    Description: Protein kinase B (PKB) is activated in response to phosphoinositide 3-kinases and their lipid products phosphatidylinositol 3,4, 5-trisphosphate [PtdIns(3,4,5)P3] and PtdIns(3,4)P2 in the signaling pathways used by a wide variety of growth factors, antigens, and inflammatory stimuli. PKB is a direct target of these lipids, but this regulation is complex. The lipids can bind to the pleckstrin homologous domain of PKB, causing its translocation to the membrane, and also enable upstream, Thr308-directed kinases to phosphorylate and activate PKB. Four isoforms of these PKB kinases were purified from sheep brain. They bound PtdIns(3,4,5)P3 and associated with lipid vesicles containing it.     Description: Polymerization of actin filaments directed by the actin-related protein (Arp)2/3 complex supports many types of cellular movements. However, questions remain regarding the relative contributions of Arp2/3 complex versus other mechanisms of actin filament nucleation to processes such as path finding by neuronal growth cones; this is because of the lack of simple methods to inhibit Arp2/3 complex reversibly in living cells. Here we describe two classes of small molecules that bind to different sites on the Arp2/3 complex and inhibit its ability to nucleate actin filaments. CK-0944636 binds between Arp2 and Arp3, where it appears to block movement of Arp2 and Arp3 into their active conformation. CK-0993548 inserts into the hydrophobic core of Arp3 and alters its conformation. Both classes of compounds inhibit formation of actin filament comet tails by Listeria and podosomes by monocytes.     Description: Cytokine production plays a critical role in diseases caused by Kaposi's sarcoma-associated herpesvirus (KSHV). Here we show that a latent KSHV gene product, kaposin B, increases the expression of cytokines by blocking the degradation of their messenger RNAs (mRNAs). Cytokine transcripts are normally unstable because they contain AU-rich elements (AREs) in their 3' noncoding regions that target them for degradation. Kaposin B reverses this instability by binding to and activating the kinase MK2, a target of the p38 mitogen-activated protein kinase signaling pathway and a known inhibitor of ARE-mRNA decay.     Description: Activation of the Raf and extracellular signal-regulated kinases (ERKs) (or mitogen-activated protein kinases) are key events in mitogenic signalling, but little is known about interactions with other signaling pathways. Agents that raise levels of intracellular cyclic adenosine 3',5'-monophosphate (cAMP) blocked DNA synthesis and signal transduction in Rat1 cells exposed to epidermal growth factor (EGF) or lysophosphatidic acid. In the case of EGF, receptor tyrosine kinase activity and association with the signaling molecules Grb2 and Shc were unaffected by cAMP. Likewise, EGF-dependent accumulation of the guanosine 5'-triphosphate-bound form of Ras was unaffected. In contrast, activation of Raf-1 and ERK kinases was inhibited.     Description: Caspase-8 has two opposing biological functions--it promotes cell death by triggering the extrinsic pathway of apoptosis, but also has a survival activity, as it is required for embryonic development, T-lymphocyte activation, and resistance to necrosis induced by tumour necrosis factor-alpha (TNF-alpha) and related family ligands. Here we show that development of caspase-8-deficient mice is completely rescued by ablation of receptor interacting protein kinase-3 (RIPK3). Adult animals lacking both caspase-8 and RIPK3 display a progressive lymphoaccumulative disease resembling that seen with defects in CD95 or CD95-ligand (also known as FAS and FASLG, respectively), and resist the lethal effects of CD95 ligation in vivo.     Description: To identify proteins that may participate in the activation of the protein kinase Raf, proteins that interact with Raf were selected in a two-hybrid screen. Two members of the 14-3-3 protein family were isolated that interacted with both the amino terminal regulatory regions of Raf and the kinase domain of Raf, but did not compete with the guanine nucleotide-binding protein Ras for binding to Raf. 14-3-3 proteins associated with Raf in mammalian cells and accompanied Raf to the membrane in the presence of activated Ras. In yeast cells expressing Raf and MEK, mammalian 14-3-3 beta or 14-3-3 zeta activated Raf to a similar extent as did expression of Ras. Therefore, 14-3-3 proteins may participate in or be required for the regulation of Raf function.     Description: The Experimental Cloud Lidar Pilot Study (ECLIPS) was initiated to obtain statistics on cloud-base height, extinction, optical depth, cloud brokenness, and surface fluxes. Two observational phases have taken place, in October-December 1989 and April-July 1991, with intensive 30-day periods being selected within the two time intervals. Data are being archived at NASA Langley Research Center and, once there, are readily available to the international scientific community. This article describes the scale of the study in terms of its international involvement and in the range of data being recorded. Lidar observations of cloud height and backscatter coefficient have been taken from a number of ground-based stations spread around the globe. Solar shortwave and infrared longwave fluxes and infrared beam radiance have been measured at the surface wherever possible. The observations have been tailored to occur around the overpass times of the NOAA weather satellites. This article describes in some detail the various retrieval methods used to obtain results on cloud-base height, extinction coefficient, and infrared emittance, paying particular attention to the uncertainties involved.

    Description: Data collected by the Stratospheric Aerosol and Gas Experiment II are presented, showing annual variations of water vapor in the stratosphere and the upper troposphere. The altitude-time cross sections of water vapor were found to exhibit annually repeatable patterns in both hemispheres, with a yearly minimum in water vapor appearing in both hemispheres at about the same time, supporting the concept of a common source for stratospheric dry air. A linear regression analysis was applied to the three-year data set to elucidate global values and variations of water vapor ratio.

    Description: The global distributions of upper tropospheric relative humidity derived from the archived SAGE II water vapor data set are presented. For both summer and winter months, vertical profiles of zonal mean relative humidity are derived for each of the six 20-deg latitude bands covering 60 deg N - 60 deg S. Some examples of global maps of upper tropospheric relative humidity are shown to illustrate the relationship between moist areas and convective activity.
