    Notes: Abstract Gibberellic acid (GA3) and abscisic acid (ABA) regulate α-amylase production in aleurone tissue of the barley (Hordeum vulgare L.) grain during germination. Since α-amylase is a Ca2+-containing protein that is synthesized on the rough endoplasmic reticulum (ER), we investigated the possibility that GA3 and ABA regulate the flux of Ca2+ into the ER of aleurone cells. Membrane vesicles were isolated from GA3- and ABA-treated aleurone layers to characterize the effect of these hormones on calcium transport (CaT) activity as measured by the uptake of 45CaCl2 in vitro. We found that ABA and GA3 greatly alter the relative amounts of CaT activity in ER and tonoplast (TP) membrane fractions. Treatment with GA3 results in CaT being associated primarily with the ER, whereas treatment with ABA results in roughly equal amounts of CaT in ER and TP. Moreover, we found that ABA prevents or reverses the GA3-induced increase in CaT in the ER. The effects of ABA and GA3 on CaT in the ER require several hours to develop, and the effects of these hormones on CaT cannot be brought about by adding the hormones to membranes in vitro. In order to determine whether the in-vitro measurements of CaT reflect changes that occur in vivo we measured the Ca2+ content of isolated ER, measured Ca2+ transport into aleurone layers, and examined differences in the ability of ER proteins from GA- and ABA-treated layers to bind Ca2+. Our results indicate that GA3 increases and ABA decreases both Ca2+ flux into the ER and the amount of calcium that accumulates in the ER of barley aleurone cells in vivo.

    Notes: Abstract The steady-state levels of Ca2+ within the endoplasmic reticulum (ER) and the transport of 45Ca2+ into isolated ER of barley (Hordeum vulgare L. cv. Himalaya) aleurone layers were studied. The Ca2+-sensitive dye indo-1. Endoplasmic reticulum was isolated and purified from indo-1-loaded protoplasts, and the Ca2+ level in the ER was measured using the Ca2+-sensitive dye indo-1. Endoplasmic reticulum was isolated and purified from indo-1-loaded protoplasts, and the Ca2+ level in the lumen of the ER was determined by the fluorescence-ratio method to be at least 3 μM. Transport of 45Ca2+ into the ER was studied in microsomal fractions isolated from aleurone layers incubated in the presence and absence of gibberellic acid (GA3) and Ca2+. Isopycinic sucrose density gradient centrifugation of microsomal fractions isolated from aleurone layers or protoplasts separates ER from tonoplast and plasma membranes but not from the Golgi apparatus. Transport of 45Ca2+ occurs primarily in the microsomal fraction enriched in ER and Golgi. Using monensin and heat-shock treatments to discriminate between uptake into the ER and Golgi, we established that 45Ca2+ transport was into the ER. The sensitivity of 45Ca2+ transport to inhibitors and the Km of 45Ca2+ uptake for ATP and Ca2+ transport in the microsomal fraction of barley aleurone cells. The rate of 45Ca2+ transport is stimulated several-fold by treatment with GA3. This effect of GA3 is mediated principally by an effect on the activity of the Ca2+ transporter rather than on the amount of ER.

    Notes: Abstract There are nearly 200 species of aquatic vertebrates, almost exclusively fishes, in the Brahmaputra River System. This faunal composition includes not only a wide variety of food-fishes, but also about 50 varieties of aquarium fishes. The most spectacular animal in the Brahmaputra is undoubtedly the river dolphin, Platanista gangetica. The population of many species, particularly of the dolphin, is in steady decline. River dolphins, mostly juveniles, often entangle themselves in gill and drift nets, while feeding on trapped fishes. Proper implementation of Indian Fisheries Act, especially a total ban on destructive nets and on killing of brooders and juveniles by explosives and poisoning should be strictly implemented. Poor communication facilities in remote riverbank areas and lack of funds are two important reasons for the slow progress of the conservation of river dolphin and this endangered species is now on the verge of extinction. The anthropogenic activities and their impact on the habitat of river dolphin are discussed.

    Notes: Abstract A model experiment on soil conservation utilising the plantation of certain herbaceous plants in a selected stretch of Majuli, the largest riverine island in the world, was carried out. The plantation followed work to establish the proper inclination for the river bank. The soil erosion in the experimental site was reduced to only about 2% compared with about 15–20% in the previous years. All the four species selected for the present study had excellent soil binding capacity and were thus helpful in stabilizing the highly vulnerable soil in the flood plain areas of the region.

    Description: The cellular machinery promoting phagocytosis of corpses of apoptotic cells is well conserved from worms to mammals. An important component is the Caenorhabditis elegans engulfment receptor CED-1 (ref. 1) and its Drosophila orthologue, Draper. The CED-1/Draper signalling pathway is also essential for the phagocytosis of other types of 'modified self' including necrotic cells, developmentally pruned axons and dendrites, and axons undergoing Wallerian degeneration. Here we show that Drosophila Shark, a non-receptor tyrosine kinase similar to mammalian Syk and Zap-70, binds Draper through an immunoreceptor tyrosine-based activation motif (ITAM) in the Draper intracellular domain. We show that Shark activity is essential for Draper-mediated signalling events in vivo, including the recruitment of glial membranes to severed axons and the phagocytosis of axonal debris and neuronal cell corpses by glia. We also show that the Src family kinase (SFK) Src42A can markedly increase Draper phosphorylation and is essential for glial phagocytic activity. We propose that ligand-dependent Draper receptor activation initiates the Src42A-dependent tyrosine phosphorylation of Draper, the association of Shark and the activation of the Draper pathway. These Draper-Src42A-Shark interactions are strikingly similar to mammalian immunoreceptor-SFK-Syk signalling events in mammalian myeloid and lymphoid cells.     Description: The immune system plays an important role in regulating tumor growth and metastasis. Classical monocytes promote tumorigenesis and cancer metastasis, but how nonclassical "patrolling" monocytes (PMo) interact with tumors is unknown. Here we show that PMo are enriched in the microvasculature of the lung and reduce tumor metastasis to lung in multiple mouse metastatic tumor models. Nr4a1-deficient mice, which specifically lack PMo, showed increased lung metastasis in vivo. Transfer of Nr4a1-proficient PMo into Nr4a1-deficient mice prevented tumor invasion in the lung. PMo established early interactions with metastasizing tumor cells, scavenged tumor material from the lung vasculature, and promoted natural killer cell recruitment and activation.     Description: Bromodeoxyuridine (BrdUrd) treatment of the prolactin nonproducing subclone of GH cells (rat pituitary tumor cells) induces amplification of a 20-kilobase DNA fragment including all of the prolactin gene coding sequences. This amplified DNA segment, which is flanked by two unamplified regions, thus designates a unit of BrdUrd-induced amplified sequence. Cloned DNA segments, 10.3 kilobases long, from the 5' end of the rat prolactin gene of BrdUrd-responsive and -nonresponsive cells, were ligated to the thymidine kinase gene of herpes simplex virus type 1 (HSV1TK), and the hybrid DNA was transferred to thymidine kinase-deficient mouse fibroblast cells by transfection. The HSV1TK gene and the rat prolactin gene were amplified together in drug-treated transfectants carrying the hybrid DNA HSV1TK gene and rat prolactin gene of BrdUrd-responsive GH cells.
