    Notes: The effects of mixing on the critical mean holding time for washout and the steady state performance of growth processes in continuous flow reactors are investigated. Macromixing, micromixing, and cell recycle arc considered. The tanks-in-series model composed of N completely mixed flow reactors, the dispersion model, the plug flow model, and a combined model composed of a plug flow reactor and a continuous stirred tank flow reactor connected in series arc used to represent the macro-mixing or residence time distribution. The extreme cases of micromixing, namely, complete segregation and maximum mixedness, as well as intermediate states of micromixing are investigated to determine their effects on washout and the occurence of multiple steady states. A technique for predicting the maximum mixedness washout condition from a knowledge of the residence time distribution is presented and used to determine the washout condition for the dispersion model under maximum mixedness conditions.

    Notes: A gram-negative bacterium strongly lytic toward living cells of the food yeast Saccharomyces fragilis was isolated by continuous-flow enrichment from compost. The organism was identified as a species of Arthrobacter. The extracellular lytic enzyme complex produced by this bacterium contained β-1,3-glucanase, mannan mannohydrolase, and proteolytic activities. The polysaccharases were inducible by whole yeast cells. In chemostat cultures on chemically defined media, synthesis of the polysaccharases was very slight and only detectable at dilution rates below 0.02 hr-1. Enzyme production in defined media was not solely dependent on growth rate but also was influenced by the growth limiting substrate and the culture history. The production of individual depolymerases and of the lytic activity was studied in batch and chemostat cultures containing yeast as the limiting substrate. The maximum specific growth rate of the Arthrobacter under these conditions was 0.22 hr-1. β-1,3-Glucanase and proteolytic activities were synthesized by exponentially growing bacteria but maximum lytic titers did not develop until the specific growth rate was declining, at which time mannan mannohydrolase syntheses was induced. In yeast limited chemostats polysaccharase syntheses were greatest at the lowest dilution rates examined, namely 0.02 hr-1. Further optimization of enzyme production was achieved by feeding the Arthrobacter culture to a second-stage chemostat. A comparison of lytic enzyme productivities in batch and chemostat cultures has been made.

    Description / Table of Contents: Microstructure and Tempering Behaviour of Evaporated resp. Thermally Sprayed ZrO2-Layers on Metallic SubstratesBecause of their application in gas turbines plasmasprayed thermal barrier coatings of partially stabilized zirconia on metallic substrates are an example for the technical use of ceramic-metal-composites. Problems especially concerning the interface adhesion are still partly not understood. In this paper several systems are examined by electron microscopy, which all consist of ceramic zirconia layers evaporated or thermally sprayed on a metallic substrate. Priority is given to structural analysis. An attempt is made to lighten the interface morphology by the method of cross section preparing, because thereby the possibility of aimed preparation is given.
    Notes: Durch ihren Einsatz in Gasturbinen sind plasmagespritzte Wärmedämmschichten aus teilstabilisierten Zirkoniumdioxid auf metallischen Substraten ein Beispiel für einen in der Praxis eingesetzten Schichtverbundwerkstoff aus Keramik und Metall. Die damit verbundenen Probleme vor allem im Bereich der Grenzflächenhaftung sind immer noch teilweise unverstanden [1,2,3]. In der vorliegenden Arbeit werden verschiedene systeme elektronenmikroskopisch untersucht, bei denen eine keramische ZrO2-Schicht auf ein metallisches Substrat aufgedampft bzw. thermisch gespritzt worden ist. Im Vordergrund steht die Gefügeanalyse des Verbundwerkstoffes, wobei der Versuch unternommen wird, mit Hilfe der durch TEM-Querschliffe möglichen Zielpräparation auch die Grenzflächenmorphologie aufzuklären.

    Notes: Interaction of macromixing and micromixing in microbiological flow reactors based on a kinetic model of the Michaelis-Menten type has been treated by considering two general types of flow systems: the system of n continuous stirred-tank reactors in series (n-CSTR's in series) and the series combination of a plug-flow reactor and a continuous stirred-tank reactor. Systematic schemes to describe the micromixing conditions of a reactor system are presented. Additional micromixing states are considered besides those proposed by Kramers and those proposed by Zwietering if the systems are employed as empirical models of a flow system. When the number of stirred-reactor units in the system of n-CSTR's in series is small, micromixing has a significant effect on the growth processes. As the number of reactor units increases, the micromixing effect on the growth processes decreases. The effect of micromixing is also important for a system described by a series combination of a plug-flow reactor and a continuous stirred reactor. In addition, the exit concentrations from the system having the same residence time distribution and the same degree of segregation may be different from each other. Information obtained in this study is pertinent in the design of biological flow reactors and sewage treatment systems.

    Notes: Complex formation between poly(U) and adenosine in solutions of salts that stabilize (Na2SO4), destabilize (NaClO4), or have little effect on the water structure (NaCl), as well as the poly(U)·poly(A) interaction in NaClO4, was studied by equilibrium dialysis and uv spectroscopy. At 3°C and neutral pH, Ado·2 poly(U) is formed in 1M NaCl and 0.33M Na2SO4. In NaClO4 solutions under the same conditions, an Ado·poly(U) was found over the whole range of salt concentration investigated (10 mM-1M), which has not been previously observed under any conditions. The Ado-poly(U) was also found in a NaCl/NaClO4 mixture, the transition from the triple- to the double-helical complex occurring within a narrow range of concentration of added NaClO4. In the presence of 1M NaCl this transition is observed on adding as little as 10 mM NaClO4, i.e., at a [ClO4-]/[Cl-] ratio of about 1:100. However, when NaClO4 is added to a 1M solution of the stabilizing salt Na2SO4, no transition occurs even at a [ClO4-]/[SO42-] ratio of 1:4. Investigation of melting curves and uv spectra has shown that in an equimolar mixture of the polynucleotides, only a double-helical poly(U)·poly(A) exists in 1M NaClO4 at low temperatures; this also holds for 1M NaCl. This changes to a triple-helical 2 poly(U)·poly(A) and then dissociates as the temperature increases. At low temperatures and the poly(U)/poly(A) concentration ratio of 2:1, a mixture of 2 poly(U)·poly(A) and poly(U)·poly(A) was observed in 1M NaClO4, in contrast to the case of 1M NaCl. Thus, sodium perchlorate, a strong destabilizer of water structure, promotes formation of double-helical complexes both in the polynucleotide-monomer and the polynucleotide-polynucleotide systems.Beginning with a sufficiently high ionic strength (μ ≃ 0.9), a further increase in the salt molarity results in an increase of the poly(U)·adenosine melting temperature in both stabilizing and neutral salts and a decrease in the destabilizing salt. In Na2SO4 concentrations higher than 1.2M Ado·2 poly(U) precipitates at room temperature. Analysis of the binding isotherms and melting profiles of the complexes between poly(U) and adenosine according to Hill's model shows that the cooperativity of binding, due to adenosine stacking on poly(U), increases in the order NaClO4 〈 NaCl 〈 Na2SO4. The free energy of adenosine stacking on the template is similar to that of hydrogen bonding between adenosine and poly(U) and ranges from -1 to -2 kcal/mol. The values of ΔHt [the effective enthalpy of adenosine binding to poly(U) next to an occupied site, obtained from the relationship between complex melting temperature and free monomer concentration at the midpoint of the transition] are -14.2, -18.3, and -16.8 kcal/mol for 1M solutions of NaClO4, NaCl, and Na2SO4, respectively. The results indicate that the effects of anions of the salts studied are related to water structure alterations rather than to their direct interaction with the complexes between poly(U) and adenosine.

    Notes: The degradation of poly-N-vinylimidazole films on copper substrates was studied by Fourier transform infrared spectroscopy and x-ray photoelectron spectroscopy. Infrared measurements on samples heated at 300°C for 15 minutes revealed that the oxidation of the polymer was accelerated by the copper. X-ray photoelectron spectroscopy showed that a layer of copper oxide was formed on top of the oxidized film. Copper ions were also detected within the polymer layer. © 1996 John Wiley & Sons, Inc.
