    Notes: The combination of supercritical fluid extraction, high resolution capillary supercritical fluid chromatography, and Fourier transform infrared microspectrometry is described for the separation and identification of polycyclic aromatic hydrocarbons in a coal tar pitch. The variable solvating power of the supercritical fluid was utilized to selectively fractionate the sample. The fluid extract was decompressed through a frit restrictor into the sample cavity of a cooled microvalve injector, where the analytes were deposited and concentrated for subsequent chromatographic analysis. Several of the analytes separated in the chromatograph were collected on a potassium bromide disc at a solvent elimination inter-face for subsequent infrared analysis involving the use of an infra-red microscope accessory. The spectra obtained show the power of this detection technique for distinguishing between isomers.

    Notes: Certain of the “meta-stable peaks” in mass spectra are abnormally wide. This extra width is shown to be associated with release of kinetic energy during fragmentation. Methods are presented for measuring this energy release and values are given for various transitions in the spectra of aromatic nitro compounds.

    Notes: We have measured the sputtering yields of ammonium chloride and of frozen glycerol films under 8 keV Ar+· bombardment using a thin film depth profiling technique. The sputtering yield of ammonium chloride was 34±3 molecules (NH4Cl)/ion; for frozen glycerol the sputter yield was 54±9 molecules/ion. The results constrain possible mechanisms of damage suppression in organic secondary ion mass spectrometry.

    Notes: A strategy for sequencing peptides using the mono- and dilithiated quasimolecular ions is proposed. Low energy collision-induced dissociation (CID) of the monolithiated species produced primarily [An+Li—H]+ and [Bn+H+OLi]+ ions while that of the dilithiated species produced [An+Li—H]+ and [Yn+Li+H]+ ions. The precursor parent ion in the monolithiated adduct is proposed to be a mixture of lithiated neutral peptide (1) and protonated lithium salt (2). Mechanisms for the formation of the [An+Li—H]+ and [Yn+Li+H]+ ions are proposed. Four peptides of known composition but unknown sequence were successfully sequenced using the combined data from the CID spectra of the mono- and dilithiated species.

    Notes: Mass Measurement accuracy of better than 0.015% on protonated gramicidin S (nominal chemical average molecular weight 1141.5 Da) and its first three isotopes has been achieved using reasonant ion ejection in the quadrupole ion-trap mass spectrometer operated at unit resolution. Analyte and Calibrant ions are subjected to identical injection and trapping conditions when desorbed by Cs+ ions from a split prone-tip and co-injected into the trap. When the mass analysis scan rate is slowed, enhanced resolution is achieved for all ions but the mass range accessed in a single scan of the digital-to-analog converter, which controls the mass analysis scan, is limited making calibration difficult. A peak-matching procedure is described for use at these higher resolution conditions in which the offset voltage which determines the origin of the RF amplitude ramp is adjusted in such a fashion that the analyte and calibrant ions occur at the same point in the mass analysis scan. This procedure gave an improved mass measurement accuracy of better than 0.007% for the protonated gramicidin S. ions.

    Notes: A method has been developed for the rapid and sensitive mass spectrometric characterization of peptides. The approach uses bioreactive mass spectrometer probe tips, incorporating covalently bound enzymes, which are capable of modifying biomolecules for analytical purposes. In the demonstrated cases, enzymatic proteolysis is initiated upon application of analyte to the probe tips, time is allowed for digestion, and the products are analyzed using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. The probe tips have been used for proteolytic mapping and partial sequence determination of picomole quantities of peptide. Analysis times were approximately 30 min. Two methods of database search were utilized. The first used limited peptide sequence information and parent molecular weight, while the second used exclusively the molecular weights of a number of endoproteolytic fragments. A simple method of comparing the match of experimental data for a tryptic digest with the results of a search is described.

    Notes: A method has been developed using reliable GC derivatization techniques interfaced with a high resolution mass spectrometer. The method has proved successful for the detection of low levels (less than 1 ppm) of clenbuterol in complex biological matrices. Selected ion recording of two characteristic isotopic fragment ions provides a specific mode of detection by verifying the GC retention time of these ions and also by comparing their relative abundance. Analysis of urine samples demands higher mass spectrometric resolution, and 40000 (10% valley) was found to be a prerequisite for accurate integration of the drug-related chromatographic peaks. The method developed is suitable for adaptation to a completely unattended automated routine incorporating sample injection, storage and retrieval of source tuning parameters, and data processing.

    Notes: An azurin, a small respiratory copper protein from the bacterium Pseudomonas fluorescens biotype G, has been studied by mass spectrometry to determine sequence information. The study of homologously related proteins by mass spectiometry is particularly attractive, since the correct nature of major parts of the deduced sequences can be confirmed by comparison with the sequences of the protein from related organisms. An oxidized tryptohan residue has been identified amongst the products from a cyanogen bromide digest of this wild type azurin. In the same digest, a product is also found to arise from cleavage of the peptide chain at the C-terminal side of the same tryptophan residue. These results are rationalized.

    Notes: It has been shown that there is no significant carrier effect when low levels of [3H]octopamine (100 pg) are extracted by ethyl acetate from aqueous solutions containing a 103 or 106 times excess of unlabelled octopamine. It is also shown that there is no carrier effect when low levels of octopamine as the tetrakis-TMS derivative are injected with 100 times the amount of the [2H3] analogue on an SE 30 column.
