data were aggregated over periods of 3-5 months using data from 4 to 5 years. Data were also stratified by satellite overpass times (primarily 06 00 and 18 00 local time). Four regions (Mexico, Amazonia, western Africa, and the western equatorial Pacific Ocean (TOGA COARE area) were chosen for their meteorological interest and relative paucity of conventional observations. The strong diurnal variation over Mexico and the southern United States was the most striking aspect of the climatologies. Pronounced morning maxima occured offshore, often in concativities in the coastline, the result of the increased convergence caused by the coastline shape. The major feature of the evening rain field was a linear-shaped maximum along the western slope of the Sierra Madre Occidental. Topography exerted a strong control on the rainfall in other areas, particularly near the Nicaragua/Honduras border and in Guatemala, where maxima in excess of 700 mm/month were located adjacent to local maxima in terrain. The correlation between the estimates and monthly gage data over the southern United States was low (0.45), due mainly to poor temporal sampling in any month and an inadequate sampling of the diurnal cycle. Over the Amazon Basin the differences in morning versus evening rainfall were complex, with an alternating series of morning/evening maxima aligned southwest to northeast from the Andes to the northeast Brazilian coast. A real extent of rainfall in Amazonia was slightly higher in the evening, but a maximum in morning precipitation was found on the Amazon River just east of Manaus. Precipitation over the water in the intertropical convergence zone (ITCZ) north of Brazil was more pronounced in the morning, and a pronounced land-/sea-breeze circulation was found along the northeast coast of Brazil. Inter-comparison of four years revealed 1992 to be the driest over Amazonia, with about a 23% decrease in mean rate compared to the 4-year mean estimated rain rate.

    Description: The story of the Aircraft to Satellite Data Relay (ASDAR) program began when airline meteorologists realized that B-747's and other commercial jets provided cockpit displays of digital values for outside air temperature and winds. Later, when a few B-747's were used to carry portable air quality monitoring equipment for the Global Air Sampling Program (GASP), scientists at NASA-Lewis explored ways in which these digital values could be used to label data collected during the GASP flights. Digital values of GASP analyses were recorded along with digital values of location and altitude, time, winds, and temperature, obtained by microprocessors from within the host aircraft's avionics. These data suggested a way in which manually recorded in-flight meteorological reports could be replaced by an automatic system, which could record winds and air temperatures as often as desired. NASA's prototype ASDAR showed that automated data relay by meteorological geostationary satellites could be accomplished from an aircraft. Testing of the instruments and analyses of its data are examined.
