    Description: Vitellogenic stage carp ovaries were collected aseptically and the cells of ovarian follicles were dispersed by trypsin treatment and cultivated as monolayer. Control cultures were grown in medium M199 alone. Media for experimental cultures were supplemented either with 100 µl ml-1 of CPH and CPS-carp GnRH (cultivated pituitary cell secretion affected by 10-6, 10-8, 10-10 M carp GnRH). The direct effect of these factors on 17-α-Hydroxy progesterone (17α -OHP) and 17β-Estradiol (E2) secretion of isolated follicular cells in cell culture was assayed by appropriate Radioimmunoassay (RIA). According to the results the E2 was a main hormone secreted by cultivated carp granulosa and theca cells. Adding 100 µl ml-1of 10-6 M CPS-carp GnRH had no significant effect on cultivated granulosa and theca cells endocrine activity (P〉 0.05). On the other hand, carp granulosa and theca cells treated with the lower contents of CPS-carp GnRH (10-8, 10 -10 M) showed a 4.02 and 3.74-fold increase in E2 concentration and 4.27 and 3.64-fold increase in 17α -OHP level, which was significantly higher than CPH (P〈 0.05). Also, there was a significant increase of steroid secretion in group stimulated with 100 µl ml-1 of CPS and CPH, but there were no significant differences between them (P〉 0.05).

    Description: In this research, morphological and histomorphometrical structure of testis of 20 silver male carp fish were studied in two classes or groups. Group1 was composed of 10 fish with average (±SD) weight of 1.247+0.656kg and average(±SD) length of 43.675+1.414cm with about 2 years age, Group2 was composed of 10 fish with average(±SD) weight of 5.716+0.519kg and average(±SD) length of 81.5+1.643cm. Average (±SD) weight of testis were 2.34+1.47gr and 83.33+25.81gr with average (±SD) GSI of 0.187+0.224 and 1.457+4.974 in groups 1 and 2 respectively. Samples from testis were taken by maximum thickness of 0.5cm and after fixation in bouin , s fixative and 5-6µm thickness section were made routine paraffin embedding method and stained by Hematoxylin-Eosin and PAS staining. The microscopic results showed that the silver carp testis was lobular and cystic type in two groups. In group 1, there was no spermatozoon activity and PGCs were only germ cells in the cysts. But in group2, the numbers of PGCs were decreased significantly and spermatogenic cells were seen in different phases including spermatogonia, primary and secondary spermatocysts, early and late spermatid, and spermatozoa which each one was located in a separated cyst. There was no significant difference in nucleus diameter of PGCs in testis of group1 (6.97+0.438µ) and group (6.13+0.438µ). In group2, the nucleolus diameter of spermatogonia was 2.97+0.112µm, primary spermatocyt 3.59+0.107µ, early spermatid 1.59+0.761µ, late spermatid 1.24+0.132µ, spermatozoa 1.16+0.054µ, and the length of spermatozoia 17.412+1.946µ. The interesting finding was immature testis in fish of group 1 with average weigh (1.247+0.656kg) and average length (43.675+1.414cm) in about 2 years age and mature testis in fish of group 2 with average weight of (5.716+0.519kg) and average length of (81.5+1.643cm) with about 4 years age in Khuzestan climate conditions.

    Description: White spot syndrome virus (WSSV) is a pathogen that causes high mortality in shrimp culture in the whole world. Sequence analysis of WSSV has shown similarity of WSSV isolates in different countries with exception of a few variable genomic loci. This study investigated the sequence variation of some Iranian WSSV isolates and previously identified isolates. Samples were collected during target surveillance and were feed, broodstock, post-larvae, artemia, crabs, and wild and cultured shrimp of northern Persian Gulf (Boushehr and Khuzestan provinces). The open reading frame (ORF) 94 sequence of different Iranian WSSV isolates were amplified using specific primers from positive samples. The ORFs 94 sequence of positive samples were sequenced and registered in the Gene Bank and then compared to other WSSV isolates. The number of repeat units in ORF94 showed that WSSV isolates were varied in number. There are SNPs (G and T) in position 48 of RUs that varies in different Boushehr and Khuzestan isolates. Also these sequences were compared to Gene Bank WSSV isolates and showed a high similarity (〉90%) to Southeast Asian countries. To our knowledge this is the first report of sequence analysis in Iranian WSSV isolates applications.

    Description: Seminal plasma has contained organics factors same as proteins and cholesterol. The information about role and variety of fish seminal plasma and relationship with other factors especially in common carp is rare. The aim of this study was evaluation of effects of total protein and cholesterol on physical and biochemical factors of cultural common carp milt in Khouzestan province. For this study 40 fish have been bought (20 in March and 20 in April) and examinations of physical and biochemical were tested. Base on the results, total protein in March and April was 0.29±0.14 and 0.37±0.35 g/dl respectively, cholesterol in March and April was 9/95±8.83 and 14/12±12.73 mg/dl respectively. The total protein and cholesterol in April compared to March increased non significantly. The ions of calcium, chloride, sodium and potassium in April compared to March increased. Based on the results there was significant and positive correlation between total protein and Albumin, cholesterol and K in march and significant and negative correlation with Cl but total protein in April had significant and positive with Cl, P, spermatocrite, cholesterol and Albumin and negative and significant correlation with weight of fish. Cholesterol in March had significant and positive with albumin and negative significant correlation with Cl and P but cholesterol in April had positive significant correlation with spermatocrite, Albumin, P and Cl. According to the data of this study, total protein, cholesterol, calcium, chloride, sodium, potassium of the Common carp seminal plasma increased in April compared to March. So because of these factors have effects on fertility, quality and quantity of common carp sperm so it is possible to adjust the parameters of protein, cholesterol and minerals of dietary in spawning season increase the quality of common carp sperm.

    Description: This study was conducted to identify intestinal helminthes parasites. The helminthes fauna is collected from body cavity and intestines of 108 specimens of Brachirus orientalis which were obtained from Abadan port (waterfront) and investigated during the years 2009-2010. Most of the 4 species of helminthes were found in the intestine and body cavity of the examined fishes. The helminthes found was composed of 2 digenetic Trematodes (Lepocreadioides zebrine, Allocreadium brachirusii) all adults, in the intestine. The general prevalence of Lepocreadioides zebrini and Allocreadium sp. was in the intestine, 58% and 73% respectively, nematode Hysterothylacium aduncum with a prevalence of 23% was the only cestod Protocephalus sp. with 16%, except Pterobothrium sp., which was reported previously. Most parasite species found in this study have been reported for the first time from Iran and Allocreadium sp. is introduced as a new species and described here.

    Description: To study viability and activity of isolated common carp, Cyprinus carpio, ovarian follicular cells (granulosa and theca cells), 17-α-Hydroxy progesterone (17α-OHP) and 17β-Estradiol (E2) levels were estimated in the culture media of cultivated carp ovarian follicular cells, using radioimmunoassay (RIA). Oocytes were isolated from the ovaries of female carp. Interstitial tissue was manually removed in order to obtain single oocytes surrounded only by the follicular envelope. Such a preparation was trypsinized at room temperature. Follicular cell suspension contained both cells and cell clumps. It was the mixture of theca (T) and granulosa (G) cells. The cells were grown as monolayer in 24-well microplates in M199 medium supplemented with FBS. Culture media were analyzed for estrogen and progesterone content by appropriate radioimmunoassay. Trypsinization of ovarian follicles resulted in the formation of the cell suspension which contained a mixture of G and T cells. The follicular cells attached to glass and grew well during culture period. E2 was the main steroid hormone secreted by cultivated cells. Estrogen secretion increased by 415.52 ± 25 pg/ml at the first 3 days up to 530.25 ± 55.8 pg/ml on day 5 and it didn't change significantly until the end of the experiment. 17α-OHP secretion, however, gradually increased from 23.84 ± 8.2 pg/ml at the beginning of culture up to 35.76 ± 5.4 pg/ml at the end of cultivating. As the result of the present study the fish follicular cells grown in tissue culture were steroidogenically active cells as expressed by the secretion of E2 and P4 and the E2 was a dominant hormone secreted by isolated follicular cells, which it correlated closely with vitellogenes stage.

    Description: The present research aimed to determine acute toxicity and evaluate the effect of sub-lethal concentrations of diazinon on some haematological and immunological parameters of Barbus sharpeyi by the Organization for Economic Cooperation and Development methods. Juvenile B. sharpeyi were exposed to 0, 5%, 10% and 25% 96h LC50 concentrations of diazinon for 21 days. Blood samples were taken after 7, 14 and 21 days of exposure to diazinon. Haematological and biochemical parameters including: RBC, Hb, PCV and WBC, as well as serum enzymes were evaluated. Immunological indices and resistance against bacterial infection, were determined. Results showed that 96h LC50 of diazinon in B. sharpeyi was estimated at 3.987 mg/L. Diazinon toxicity at a level of 1 and 0.4 mg/L decreased RBC, Hb, and Hematocrite, in almost all sampling periods compared. WBC and globular index decreased significantly in fish exposed to 1 mg/L diazinon on days 14 and 21(p〈0.05). Dose dependent increase in serum enzymes were seen in fish exposed to diazinon. Diazinon toxicity showed no effect on serum LDH level (p〉0.05). Dose dependent decrease in serum total protein and globulin were indicated in diazinon exposed fish. Serum lysozyme and bactericidal activity decreased in T3 and T4 at days 14 and 21. Mortality following challenge to A. hydrophila increased in fish exposed to 1 mg/l diazinon. It can be concluded that diazinon is toxic to B. sharpeyi. Therefore, the strict biosecurity should be taken into consideration when this pesticide is used in agricultural fields surrounding freshwater sources of fish cultivation.

The first group was the control group put in dechlorinated tap water and four respectively in the next group were kept with salinity 4ppt, 8ppt, 12ppt, 16ppt in the same condition. In days (1, 3, 7, 14, 21, 28) samples were collected from skin of head, ventral and lateral lines. Sections with 5- 6µm thickness were made and stained with PAS and H&E methods. In histometrical survey, epiderm thickness, numbering of club cells and goblet cells in 200 µm of epiderm length in any area, was investigated comparatively. The results showed that in different salinity concentrations, skin of head, ventral and lateral lines were influenced and changes in epiderm thickness, reduce the number of club cells and increase the number of goblet cells were demonstrated. These changes in different concentrations for each parameter, for significantly increased (p〈0.05).
