    Notes: Six different types of materials including PVC, chitosan, chitin, agarose, Sepharose, and Trisacryl were evaluated for their lipase-coupling efficiencies. Among those tested, chitosan yielded the highest amount of lipase (79 mg/mL packed gel) immobilized but with lowest oil hydrolytic activity (0.03 mg eq/mL gel). The amount of lipase immobilized was affected by the length of the hydrocarbon chain attached to the PVC matrix but not by the pore size of the supports used. On the other hand, the specific activity of the immobilized lipase was affected by the pore size but not by the chain length of the hydrocarbon attached to the support. After immobilization, the optimal reaction pH was shifted from 7.5 to 8.5 and the optimal reaction temperature from 35 to 45-55°C. Lipase immobilized on PVC exhibited higher thermal stability than that on agarose. The half-life of the PVC immobilized lipase operating at 30°C in a packed-bed reactor was estimated to be about 400 h.

    Notes: Poly(vinylamine) was synthesized and used to replace poly-L-lysine in forming microcapsule with alginate. Test results indicated that capsules with good mechanical strength and permeability could be obtained under the controlled treatment conditions of poly(vinylamine) and alginate. Application of the current microcapsular system to cell culture was demonstrated by the usage of erythropoietin- (EPO-) producing IW32 mouse erythroleukemia cells. The encapsulated IW32 cells grew to a density of 8 × 107 cells/mL, two times that found in the corresponding poly-L-lysine/alginate capsules. The EPO accumulation inside the microcapsule with the current encapsulation system was also higher. A concentration of 7.3 U/mL was attained as compared to 4.3 U/mL in the poly-L-lysine/alginate microcapsule. © 1992 John Wiley & Sons, Inc.

    Notes: A dramatic change of the reaction rate was observed for the lipase-catalyzed hyrolysis of tributyrin in a batch reactor. Immediately after the addition of the enzyme, the lipolysis rate increased continuously until a maximal reaction rate was reached. The duration of the induction was mainly controlled by the bulk enzyme concentration and the reactor stirring speed. The reaction rate dropped sharply after reaching its maximal value. The lipolysis decayed at a rate of about 0.012 min-1, and was not affected by changes of the stirring speed. This decay was attributed to the fast deactivation of the surface-adsorbed lipase, and possibly to the extremely slow desorption of the inactivated species. For reaction time longer than 120 minutes, the lipolysis decreased at a much slower rate. Several mechanisms for the decay of the lipolysis rate were discussed.

    Notes: The gram-positive bacteria, Acinetobacter calcoaceticus, is capable of accumulating biopolymer in the carrier matrix of an immobilized cell system. Several possible mechanisms for the biopolymer accumulation are evaluated. It appears that direct solid surface polymer adsorption and polymer diffusion limitation within the pore space are minor factors in biopolymer accumulation. Calculations demonstrate that the cell bound polymer to dry cell weight ratio is much higher for immobilized cells than for free cells. The higher cell-bound polymer to dry-cell-weight ratio for immobilized cells as well as the accumulation of the immobilized cells in the Celite matrix are believed to be the main factors for biopolymer accumulation in the Carrier matrix. Further studies reveal that the cell-bound polymer to dry-cell-weight ratio is strongly affected by shear forces. At zero shear stress, such as would be present in the carrier matrix, cell bound polymer to dry cell weight ratio can be as high as 1.6. As the shear stress increases, this ratio decreases. When shear stress increases above 5 dyn/cm2, a level equivalent to the shear experienced by free cells in a stirred tank fermentation, cell-bound polymer decreases to less than 20% of dry cell weight. A macroscopic model is developed to describe the effect of shear stress on the cell-bound polymer to dry-cell-weight ratio.

    Notes: Despite the importance of biomass as a parameter in fermentation processes, there are no commercially available sensors suitable for its measurement. An indirect approach for the assessment of biomass concentration can be based on material balances and on the direct monitoring of fermentation parameters for which there are established sensors (e.g., gaseous oxygen and carbon dioxide). As a consequence, this method requires no assumption of cellular yield coefficients or rate constants. This approach is also readily adaptable to general use since it requires only some knowledge of the compositions of the substrate, cells, and noncellular products.

    Notes: The economics of yeast production depend heavily upon the cellular yield coefficient on the carbon source and the volumetric productivity of the process. The application of an on-line computer to maximize these two terms during the fermentation requires a continuous method of measuring cell density and growth rate. U fortunately, a direct sensor for biomass concentration suitable for use in industrial fermentations is not available. Material balancing, with the aid of on-line computer monitoring, offers an indirect method of measurement. Laboratory results from baker's yeast production in a 14-liter fermentor (with a PDP-11/10 computer for on-line analyses) show this indirect measurement technique to be a viable alternative. From the oxygen uptake and carbon dioxide production data, gas flow rate, and ammonia addition rate, the cell density during the fermentation has been estimated and found to compare well with actual fermentation data.

    Notes: The Biotechnology and Bioengineering study presented here was undertaken to demonstrate the usefulness of computer control for the production of yeast from molasses. A flexible control system was developed by using an on-line computer for the monitoring of cell mass and employing anticipatory control to maintain the maximum productivity. Process disturbances were minimized by employing a multivariable feedback control system to prevent ethanol formation. The control strategy acted to keep overall conversion yield at its maximum level, about 0.5 g cell/ g sugar, while maintaining high volumetric productivity between 3 and 5 g/liter-hr. Results are presented to show the effectiveness of simultaneous anticipatory and feedback contol in overcoming problems of oxygen starvation, molasses quality, and variable inoculum size.
