    Notes: Abstract Patients suffering from rheumatoid arthritis (RA) may experience a temporary reduction of disease symptoms during pregnancy. As indicated by the occurrence of RA-disease symptoms during pregnancy, three categories of patients were defined, namely, remission, relapse and unchanged. In all three categories changes in the plasma level and glycosylation of α1-acid glycoprotein (AGP) were determined longitudinally in comparison to those occurring in pregnancy of healthy women. In healthy pregnancy, we observed: (i) a peak in the plasma concentration at week 18 and a minimum at week 30; (ii) a continuous increase in the degree of branching of the glycans during the entire pregnancy period, and (iii) a decrease in the degree of α3-fucosylation of AGP-glycans with a minimum occurring at week 25. Comparable pregnancy-induced changes in glycosylation were found for two other acute-phase proteins α1-protease inhibitor (PI) and α1-antichymotrypsin (ACT). Increased oestrogen levels, known to occur during pregnancy, may be one of the factors that induce these changes, because the increased branching and decreased α3-fucosylation is in agreement with our earlier findings regarding an involvement of this hormone in the regulation of acute phase protein glycosylation in oestrogen-treated males as well as females. In all three clinical categories in RA, pregnancy also induced a continuous increase in the degree of branching of the glycans of AGP. However, similar changes in concentration and fucosylation were only found during remission of the disease symptoms. In the relapse and unchanged categories in RA, the degree of fucosylation and the plasma concentration of AGP remained constant throughout pregnancy. This indicates a relationship between changes in α3-fucosylation of AGP and RA disease activity.

    Notes: The synthesis of new photolabeling analogues of luteinizing hormone releasing hormone is described. Peptides were prepared by the solid-phase method, and the photolabeling moiety was introduced as the 4′-nitrophenyl-alanine precursor. This amino acid, either of the L or D configuration, was placed at different positions and was modified afterwards to 4′-azidophenylalanine. Structures with agonistic or antagonistic character were prepared, together with a peptide containing simultaneously the photolabeling moiety and a biotin substituent for future receptor isolations. Binding studies on placental and pituitary membranes indicate that these compounds represent promising candidates for receptor labeling studies.

    Description / Table of Contents: Boron Imides from the Thermal Decomposition of Dialkyl[sily(silyloxy)amino]boranesDialkyl[trimethylsilyl)(trimethylsilyloxy)amino]boranes R2B—N(OSiMe3SiMe3 (1a-c), prepared from (Me3Si)2N—OSiMe3 and R2BCl, can be thermolyzed in the liquid phase at 70°C to form (Me3SI)2O and a mixture of R2B—NR—Br—N(OSiMe3)SiMe3 (2a-c) and (RBNR)3 (3a-c). In the presence of an excess of BR′3, 1b, c can be transformed into (Me3Si)2O and diborylamines RR′B—NR—BR′2 (4b), c, (Me3Si)2O and a mixture of (RBNR)3 and polymer material are formed from the gas-phase thermolysis of 1b, c at 270°C The polymer (EtBNEt)n depolymerizes to the borazine 3b above 150°C. An intermediate from the gas-phase thermolysis of 1b can be trapped by liquid 1c, yielding 2c, A. conclusive explanation of all these observation is possible by the assumption that boron imides RB—NR(5a-c) occur as intermediates.
    Notes: Die aus (Me3Si)2N-OSiMe3 und R2BCl zugänglichen Dialkyl[trimethylsilyl)(trimethylsilyloxy)amino]borane R2B—N(OSiMe3)SiMe3 (1a-c zerfallen in flüssiger Phase bei 70°C unter Abspaltung von (Me3Si)2O in ein Gemisch aus R2B—NR—BR—N(OSiMe3)SiMe3 (2a-c) und (RBNR)3 (3a-c). Im Überschuß von BR′3 erhält man aus 1b, c neben (Me3Si)2O die Diborylamine RR′B—NR—BR′2 (4b-c). In der Gasphase entstehen bei 270°C aus 1b, c (Me3Si)2O und ein Gemisch aus (RBNR)3 und Polymeren. Das Polymere (EtBNEt)n depolymerisiert oberhalb 150°C zu 3b. Flüssiges 1c regiert mit einer aus 1b durch Gasphasenthermolyse gebildeten Zwischenstufe zum Abfangprodukt 2c′. Einer geschlossenen Deutung aller Befunde liegt die Annahme von Borimiden RB—NR (5a-c) als Zwischenstufen zugrunde.

    Notes: An efficient synthesis of HR 916 B (4), the orally active 1-(RS)-(pivaloyloxy)ethyl prodrug ester of the cephalosporin cefdaloxime, was developed and applied on a multi-kg scale. AMCA (8) was prepared by exchange of the acetoxy group of fermentation product ACA (7) with the nucleophile methanol under acidic conditions. Its 7-amino group was acylated with mixed anhydride 14 to give 15. Carboxylic acid 15 was esterified with iodohydrin ester 27 or bromohydrin ester 30, respectively, to provide the acylal 16. Simultaneous removal of both the amino- and the oximo-protecting group furnished the prodrug HR 916 3, which was purified and stabilized by precipitation of its tosylate salt 4. The overall yield of 4 (ratio 5/6 = 0.65) was 39% relative to AMCA (8) (four steps), 15% relative to ACA (7) (five steps).

    Notes: A number of acute phase proteins were determined by electroimmunoassay in media from CBA mouse hepatocytes cultured for 2 days with human recombinant IFN β2/IL-6, as well as with conditioned media from LPS-stimulated rat macrophages, and of murine L fibroblasts. It was found that human recombinant IL-6 caused three-fold increase in secretion of fibrinogen, while haptoglobin, complement C3 and transferrin were increased respectively, to 168 per cent, 151 per cent, and 145 per cent of the control. DEX(10-7M) in DMEM supplemented with 5 per cent FCS, enhanced the IL-6 effect on the three positive acute phase proteins. IL-6 elevated haptoglobin mRNA in mouse hepatocytes to a degree comparable with the concentration of the protein in the culture medium. The effect of conditioned media from murine fibroblasts and peritoneal rat macrophages was generally similar to that of recombinant IL-6. However, both natural preparations of the cytokines caused decrease in albumin and alpha-1-proteinase inhibitor secretion.

    Notes: A model system for studies of mechanisms governing the alterations of glycosylation of plasma glycoproteins was developed. The system employs two human hepatoma cell lines, Hep 3B and Hep G2, as target cells and agarose affinity electrophoresis with lectins for studies of microheterogeneity of α1-protease inhibitor (PI), a model glycoprotein synthesized by hepatocytes. As an example for the application of the system, the effect of cytokines on major microheterogeneity of plasma proteins is demonstrated. The results indicate that interleukin 6, transforming growth factor β1 and, to some extent, tumor necrosis factor α are directly involved in regulating the pattern of glycosylation of plasma proteins in vitro, but the major effect is obtained by using combinations of interleukin 6, transforming growth factor β1, tumor necrosis factor a and interleukin 1. In addition, the results underline the dissociation between alteration of gene expression and the changes in the pattern of plasma protein glycosylation.

    Notes: Affinity electrophoresis, with concanavalin A and Lens culinaris agglutinin as ligands, was applied to study the microheterogeneity of serum proteins, with special emphasis on α1-fetoprotein and α1-acid glycoprotein, two proteins of potential clinical value. A total of 602 samples of serum from patients with various neoplastic and inflammatory conditions were evaluated. Affinity electrophoresis provided useful information for differential diagnosis of cancers of various origins including hepatomas, metastatic liver cancers and yolk sac tumors. The method also proved indispensable for the detection of intercurrent infection in patients with rheumatoid arthritis, systemic lupus erythematosus and scaled burns.
