    Notes: Abstract X-ray and γ-radiation techniques for evaluating bone density under field conditions in cattle and sheep were investigated. The best results were obtained from measurements of the density of radiographs of tibial tarsal bones of sheep and from the absorption of γ-radiation of the tibial tarsal bones of sheep and the fibular tarsal bones of cattle. The γ-radiation techniques may be carried out rapidly under field conditions (up to 30 animals an hour) and bone density can be estimated with a standard error of 0.042–0.046 SG units.

    Notes: Abstract The specific gravities (SG) of bone samples taken from various parts of the skeleton of cattle and sheep were determined gravimetrically in an effort to establish which sites give the most reproducible and uniform results, so that between animals, or sequential within animals, comparisons may be made with maximum sensitivity. Samples obtained from the mandible of sheep and the rib of cattle and sheep were found to be too variable to be useful for most purposes. Best results were obtained using whole bones which are easily prepared, such as the tibial tarsal bones of cattle and sheep and the fibular tarsal bone of cattle. These bones gave within animal deviations of 0.012–0.024 SG units, when comparing left bone with right bone. Slightly higher values were obtained for coccygeal vertebrae from cattle and distal metacarpal condyles from sheep. SG was related to ash, calcium and phosphorus contents of coccygeal vertebrae of cattle and it is considered that, for survey work on the mineral status of the bones of cattle and sheep, the correlations are sufficiently high to make their determination unnecessary. This will enable radiation determined SG techniques to be applied to this type of work.

    Notes: A carefully Standardized technique is described for the preparation of glass capillary columns which can be used successfully for routine quantitative amino acid analysis. Comparison is made between two different modes of sample injection. Preliminary quantitative results from “split” injection and “on-column” injection techniques are evaluated statistically and it is concluded that the “on-column” system is a prerequisite for quantitative amino acid analysis by glass capillary gas chromatography. An analysis of fish muscle protein hydrolyzate illustrates an application of this technique and results are compared with those from a packed column analysis.

    Notes: A new procedure, requiring expired sample volumes of approximately 2L, has been developed for the quantitative determination of n-pentane in human breath. The methodology capitalizes on the superior resolving power and separating efficiency of the open tubular capillary column and incorporates an effective prechromatographic cryofocusing technique. Molecular sieve is used to simultaneously remove moisture and carbon dioxide from breath samples which are “spiked” with an internal standard to optimize the validity of analysis. Reproducible data for both internal standard spiking (CV = 5.8%) and sample n-pentane level (CV = 3.2%) have been obtained, together with a mean recovery of 104%. Preliminary data show n-pentane levels in normal female subjects (n = 10) to range from 0.62 to 3.16 nmole/L (mean 1.76 nmole/L) and in males (n = 10) from 3.20 to 8.76 nmole/L (mean 5.66 nmole/L).

    Notes: A procedure is described in which a wide bore capillary column is used as an alternative to the more traditional packed column for the quantitative analysis of amino acids as their N-heptafluorobutyryl isobutyl ester (HBB) derivatives. The column, installed in a gas chromatograph previously configured for use with a packed column, is shown to give good reproducibility by repeated determination of amino acid response factors (RSD values for all amino acids are below 3%). A number of problems, encountered during the use of this column, are discussed and suitable techniques to overcome them are reported.

    Notes: A procedure is described in which a wide bore glass capillary column is used as an alternative to the more traditional packed column in the analysis of amino acid levels in plasma. The coefficients of variation for all amino acids (with the exception of aspartic acid) were better than 11% with recoveries ranging from 81% to 122%. The data are compared with the corresponding results obtained using a packed column and show significant differences (p 〈 0.005) between values for glycine, serine, isoleucine, proline, methionine, aspartic acid, phenylalanine, and lysine. A similar comparison between results from the wide bore and the fused-silica open tubular (FSOT) column shows better agreement. Adjustment of chromatographic conditions for the wide bore analysis yields results in good agreement with those from FSOT analysis but which still differe significantly from the backed column data.
