    Notes: Summary New methods have been applied to the determination of single copy DNA sequence differences between the sea urchin speciesStrongylocentrotus purpuratus, S. franciscanus, S. drobachiensis, andLytechinus pictus. The thermal stability of interspecies DNA duplexes was measured in a solvent (2.4 M tetraethylammonium chloride) that suppresses the effect of base composition on melting temperature. The lengths of duplexes were measured after digestion with S1 nuclease and correction made for the effect of length on thermal stability. The degree of base substitution that has occurred in the single copy DNA during sea urchin evolution is significantly larger than indicated by earlier measurements. We estimate that 19% of the nucleotides of the single copy DNA are different in the genomes of the two sea urchin congeners,S. purpuratus, andS. franciscanus, which apparently diverged only 15 to 20 million years ago.

    Notes: Summary A comparison has been made between the Sp88 gene regions of the DNAs of the sea urchinsStrongylocentrotus purpuratus (Sp.) andStrongylocentrotus drobachiensis (Sd.). Examination of the 3′ terminal part of the transcribed region revealed a short repetitive sequence present in Sd. but absent from Sp. A 12-nucleotide sequence present once in Sp. is almost perfectly duplicated at both ends of the repeat in Sd., suggesting that a mobile repeat was inserted in the Sd. genome. Other members of this family of repeated sequences occur in many interspersed locations in the genomes of both species. Except for the insertion duplication, the inserted sequence lacks direct or reverse repeats.

    Notes: Summary Cells in fetal adipose tissue and cells in vitro are characterized by rapid proliferation. Serum factors have been shown to be important for the rapid proliferation of cells in vitro. The present experiment was performed to determine if neuroendocrine regulatory mechanisms of the fetus can influence the actions of serum factors on preadipocyte proliferation and differentiation in vitro. Sera were obtained from decapitated fetal pigs and intact littermates during gestation. Sera were tested for their effects on primary cultures of preadipocytes and stromalvascular cells derived from inguinal adipose tissue of young Sprague-Dawley rats. Coverslip cultures were used for histochemical analysis of enzymes after 12 days of incubation with test media. Analysis of growth curves produced from sequential [3H]-thymidine labeling indicated that fetal age influences rates of proliferation. Sera from decapitated fetal pigs specifically reduced the number of proliferating preadipocytes in culture. Sera from decapitated fetal pigs induced a minimum of 50% less differentiation of sn-glycerol-3-phosphate dehydrogenase activity than sera from intact pigs at all fetal ages. Histochemical staining for enzymes of differentiating preadipocytes was also reduced in cultures incubated with sera from decapitated fetal pigs in comparison to sera from intact pigs. The present study has demonstrated that the in vivo effect of decapitation on fetal adipose tissue development is a consequence of alterations in systemic factors present in serum in response to removal of central regulation by the hypothalamic-pituitary axis.

    Notes: Summary This experiment was conducted to determine if serum factors are responsible for differences in cellularity of prenatal and postnatal pig adipose tissue as determined by in vitro measurement of cellular proliferation and enzyme-histochemical metabolic development. Cellular proliferation of stromal-vascular cells derived from rat inguinal adipose tissue was measured by [3H]-thymidine incorporation. Coverslip cultures were used for analysis of histochemical differentiation. Cells were incubated in media containing 10% fetal bovine, fetal pig, mature pig, or various combinations of these sera. Fetal bovine serum promoted more [3H]-thymidine incorporation than fetal or postnatal pig sera. Fetal pig sera also stimulated more [3H]-thymidine incorporation than mature pig sera. Sera from adult pigs promoted differentiation and lipid filling of adipocytes. Fetal pig sera stimulated histochemical expression of enzymes, but did not induce lipid filling. Fetal bovine serum produced histochemically undifferentiated cells. Addition of fetal bovine serum to media containing mature pig sera reduced lipid accumulation and histochemical reactivity of cells. This effect of fetal serum was thus due to specific inhibition of lipid deposition and not substrate restriction. These experiments demonstrated that serum factors have a major influence on morphological development of fetal and postnatal adipose tissue.

    Description / Table of Contents: Hochreine ferritische CrMo-Stähle - Fünf erfolgreiche Jahre für die Korrosionsbe-kämpfung in der VerfahrensindustrieIm Laufe von 5 Jahren hat der ferritische nichtrostende Stahl E-Brite 26-1 eine beträchtliche Bedeutung innerhalb der für Chemieanlagen infrage kommenden Werk-stoffe erreicht. Als nichtrostende Stähle ist er hinsichtlich seiner Anwendbarkeit mit viel kostspieligeren Legierungen vergleichbar. Der Stahl hat sich als wirtschaftliche Alternative für Anlagen erwiesen, die mit Alkalien, Salpetersäure-Harnstoff, organischen Chemikalien, Zellstofflaugen, Raffinerieprodukten u. v. a. in Kontakt kommen. Das Vertrauen der Verbraucher in die Zuverlässigkeit dieses Stahls ist so stark, daß heute auch große kritische Anlagen auf Routinebasis aus diesem Stahl verlangt werden. Aufgrund der guten Aufnahme des neuen Materials auf dem Markt hat man versucht, Ersatzversionen zu entwickeln. Diese Nachahmungen sind jedoch mit Vorsicht zu betrachten. Ein stabilisierter 26-IS muß noch längere Zeit untersucht werden, um festzustellen, ob er hinsichtlich Korrosionsbeständigkeit, Duktilität, Verarbeitbarkeit und Reproduzierbarkeit der Eigenschaften mit E-Brite 26-1 vergleichbar sein kann.
    Notes: In five years, E-BRITE 26-1 ferritic stainless steel has won an important place in the spectrum of materials suitable for use in chemical process equipment. It provides, in stainless steel, performance-capability characteristics comparable to more expensive alloys. It has demonstrated cost-effectiveness in equipment used for caustic, nitric-urea, organic chemicals, pulping liquors, refinery streams, and elsewhere. User confidence in the reliability and integrity of Grade XM 27 has increased to the point where large critical systems are now routinely specified in the alloy. The market acceptance of this material has attracted attempts to produce substitute versions of the alloy. Imitation, should be viewed with caution. Stabilized 26-IS must be examined over a lengthy period of time to determine if its own corrosion resistance, ductility, fabricability, and reproducibility properties could ever be likened to those of E-BRITE 26-1.

    Notes: The thermal decomposition of normal alkanes and linear poly(α-olefins) was found to occur predominately by random scissions, followed by volatilization into an open system. To show these processes the fragment distribution from high molecular weight linear polyethylene (fragments to C55), n-alkanes, polypropylene, and polyisobutylene (fragments to C60) are reported and discussed in depth. Based on the results, a mechanism of random scissions followed by volatilization is postulated for polyethylene and n-alkanes as well as for polyisobutylene and polypropylene. Polystyrene is shown not to follow the same mechanism, for little evidence for fragments above trimers was found. In addition, temperature rise-time measurements at the pyrolyzer probe are reported and explained.

    Notes: Flash pyrolyses at 400, 650, and 900°C of polyisopropenylcyclohexane (PIC) and copolymers, produced by partial hydrogenation of poly-α-methylstyrene (PAMS), have shown that the decomposition of the PIC or IC sequences is complex. In comparison, PAMS and AMS sequences give only depolymerization to monomer at the two lower temperatures but a more complex degradation at 900°C, based on the products identified. It is interesting to note that traces of decalin solvent initiated the depolymerization of PAMS and the AMS sequences in the copolymers at 400°C. The products identified by copolymer pyrolyses indicated that no reaction occurred between S and IC units during decomposition. Average sequence data on the copolymers could not be determined because of the facile unzipping of the AMS sequences and the greater thermal stability of the IC sequences. The fragmentation pattern did indicate that partial hydrogenation of PAMS had produced random copolymer and not a blend of two homopolymers.

    Description: A laser-induced iodine fluorescence technique that is suitable for the planar measurement of temperature in cold nonreacting compressible air flows is investigated analytically and demonstrated in a known flow field. The technique is based on the temperature dependence of the broadband fluorescence from iodine excited by the 514-nm line of an argon-ion laser. Temperatures ranging from 165 to 245 K were measured in the calibration flow field. This technique makes complete, spatially resolved surveys of temperature practical in highly three-dimensional, low-temperature compressible flows.

    Description: A technique is described for imaging the injectant mole-fraction distribution in nonreacting compressible mixing flow fields. Planar fluorescence from iodine, seeded into air, is induced by a broadband argon-ion laser and collected using an intensified charge-injection-device array camera. The technique eliminates the thermodynamic dependence of the iodine fluorescence in the compressible flow field by taking the ratio of two images collected with identical thermodynamic flow conditions but different iodine seeding conditions.

    Description: A unified laser-induced fluorescence technique for conducting planar measurements of temperature, pressure and velocity in nonreacting, highly compressible flows has been developed, validated and demonstrated. Planar fluorescence from iodine, seeded into air, was induced by an argon-ion laser and collected using a liquid-nitrogen cooled CCD camera. In the measurement technique, temperature is determined from the fluorescence induced with the laser operated broad band. Pressure and velocity are determined from the shape and position of the fluorescence excitation spectrum which is measured with the laser operated narrow band. The measurement approach described herein provides a means of obtaining accurate, spatially-complete maps of the primary flow field parameters in a wide variety of cold supersonic and transonic flows.
