    Notizen: Abstract Lemna minor fronds transferred to a sterile culture medium containing 50% (v/v) deuterium oxide (2H2O) rapidly undergo a loss of soluble protein with a corresponding increase in free amino acids. The loss of protein is due to two factors: (i) the inhibition of protein synthesis for 4 h followed by a slower rate of synthesis than normal, (ii) a rapid 9–10 fold increase in protein degradation. In plants grown for longer periods (3–6 days) in 50% 2H2O medium, protein synthesis is inhibited by 20% and the rate constant of degradation is 2–3 times that measured in fronds growing in normal (H2O containing) complete medium. The initial loss of protein is not due to the breakdown of any specific protein fraction. Investigation of several enzymes indicates that all proteins are catabolised in response to 2H2O treatment. The implications of these results with regard to the interpretation of density-labelling experiments are discussed.

    Notizen: Abstract Transfer of Lemna minor fronds to culture medium containing 50% (v/v) deuterium oxide induces a large increase in the rate of protein breakdown, which is not due to an increase in the activity of acidic or neutral proteolytic enzymes or peptidases. Biochemical and ultrastructural evidence indicates that deuterium oxide affects the properties of certain membranes, particularly the tonoplast, and allows vacuolar proteolytic enzymes to pass into the cytoplasm and cause the increased protein breakdown.

    Notizen: The thermodynamical parameters (free energy, enthalpy, and entropy) of complex formation between ethidium bromide and single-stranded and double-stranded tetranucleotides of different base sequence [5′-d(TpGpCpA), 5′-d(ApCpGpT), and 5′-d(ApGpCpT) have been determined from the temperature dependencies of 500 MHz proton nmr chemical shifts. The analysis enables the contributions to be differentiated for the formation of different types of complexes (1:1, 2:1, 1:2, and 2:2) in aqueous solution. The results have been interpreted in terms of the main types of intermolecular interactions responsible for formation of the different complexes; van der Waals and electrostatic interactions are important for formation of complexes of ethidium bromide with single-stranded tetranucleotides, whereas van der Waals and hydrophobic interactions play a significant role in the binding of the dye to the tetramer duplexes. © 1997 John Wiley & Sons, Inc.     Notizen: Phosphorous, as sodium dihydrogen phosphate, and nitrogen, in the form of a urea-for-maldehyde condensate, were applied to cotton fabric. The flame-retarding character of each treated fabric was assessed by determining the respective limiting oxygen index. Analysis of the results showed that at the lowest levels of applied phosphorous (P = 2%), the presence of nitrogen produced less than an additive combined flame-retarding effect. At higher phosphorous levels, this deviation from additivity increased until at P 〉 6%, the presence of nitrogen antagonized the flame-retarding effect of the phosphorous. These observations are discussed within the current ideas of nitrogen-phosphorous interaction in flame-retardant systems, for cotton-containing fabrics.

Die Polymerisationsgeschwindigkeit erwies sich als direkt abhängig von der Stromstärke, der Zinkbromid-Konzentration und dem anfänglichen Wassergehalt des Solvens.Die Molekulargewichte der entstandenen Polymeren (1000-6000) sind unabhängig von der Stromstärke, der Zinkbromid-Konzentration und dem Umsetzungsgrad des Monomeren zum Polymeren, werden jedoch beeinflußt durch Wasser, die Monomerkonzentration und das Elektrodenmaterial. Die chemische Zusammensetzung der Polymeren ändert sich nicht mit der Stromstärke, der Zinkbromid-Konzentration, dem Umsetzungsgrad und dem Elektrodenmaterial. Sie ändert sich jedoch ein wenig mit dem Wassergehalt des Acetons.
    Notizen: The electroinitiated polymerization of N-vinylcarbazole (VCZ) in acetone solvent at 25°C with zinc bromide, as catalyst, and platinum electrodes was investigated in detail. The polymerization rate was found to be directly dependent on current strength, zinc bromide concentration and the initial water content of the solvent.The molecular weights of the formed polymers (1000-6000) are independent of current strength, zinc bromide concentration and degree of conversion of monomer to polymer, but are affected by water, monomer concentration, and by the type of electrodes used. The chemical composition of the polymer is invariant with current strength, zinc bromide concentration, degree of conversion, and electrode material but does vary slightly with the water content of acetone.

    Notizen: Complexation of the trypanocidal drug, ethidium bromide (EB), and the self-complementary deoxytetraribonucleoside triphosphates, 5′-d(ApCpGpT), 5′-d(ApGpCpT), and 5′-d(TpGpCpA), in aqueous salt solution has been investigated using one-dimensional and two-dimensional 500/600 MHz 1H-nmr spectroscopy. Six hundred megahertz two-dimensional homonuclear 1H-nmr spectroscopy (nuclear Overhauser effect spectroscopy) was used for a qualitative determination of the structures of EB binding with the deoxytetranucleotides. Concentration dependencies of proton chemical shifts of the molecules have been measured at constant temperatures (T = 303 or 308 K). Different successive schemes of complex formation between the dye molecule and the tetranucleotides have been examined by taking into account various molecular associations in solution, viz., 1:1, 1:2, 2:1 and 2:2 complexes. Equilibrium reaction constants and the limiting proton chemical shifts in the complexes have been determined. The relative contributions of different types of complexes in the equilibrium mixture have been determined and special features of the dynamic equilibrium have been revealed by analysis of chemical shifts as a function of both the dye and tetranucleotide concentrations. The present analysis leads to the conclusion that EB binds preferentially to the pyrimidine-purine sites of the tetranucleotide duplexes. The results show that the energy of EB binding depends on the base content in the pyrimidine-purine sites of the tetramers and on the nucleotide residuals flanking the preferential site. The most favorable structures of the 1:2 and 2:2 complexes of the dye with the tetranucleotides have been constructed using calculated values of induced chemical shifts of EB protons in conjunction with intermolecular nuclear Overhauser effects. The structures of the EB:tetranucleotide complexes depend on tetramer base sequence and are characterized by differences in helix parameters. © 1996 John Wiley & Sons, Inc.

    Notizen: The light-induced degradation of aqueous hydroxypropylmethylcellulose (HPMC; D.S. 1.9) solutions has been investigated under a variety of conditions. The HPMC was sensitized to short-wavelength radiation (〈500 nm) by the addition of sodium nitrite, the degradation being accompanied by increased acidity and the generation of oxides of nitrogen. Addition of eosin dye increased the sensitivity to visible light (〉500 nm), and the effect was considerably more pronounced when strong electron donors were added. Analysis of the shear-viscosity relationship of the solutions, during the degradation, indicated that both chain disentanglement and chain scission occurred but that the primary process was the depolymerization of the cellulose chain. It is proposed that both the dye- and nitrite-induced depolymerizations are a consequence of peroxide attack on the β-glucosidic ether link. The results indicate that, in the case of sodium nitrite addition, the peroxide was generated by the aqueous photolytic decomposition of the nitrite ion. The peroxide formation in the eosin - TEA system was a consequence of leuco-dye production, under the influence of the light, followed by the reduction of traces of dissolved oxygen by this activated dye.
