    Notes: Abstract A highly sensitive immunoassay has been used for the detection of a major storage protein, legumin, in embryos of Pisum sativum L.; with this technique nanogram quantities could be measured. In the two varieties tested, legumin could be detected in embryos in vivo, when they had attained a fresh weight of 2·10-3 g and 3·10-3 g, respectively. Contrary to earlier claims, embryos cultured in vitro were shown to be capable of initiating legumin synthesis. This capacity to initiate legumin synthesis was confirmed by two-dimensional isoelectric focusing-electrophoresis and fluorography; embryos harvested before initiation of legumin synthesis and cultured in radioactive medium were shown to have synthesized legumin subunits. The amounts of legumin and total protein synthesized per unit fresh weight were consistently greater in vitro than in equivalent embryos grown in vivo.

    Notes: Summary Pollen of Pisum sativum was exposed to doses of 900 to 6,000 r of X-rays prior to pollinating a multiply marked genotype. The first generation progeny closely resembled that produced with unirradiated pollen. In the second generation, five loci were monitored, and the results showed that irradiation enhanced the proportion of maternal information transmitted to the progeny; the practical implications of the data, as well as the mechanism underlying the effect are discussed.

    Notes: Summary The organization of three classes of legumin gene has been studied by exploiting restriction fragment length polymorphisms. Hybridization of cDNA probes, representing different sub-families of legumin gene, to DNA from F2 and F6 plants from selected crosses showed that there was a tight linkage of individual genes within each of two sub-families. One of these sub-families has been mapped to a locus on chromosome 7 close to r, while the other maps to a locus near a on chromosome 1.     Notes: Summary The cotyledon cells ofPisum sativum have high DNA contents. By appropriate culture techniques, some of these cells can be triggered into division. Two types of dividing nuclei were seen. Firstly those that were polyploid with metaphases containing chromosome numbers ranging in value from 4 x to 32 x. Included among these were unexpected numbers equivalent to 12 x and 14 x. Secondly there were cells containing giant polytene chromosomes and these progressed from prophase to a metaphase where the polytene chromosomes separated into constituent single chromosomes.

    Notes: The thermodynamical parameters (free energy, enthalpy, and entropy) of complex formation between ethidium bromide and single-stranded and double-stranded tetranucleotides of different base sequence [5′-d(TpGpCpA), 5′-d(ApCpGpT), and 5′-d(ApGpCpT) have been determined from the temperature dependencies of 500 MHz proton nmr chemical shifts. The analysis enables the contributions to be differentiated for the formation of different types of complexes (1:1, 2:1, 1:2, and 2:2) in aqueous solution. The results have been interpreted in terms of the main types of intermolecular interactions responsible for formation of the different complexes; van der Waals and electrostatic interactions are important for formation of complexes of ethidium bromide with single-stranded tetranucleotides, whereas van der Waals and hydrophobic interactions play a significant role in the binding of the dye to the tetramer duplexes. © 1997 John Wiley & Sons, Inc.     Notes: Phosphorous, as sodium dihydrogen phosphate, and nitrogen, in the form of a urea-for-maldehyde condensate, were applied to cotton fabric. The flame-retarding character of each treated fabric was assessed by determining the respective limiting oxygen index. Analysis of the results showed that at the lowest levels of applied phosphorous (P = 2%), the presence of nitrogen produced less than an additive combined flame-retarding effect. At higher phosphorous levels, this deviation from additivity increased until at P 〉 6%, the presence of nitrogen antagonized the flame-retarding effect of the phosphorous. These observations are discussed within the current ideas of nitrogen-phosphorous interaction in flame-retardant systems, for cotton-containing fabrics.
